Claims (7)
- 오로트산과 콜린 및/또는 포스포릴콜린으로부터 시티딘 디인산 콜린을 생성할 수 있는, 시티딘-5'-트리인산 신세타제(이하에서는 pyrG라고 한다), 콜린포스페이트 시티디릴 트랜스퍼라제 (이하에서는, CCT라고 한다) 및 콜린 키나제 (이하에서는 CKI라고 한다)의 효소활성을 갖는 미생물 (이하에서는 미생물 A1라고 한다) 또는 pyrG 및 CCT의 효소활성을 갖는 미생물(이하에서는 미생물 A2라고 한다)에서 선택한 1종의 미생물과 오로트산으로부터 우리딘-5'-트리인산을 생성하는 활성을 갖는 미생물(이하에서는 미생물 B라고 한다)의 배양액 또는 이의 배양액의 농축물 또는 건조물, 배양액을 원심분리하여 수득되는 균체, 균체의 건조물, 계면활성제 및/또는 유기 용제처리물, 용균효소처리물, 고정화균체 또는 균체로부터의 추출효소표퓸등으로 이루어진 군에서 선택한 1종이상의 처리물을 효소원으로서 사용하고, 오로트산과 콜린 및/또는 포스포릴콜린을 기질로서 사용하여 반응을 수행하고, 반응액중에 시티딘 디인산 콜린을 생성 축적시키고, 이 반응액으로부터 시티딘 디인산콜린을 채취하는 것을 특징으로 하는 시티딘 디인산 콜린의 제조방법.
- 제1항에 있어서, 미생물 A1이 pyrG, CCT 및 CKI를 코딩하는 유전자를 함유하는 DNA단편과 벡터와의 재조합체 DNA를 보유하는 것임을 특징으로하는 방법.
- 제1항에 있어서, 미생물 A2가 pyrG 및 CCT를 코딩하는 유전자를 함유하는 DNA단편과 벡터와의 재조합체 DNA를 보유하는 것임을 특징으로 하는 방법.
- 제2항 또는 제3항에 있어서, CCT 및 CKI를 코딩하는 유전자가 사카로마이세스·세레비지아 (Saccharomyces cerevisiae)유래이며, pyrG를 코딩하는 유전자가 에스케리키아·콜리 (Escherichia coli) 유래인 것임을 특징으로 하는 방법.
- 제1항에 있어서, 미생물 A1 또는 미생물 A2가 에스케리키아속에 속하는 미생물이며, 미생물 B가 코리네박테륨(Coynebacterium)속에 속하는 미생물인 것을 특징으로하는 방법.
- 제5항에 있어서, 미생물 A1이 에스케리키아·콜리 MM294/pCKG 55 (FERM BP-3717) 인 것을 특징으로 하는 방법.
- 제5항에 있어서, 미생물 B가 코리네박테륨·암모니아게네스 ATCC21170 인 것을 특징으로하는 방법.
