Claims (20)
- 모낭을 포함하는 두피조직을 잘게 절개한 다음 절개된 조직을 단백질 가수분해 효소, 교원분해(collagenolytic) 효소 및 DNAse 효소로 이루어진 효소 복합체와 디스페이즈가 함유된 배지에서 1차 화학적 분해를 행하는 단계; 콜라게나아제가 함유된 배지에서 2차 화학적 분해를 행하는 단계;화학적 분해된 조직을 회수하여 두피세포를 혈청 1-30%가 포함된 배지에서 배양한 다음, 세포부착후 무혈청 배지에 배양하는 단계;배양증식된 두피세포를 회수한 다음 이로부터 CD34+ 세포를 분리하여 모낭 줄기세포를 분리하는 단계; 및상기 분리된 모낭 줄기세포를 두피조직에 피하주사로 투여하여 모낭 세포로 분화시키는 단계를 포함하는, 인체를 제외한 포유류에서 모낭 줄기세포의 분리 및 모낭 세포로의 분화 방법.
- 제 1항에 있어서, 1차 화학적 분해에 사용되는 상기 효소 복합체와 디스페이즈가 함유된 배지는 우태아혈청 1-30%, 노르모신 10-200ug/ml, 페니실린-스트렙토마이신 1-10%가 포함된 DMEM배지에 디스페이즈 0.1-2mg/ml 및 단백질 가수분해 효소, 교원분해(collagenolytic) 효소 및 DNAse 효소로 이루어진 효소 복합체 1-10%가 첨가된 배지인 것을 특징으로 하는 방법.
- 삭제
- 제 1항에 있어서, 2차 화학적 분해에 사용되는 콜라게나아제가 함유된 배지는 우태아혈청 1-30%, 노르모신 10-200ug/ml, 페니실린-스트렙토마이신 1-10%가 포함된 DMEM배지에 콜라게나아제 타입ⅠA를 0.1-10mg/ml 첨가한 배지인 것을 특징으로 하는 방법.
- 제 1항에 있어서, 화학적 분해는 50-200m/min, 30-40℃에서 0.5-24시간 동안 중력대류배양기에서 행하는 것을 특징으로 하는 방법.
- 제 1항에 있어서, 회수된 두피세포의 배양은 우태아혈청 1-30%가 포함된 M199/F12배지에서 배양한 다음, 세포부착후 우태아혈청이 없는 M199/F12배지에서 배양하는 것을 특징으로 하는 방법.
- 제 6항에 있어서, 처음에 사용되는 M199/F12배지는 M199와 F12를 1:1로 섞은후 인슐린 0.1-1.0ug/ml, 트랜스페린 0.1-1.0ug/ml, 페니실린-스트렙토마이신 1-10%, rEGF(receptor for human epidermal growth factor) 1-100ng/ml, bFGF(basic fibroblast growth factor) 1-100ng/ml, 노르모신 10-200ug/ml, 우태아혈청 1-30%, N-아세틸-L-씨스테인 0.1-10mM 을 첨가한 배지인 것을 특징으로 하는 방법.
- 제 6항에 있어서, 나아가 우태아 혈청이 없는 M199/F12배지로 배양 일주일후 노르모신이 없고 아스코르빈산 0.1-10mM이 포함된 무혈청 케라티노사이트용 배지에서 배양증식하는 것을 특징으로 하는 방법.

- 제 1항에 있어서, 분리된 모낭 줄기세포의 면역표현형 항원은 CD34 양성을 나타내며, 또한 CD44 양성, CD45 양성, CD133 양성, CD29 양성 중에서 어느 하나 또는 복수의 면역학적 특성을 나타내는 것을 특징으로 하는 방법
- 제 1항에 있어서, 상기 두피조직은 포유류로부터 얻어진 것을 특징으로 하는 방법.
- 모낭을 포함하는 두피조직을 잘게 절개한 다음 절개된 조직을 단백질 가수분해 효소, 교원분해(collagenolytic) 효소 및 DNAse 효소로 이루어진 효소 복합체와 디스페이즈가 함유된 배지에서 1차 화학적 분해를 행하는 단계; 콜라게나아제가 함유된 배지에서 2차 화학적 분해를 행하는 단계;화학적 분해된 조직을 회수하여 두피세포를 혈청 1-30%가 포함된 배지에서 배양한 다음, 세포부착후 무혈청 배지에 배양하는 단계; 및배양증식된 두피세포를 회수한 다음 이로부터 CD34+ 세포를 분리하여 모낭 줄기세포를 분리하는 단계에 의해 분리된 모낭 줄기세포를 유효성분으로 하는 발모촉진용 조성물.
- 제 12항에 있어서, 1차 화학적 분해에 사용되는 상기 효소 복합체와 디스페이즈가 함유된 배지는 우태아혈청 1-30%, 노르모신 10-200ug/ml, 페니실린-스트렙토마이신 1-10%가 포함된 DMEM배지에 디스페이즈 0.1-2mg/ml, 단백질 가수분해 효소, 교원분해(collagenolytic) 효소 및 DNAse 효소로 이루어진 효소 복합체 1-10% 첨가한 배지인 것을 특징으로 하는 발모촉진용 조성물.

- 제 12항에 있어서, 상기 모낭 줄기세포는 1×103-1×1012 세포의 양으로 투여되는 것을 특징으로 하는 발모촉진용 조성물.
- 제 12항에 있어서, 회수된 두피세포의 배양은 우태아혈청이 포함된 M199/F12배지에서 배양한 다음, 세포부착후 우태아혈청이 없는 M199/F12배지에서 배양하는 것을 특징으로 하는 발모촉진용 조성물.
- 제 17항에 있어서, 나아가 우태아 혈청이 없는 M199/F12배지로 배양 일주일후 노르모신이 없고 아스코르빈산 0.1-10mM이 포함된 무혈청 케라티노사이트용 배지에서 배양하는 것을 특징으로 하는 발모촉진용 조성물.
- 제 12항에 있어서, 유효성분이 되는 모낭 줄기세포의 면역표현형 항원은 CD34 양성을 나타내며, 또한 CD44 양성, CD45 양성, CD133 양성, CD29 양성 중에서 어느 하나 또는 복수의 면역학적 특성을 나타내는 것을 특징으로 하는 발모촉진용 조성물.
- 제 19항에 있어서, 복수의 면역학적 특성을 나타내는 경우 CD44 양성, CD45 양성, CD133 양성, CD29 양성에 대해서 양성을 공통적으로 나타내는 것을 특징으로 하는 발모촉진용 조성물.
