Description
Claims (12)
- 다음의 단계를 포함하는 중간엽 줄기세포로부터 연골세포를 분화시키는 방법:(a) 중간엽 줄기세포를 단층 배양한 다음, 상기 단층배양된 중간엽 줄기세포를 원심분리하여 세포펠렛을 수득하는 단계;(b) 상기 중간엽 줄기세포의 세포펠렛을 포어(pore)를 가진 삼차원 생분해성 합성고분자 또는 포어(pore)를 가진 삼차원 천연고분자를 함유하는 배지에서 배양하여 중간엽 줄기세포가 고정된 삼차원 지지체를 제조하는 단계;(c) 상기 중간엽 줄기세포가 고정된 삼차원 지지체를 인간을 제외한 동물에 주입하는 단계; 및(d) 상기 동물의 중간엽 줄기세포가 고정된 삼차원 지지체 주입부위를 초음파로 처리하여 연골세포로의 분화를 촉진시키는 단계.
- 삭제
- 제1항에 있어서, 고분자는 PGA(polyglycolic acid), PLA(polylatic acid), PCL(polycaprolactone), polylatic acid/polyglycolic acid, 히알유론산(hyaluronic acid), 콜라겐, 알지네이트 및 이들의 공중합체로 구성된 군에서 선택되는 것을 특징으로 하는 방법.
- 제1항에 있어서, 상기 초음파는 파장 10kHz~100MHz, 출력강도 10~5000mW/cm2인 것을 특징으로 하는 방법.
- 제1항에 있어서, 상기 중간엽 줄기세포는 배아, 성체조직, 또는 골수(bone-marrow) 유래인 것을 특징으로 하는 방법.
- 제1항에 있어서, 상기 초음파 처리는 6~24시간 간격으로 1~30분간, 3~50일 동안 연속하여 실시하는 것을 특징으로 하는 방법.
- (a) 중간엽 줄기세포를 단층 배양한 다음, 상기 단층배양된 중간엽 줄기세포를 원심분리하여 세포펠렛을 수득하는 단계; 및 (b) 상기 중간엽 줄기세포의 세포펠렛을 포어(pore)를 가진 삼차원 생분해성 합성고분자 또는 포어(pore)를 가진 삼차원 천연고분자를 함유하는 배지에서 배양하여 중간엽 줄기세포가 고정된 삼차원 지지체를 제조하는 단계를 거쳐 제조된, 중간엽 줄기세포가 고정된 삼차원 지지체를 함유하는 연골질환 치료용 조성물.
- 삭제
- 제7항에 있어서, 상기 고분자는 PGA(polyglycolic acid), PLA(polylatic acid), PCL(polycaprolactone), polylatic acid/polyglycolic acid, 히알유론산(hyaluronic acid), 콜라겐, 알지네이트 및 이들의 공중합체로 구성된 군에서 선택되는 것을 특징으로 하는 조성물.
- 제7항에 있어서, 상기 연골질환은 퇴행성 관절염, 류마티스성 관절염, 골절, 근육조직의 손상, 족저근막염, 상완골외과염, 석회화근염, 골절의 불유합 또는 외상에 의한 관절손상인 것을 특징으로 하는 조성물.
- 삭제
- 제1항의 방법으로 분화된 연골세포를 사용하는 것을 특징으로 하는 인공관절의 제조방법.
