Claims (5)
- a) 목표 물질(T, target)에 결합하는 항체와 항원, 효소와 기질, 펩타이드(peptide), 핵산(nucleotide), 앱타머(aptamer), 염료(dye) 및금속이온(metal ion)과 같은 리간드(ligand) 및 어셉터(acceptor)로 작용 가능한 물질로 구성된 군으로부터 선택되는 분자특이적 결합력이 있는 물질(B, binding)에 HRP(horseradish peroxidase), 알칼라인 포스파타제(Alkaline phosphatase), 베타-갈락토시다제(beta-galactosidase), 리소자임(lysozyme), 우혈청 알부민(bovine serum albumin)과 같은 폴리펩티드 및 펩티드로 구성된 군으로부터 선택되는 분배향상 물질(P, partition)을 접합시켜 목표 물질과 다른 상(phase)에 위치하도록 B의 분배계수를 변화시킨 B-P 접합체를 제조하는 단계;b) 단계 a)의 상기 접합체를 T가 함유된 수성이상계에 투입하는 단계; 및c) 단계 b)에서 투입 후 생성된 T-B-P 결합체를 분리해내는 단계를 포함하는 친화도에 의한 생물물질 분리방법.

- 제 1항에 있어서, 단계 3의 수성이상계는 PEG(polyethylene glycol)와 인산칼륨 완충용액, PEG와 덱스트란, 폴리비닐알코올(polyvinylalcohol), 덱스트란(dextran), 폴리에틸렌 폴리프로필렌 블록 코폴리머(polyethylene polypropylene block copolymer), 올리고사카라이드(oligosaccharide), 인산칼륨(potassium phosphate), 인산나트륨(sodium phosphate), 암모늄 설페이트(ammonium sulfate) 및 마그네슘 설페이트(magnesium sulfate)로 구성된 군으로부터 선택되는 것을 특징으로 하는 분리방법.
- 제 1항에 있어서, 목표물질은 항-인간 IgG이고 분자특이적 결합력이 있는 물질은 인간 IgG것을 특징으로 하는 분리방법.
