Claims (6)
- (a) 타겟 분절의 3'-하위분절의 서열에 충분히 상보적인 서열과 여기에 조작적으로 결합되는 프로모터 서열을 포함하는 단일-스트랜드 DNA인 제1프라이머 및 (b) 타겟 분절의 5'-하위 분절의 일부 서열을 포함하는 단일-스트랜드 DNA 인 제2프라이머; (c) 상기 프로모터를 인식하는 RNA 중합효소 활성을 지닌 효소, (d) DNA 의존성 DNA 중합효소 활성을 지닌 효소, (e) RNA 의존성 DNA 중합효소 활성을 지닌 효소, (f) 리보뉴클레오사이드 및 데옥시리보뉴클레오사이드 트리포스페이트 및 (g) RNase H 활성을 지닌 효소를 포함하는 반응 혼합물에 타겟 RNA를 접촉시키고 상기 반응 혼합물을 기능적 이중 스트랜드 프로모터와 상기 타겟 RNA 분절의 서열을 포함하는 템플레이트의 형성 및 RNA 중합효소 활성을 지닌 효소에 의하여 상기 프로머터를 포함하는 테플레이트로부터 전사에 의한 RNA의 형성(이렇게 함으로써 전사에 의해 만들어지는 RNA는 RNA 의존성 DNA중합효소 활성, DNA 의존성 DNA 중합효소 활성을 지닌 효소, RNA 중합효소 활성을 지닌 효소 및 두 개의 프라이머를 사용하여 추가 증폭을 위한 타겟으로 사용된다)을 수반하는 반응이 일어나도록 하는 적절한 조건 하에서 유지시키는 것을 포함하는, 핵산 듀플렉스의 임의의 실질적 열 변성을 수반하지 않는 연속 반응인, 중복되지 않은 5'-하위분절 및 3'-하위단편을 포함한 타겟 RNA 분절을 증폭시키는 방법.
- 제1항에 있어서, RNase H 활성을 지닌 효소가 이. 콜라이 RNase H임을 특징으로 하는 방법.
- 제1항 또는 제2항에 있어서, DNA-의존성 DNA 중합효소 및 역전사효소 활성 모두가 AMV 및 MMLV로 구성된 군 중에서 선택되는 레트로바이러스의 역전사효소에 의하여 제공되고 RNA 중합효소 활성이 T7, T3 및 SP6로 구성된 군 중 중에서 선택되는 박테리오파지의 RNA 중합효소에 의하여 제공되며 용액 내에는 하나의 RNA 중합효소가 존재함을 특징으로 하는 방법.
- 제1항 또는 제2항에 있어서,상기 반응 혼합물이 33℃ 내지 46℃ 온도에서 유지됨을 특징으로 하는 방법.
- 제1항 또는 제2항에 있어서, 증폭 후에 타겟 RNA 및/또는 상기 타겟 RNA에 상보적인 서열을 지닌 RNA가 핵산 프로브 하이브리드화 분석에 의하여 검출됨을 특징으로 하는 방법.
- 제1항 또는 제2항에 있어서, 제2 프라이머가 반응에서 형성된 RNA/DNA 듀플렉스의 RNA 스트랜드의 선택적인 분해 결과인 방법.
