本出願は平成30年6月18日に出願された米国仮特許出願第62/686,508号の利益を主張する、令和1年6月17日に出願された米国特許出願第16/443,658号の利益を主張し、平成29年7月3日に出願された米国特許出願第15/641,114号の一部継続出願であり、その3つすべてが参照により本明細書に組み込まれる。
本発明は一般に、微生物発酵及び工業的バイオテクノロジーの分野に関し、より詳細には、従属栄養生物の培養のための栄養豊富な増殖培地の製造に関する。

倍以上に増加させたことを示す。追加のグルコースは特に0.5%添加の後期段階で、このレベルを超えるそれらの増殖を刺激したが、1%グルコースは培養期間が長くなるにつれて効果が低くなる可能性がある。これは、第2の培地が完全培地又は有意な培地成分のいずれかとして従属栄養細胞を培養するために使用され得ることを実証する。この方法はまた、従属栄養細胞をガス上で間接的に効果的に培養することを可能にし、それによって、それらが代謝的に適していない供給原料及び培養条件を利用する。また、本発明は、元の抽出されたバイオマスを超えて、ガス発酵の生成物から付加的な価値を抽出することを可能にする。
Claims (22)
- バイオリアクターへ有機化合物を含まない培地を提供することと、
化学合成独立栄養細胞を含む接種材料を前記バイオリアクター中の前記培地に接種することと、
前記培地中のバイオマスの細胞密度が閾値を満たすまで、前記バイオリアクターへのガス投入を提供することによって、前記バイオリアクター中でバイオマスを増殖させるために、化学合成独立栄養的に、接種材料を培養し、それによって、培養中に前記培地が濃縮され、濃縮培地となることと、
を含み、
前記接種材料が従属栄養細胞も含み、
前記ガス投入がCH4及びO2を含み、
前記化学合成独立栄養細胞がMethylococcus capsulatusの細胞、Cupriavidus necatorの細胞、Rhodobacter capsulatusの細胞、及びRhodococcus opacusの細胞を含む、前記濃縮培地を得る方法。 - 前記化学合成独立栄養細胞が、成長因子、ホルモン、抗生物質、アミノ酸、ペプチド、タンパク質、ビタミン、着色剤、カロチノイド、脂肪酸、又は油を産生する、請求項1に記載の方法。
- 前記接種材料を培養することが、有益な分子を前記濃縮培地に添加することを含む、請求項1又は2に記載の方法。

- 前記接種材料が一酸化炭素資化性(カルボキシドトロフ)の細胞を含む、請求項1~4
又は5に記載の方法。 - 前記培地に接種する前に、前記接種材料を調製することをさらに含む、請求項1~5又は6に記載の方法。
- 前記培地を調製することをさらに含む、請求項1~6又は7に記載の方法。
- 前記培地に前記接種材料を接種する前に、前記培地及び前記バイオリアクターを滅菌することをさらに含む、請求項1~7又は8に記載の方法。
- 細胞の内容物を前記濃縮培地中に放出するように、前記濃縮培地中の細胞を破壊することをさらに含む、請求項1~8又は9に記載の方法。
- 前記細胞を破壊することが、前記細胞を溶解することを含む、請求項10に記載の方法。
- 前記濃縮培地が、成長因子、ホルモン、抗生物質、アミノ酸、ペプチド、タンパク質、ビタミン、着色剤、カロテノイド、脂肪酸、又は油の1つ以上を含む、請求項1~10又は11に記載の方法。
- 不溶性バイオマスを前記濃縮培地から分離することをさらに含む、請求項1~11又は12に記載の方法。
- 前記濃縮培地が、分離後、リットル当たり少なくとも1グラムのD-グルコースを含む、請求項13に記載の方法。
- 分離後、無機塩を前記濃縮培地に添加すること、前記濃縮培地のpHを調節すること、前記濃縮培地を濾過すること、前記濃縮培地に酵素処理を施すこと、前記濃縮培地上でクロマトグラフィーによる分離を行うこと、又は前記濃縮培地から選択的に沈殿させることのうちの1つ以上をさらに含む、請求項13又は14に記載の方法。
- 前記バイオマスを前記濃縮培地から分離することが、遠心分離、濾過、又は重力に基づく分離を含む、請求項13、14又は15に記載の方法。
- 前記濃縮培地からアンモニウムイオンを除去することをさらに含む、請求項13~15又は16に記載の方法。
- 前記バイオマスから分離された前記濃縮培地中で従属栄養細胞の細胞を培養し、それによって前記濃縮培地を枯渇させることをさらに含む、請求項13~16又は17に記載の方法。
- 前記従属栄養細胞が酵母、真菌、藻類、古細菌、細菌、又は哺乳動物を含む、請求項18に記載の方法。
