Claims (52)
- a) 비-사람 다세포 동물에게, 발현을 촉진하는 능력을 분석하고자 하는 내인성 유전자의 프로모터에 작동적으로 연결된 형광단을 암호화하는 핵산을 전달하거나 이러한 핵산을 함유하는 세포를 전달하는 단계; 및b) 전신 외부 형광 광학 영상화(whole-body external fluorescent optical imaging)에 의해 상기 동물 내의 각종 위치에서 상기 형광단에 의해 발생된 형광의 존재, 부재 또는 이의 강도를 관찰함으로써, 발현을 촉진하는 상기 프로모터의 능력을 모니터하는 단계를 포함하고, 이때 상기 형광단이 자체 형광성이어서 이것이 형광 발생하는데 있어 기질 또는 조인자를 필요로 하지 않는 단백질인, 비-사람 다세포 동물에서 상기 프로모터 조절하에 있는 내인성 유전자의 발현을 촉진하는 프로모터의 능력을 모니터하는 방법.

- a) 내인성 유전자의 프로모터의 지시하에 형광단을 발현하는 비-사람 다세포 동물에게 시험 물질을 투여한 다음, 전신 외부 형광 광학 영상화에 의해 상기 다세포 동물 내의 각종 위치에서 상기 형광단에 의해 발생된 형광의 존재, 부재 또는 이의 강도를 관찰함으로써 상기 프로모터의 발현도를 측정하는 단계;b) 상기 유전자의 프로모터의 지시하에 상기 형광단을 발현하는 대조군 비-사람 다세포 동물에서, 전신 외부 형광 광학 영상화에 의해 각종 위치에서 상기 형광단에 의해 발생된 형광의 존재, 부재 또는 이의 강도를 관찰함으로써 상기 내인성 프로모터의 발현도를 측정하는 단계; 및c) 상기 단계 a)와 b)에서 측정된 프로모터의 발현도를 비교하는 단계를 포함하고, 이때 상기 시험 물질이 상기 유전자 발현을 조정하는 경우, 단계 a)에서 측정된 발현도는 단계 b)에서 측정된 발현도와 상이하고, 상기 형광단이 자체 형광성이어서 이것이 형광 발생하는 데 있어 기질 또는 조인자를 필요로 하지 않는 단백질인, 비-사람 다세포 동물에서 유전자 발현 조절제를 스크리닝하는 방법.

- a) 내인성 유전자의 프로모터의 지시하에 형광단을 발현하는 비-사람 다세포 동물에게 돌연변이-유발제를 투여하거나 돌연변이 처리한 다음, 전신 외부 형광 광학 영상화에 의해 상기 다세포 동물 내의 각종 위치에서 상기 형광단에 의해 발생된 형광의 존재, 부재 또는 이의 강도를 관찰함으로써 상기 프로모터의 발현도를 측정하는 단계;b) 상기 유전자의 프로모터의 지시하에 상기 형광단을 발현하는 처리되지 않은 대조군 다세포 동물에서, 전신 외부 형광 광학 영상화에 의해 각종 위치에서 상기 형광단에 의해 발생된 형광의 존재, 부재 또는 이의 강도를 관찰함으로써 상기 내인성 프로모터의 발현도를 측정하는 단계; 및c) 상기 단계 a)와 b)에서 측정된 프로모터의 발현도를 비교하는 단계를 포함하고, 이때 상기 다세포 유기체가 이상 수준으로 상기 유전자를 발현하는 경우, 단계 a)에서 측정된 발현도는 단계 b)에서 측정된 발현도와 상이하고, 상기 형광단이 자체 형광성이어서 이것이 형광 발생하는데 있어 기질 또는 조인자를 필요로 하지 않는 단백질인, 이상 수준으로 유전자를 발현하는 비-사람 다세포 동물을 스크리닝하는 방법.
- 제39항에 있어서, 돌연변이-유발제의 투여 또는 돌연변이 처리가 당해 다세포 동물의 배선(germ line) 세포에서의 돌연변이를 유발시켜, 이러한 돌연변이가 상기 다세포 동물의 자손에게 안정적으로 전이될 수 있도록 하는 방법.
- 제1항, 제6항 또는 제7항 중 어느 한 항에 있어서, 형광단이 그린 형광 단백질(GFP), 블루 형광 단백질(BFP) 및 레드 형광 단백질(RFP)로 이루어진 그룹 중에서 선택되는 방법.
- 제1항, 제6항 또는 제7항 중 어느 한 항에 있어서, 동물이 마우스, 랫트, 토끼, 또는 비-사람 영장류인 방법.
- 제1항, 제6항 또는 제7항 중 어느 한 항에 있어서, 내인성 유전자가 조직 또는 기관 특이적 방식으로 정상적으로 발현되는 방법.
- 제43항에 있어서, 조직이 연결 조직, 상피 조직, 근육 조직 및 신경 조직으로 이루어진 그룹 중에서 선택되는 방법.
- 제37항에 있어서, 형광단이 그린 형광 단백질(GFP), 블루 형광 단백질(BFP) 및 레드 형광 단백질(RFP)로 이루어진 그룹 중에서 선택되는 방법.
