Claims (11)
- (ⅰ)형광 단백질의 N 말단 영역과 융합된 융합 단백질의 형태로 발현시키는 벡터와 형광 단백질의 C 말단 영역과 융합된 융합 단백질 형태로 발현시키는 벡터 각각에, 목적 단백질을 코딩하는 핵산 또는 분석 대상 단백질을 코딩하는 핵산을 도입하는 단계;상기 벡터는 DNA 서열 교환(exchange)을 위한 DNA 재조합 서열(DNA recombination sequence) 쌍을 포함하는 부위-특이적 재조합이 일어나는 벡터로, 프로모터 하부에 핵 위치 시그널(nuclear localization signal)을 포함한다.(ⅱ)상기 단계에서 준비한 목적 단백질을 발현하는 벡터 및 분석 대상 단백질을 발현하는 벡터를 세포에 형질전환시키고 배양하는 단계;(ⅲ)목적 단백질과 분석 대상 단백질 간의 상호작용을 형광 단백질의 형광으로 측정하는 단계;를 포함하는 세포내 단백질-단백질 상호작용(in vivo protein-protein interaction)을 분석하는 방법.
- 제1항에 있어서, 형광단백질이 YFP(yellow fluorescent protein), GFP(green fluorescent protein), enhanced GFP, BFP(blue fluorescent protein) 또는 CFP(cyan fluorescent protein)인 방법.
- 제1항에 있어서, 상기 벡터의 DNA 재조합 서열이 attR인 것인 방법.
- 제1항에 있어서, 벡터의 핵 위치 시그널이 PPKKRKV를 코딩하는 핵산 서열을 포함하는 것인 방법,상기에서 P는 프롤린, K는 라이신, R은 아르기닌 및 V는 발린을 나타낸다.
- 제1항에 있어서, 벡터의 융합단백질을 코딩하는 서열의 하부에 태그(tag)를 코딩하는 서열을 추가로 포함하는 벡터를 포함하는 것인 방법.
- 제5항에 있어서, 태그가 플래그 태그(Flag tag) 또는 HA 태그인 벡터를 포함하는 것인 방법.
- 삭제
- 제1항에 있어서, 상기 벡터가 CMV 프로모터를 포함하는 벡터를 포함하는 것인 방법.
- 제1항 내지 제6항 및 제8항 중 어느 한 항에 있어서, 상기 벡터 중 형광 단백질의 N 말단 영역과 융합된 융합 단백질 형태로 발현시키는 벡터가 도 1A에 개시된 pFPIA-YN 벡터이고 형광 단백질의 C 말단 영역과 융합된 융합 단백질 형태로 발현시키는 벡터가 도 1B에 개시된 pFPIA-YC 벡터인 방법.
- 제1항에 있어서, (ⅲ) 단계에서 형광을 형광 현미경 또는 콘포컬 현미경으로 측정하는 것인 방법.
- 세포내 단백질-단백질 상호작용 분석을 위한 도 1A에 개시된 pFPIA-YN 및 도 1B에 개시된 pFPIA-YC 벡터.
