Claims (7)
- 효소 촉매와 화학 촉매를 병용하는 동적속도론적 광학 분할방법에 의해 라세믹 2-아릴프로피온산 씨오에스테르로부터 (S)-2-아릴프로피온산 유도체를 제조하는 방법에 있어서,라세믹 2-아릴프로피온산 씨오에스테르를 효소에 의해 입체선택적으로 가수분해반응하여 (S)-2-아릴프로피온산을 제조하는 생물학적 광학 분할 반응기와,(R)-2-아릴프로피온산 씨오에스테르를 고체화합물에 의해 라세미화하는 화학적 라세미화 반응기가 별도로 설치되어 있고,상기 두 반응기는 용액이송관과 펌프로 연결되어 반응용액이 40 내지 140 mL/min의 유속으로 상기 두 반응기를 순환하도록 하여 동적속도론적 광학 분할(dynamic kinetic resolution)을 수행하는 것을 특징으로 하는 (S)-2-아릴프로피온산 유도체의 제조방법.
- 제 1 항에 있어서, 상기 생물학적 광학 분할 반응기는 (S)-2-아릴프로피온산 씨오에스테르를 입체선택적으로 가수분해하는 효소 촉매가 고정되어 있는 고정층 반응기로서, 상기 반응기의 상단 및 하단에는 효소 촉매가 반응기 밖으로 배출되지 않고 반응용액만 통과하도록 하는 필터가 장착되어 있는 것을 특징으로 하는 제조방법.
- 제 1 항 또는 제 2 항에 있어서, 상기 효소는 리파아제, 에스터라아제 및 프로테아제 중에서 선택된 것을 특징으로 하는 제조방법.
- 제 1 항에 있어서, 상기 생물학적 광학 분할 반응기 내부는 30 내지 60 ℃ 온도, pH 5 내지 7 범위를 유지하는 것을 특징으로 하는 제조방법.
- 제 1 항에 있어서, 상기 화학적 라세미화 반응기는 (R)-2-아릴프로피온산 씨오에스테르를 라세미화하는 고체 염기촉매, 고체 산촉매 및 금속 촉매 중에서 선택된 화학 촉매가 고정되어 있는 고정층 반응기로서, 상기 반응기의 상단 및 하단에는 화학 촉매가 반응기 밖으로 배출되지 않고 반응용액만 통과하도록 하는 필터가 장착되어 있는 것을 특징으로 하는 제조방법.
- 제 5 항에 있어서, 상기 금속 촉매는 루테늄 또는 팔라듐 금속 착화합물인 것을 특징으로 하는 제조방법.
- 제 1 항에 있어서, 상기 화학적 라세미화 반응기 내부는 70 내지 90 ℃ 온도를 유지하는 것을 특징으로 하는 제조방법.
