Claims (8)
- (ⅰ) 반응기(X)-스페이서 부위(R)-타겟프로브 인식부위(V)로 구성된 단일가닥핵산인 캡쳐프로브들을 고체 기판 위에 고착시켜 어레이를 제작하는 제 1단계; (ⅱ) 분석시약의 처리에 의해 특정물질과 타겟프로브를 반응시켜 특정물질-타겟프로브 복합체를 제조하는 제 2단계; (ⅲ) 상기 제 2단계에서 제조된 특정물질-타겟프로브 복합체를 세척 및 분리하는 제 3단계; (ⅳ) 분리된 복합체에 혼성화용액을 첨가하고 95℃에서 5분간 처리하여 타겟프로브를 해리시키는 제 4단계; (ⅴ) 캡쳐프로브와 표지된 타겟프로브를 반응시키는 제 5단계; 및 (ⅵ) 상기 타겟프로브에 표지된 물질을 탐색하는 제 6단계로 구성된 단일가닥핵산 어레이를 이용한 특정물질의 동정 및 정량방법.
- 제 1항에 있어서, 상기 제 2단계의 분석시약은(ⅰ) 20 내지 100mM 트리스·염산(pH 7.4),(ⅱ) 각각 1 내지 200mM의 염화칼륨, 염화나트륨, 염화마그네슘, 또는 이들의 혼합용액,(ⅲ) 0.05 내지 0.2%의 소듐 아지드,(ⅳ) 0.1 내지 0.3%의 우혈청알부민 또는 비특이적인 결합을 저해하는 핵산을 포함하는 셀렉스 선택버퍼인 것을 특징으로 하는 방법.
- 제 2항에 있어서, 상기 분석시약은 50mM 트리스·염산(pH 7.4), 5mM 염화칼륨, 100mM 염화나트륨 또는 1mM 염화마그네슘, 0.1% 소듐아지드, 0.2% 우혈청알부민 또는 단일가닥핵산인 것을 특징으로 하는 방법.
- 제 1항에 있어서, 상기 제 2단계의 특정물질은 세균, 진균류, 바이러스, 세포주, 조직, 이로부터 분리된 단백질, 탄수화물, 지질, 다당체, 당단백, 호르몬, 수용체, 항원, 항체 및 효소로 이루어진 군으로부터 선택된 1종 이상인 것을 특징으로 하는 방법.
- 제 1항에 있어서, 상기 제 3단계의 세척버퍼는 0.01 내지 1 x 세렉스버퍼 또는 1 내지 500 mM EDTA 용액인 것을 특징으로 하는 방법.
- 제 1항에 있어서, 상기 제 4단계의 핵산 혼성화용액은(ⅰ) 0.5 내지 1.5 M 염화나트륨,(ⅱ) 0.1 내지 1.0 M의 소듐 시트레이트,(ⅲ) 비특이적인 결합을 저해하는 물질인 0.1 내지 0.3%의 우혈청알부민, 0.1 내지 1.0% SDS, 0.1 내지 10 배의 덴하르트 용액(Denhart solution) 또는 0.1 내지 1% 비지방 건조 우유(nonfat dried milk),(ⅳ) 비특이적인 결합을 저해하는 핵산,(v) 30 내지 60 % (v/v) 포름아미드를 포함하는 것을 특징으로 하는 방법.
- 제 6항에 있어서, 상기 핵산 혼성화 용액은 1 M 염화나트륨, 0.3 M 소듐 시트레이트, 0.5% SDS 또는 100㎍/㎖ 살몬스펌 DNA, 0.2% 우혈청알부민 또는 단일가닥핵산인 것을 특징으로 하는 방법.
