Description
図1に、本実施形態の脱細胞化組織の製造方法の一例を示す。
脱細胞化前処理装置は、液化ガスを含む液体を用いて、生体組織の細胞を破壊することが可能であれば、特に限定されない。
核酸成分分解装置は、脱細胞化前処理装置により破壊された細胞に含まれる核酸成分を、核酸分解酵素を用いて、分解させることが可能であれば、特に限定されず、例えば、公知の振とう機を用いることができる。
洗浄装置は、核酸成分分解装置により核酸が分解した生体組織を洗浄することが可能であれば、特に限定されず、例えば、公知の振とう機を用いることができる。
(工程(S1))
図4に示す脱細胞化前処理装置を用いて、生体組織としての、ブタ由来の大動脈57の細胞を破壊した。
工程(S1)により得られた細胞が破壊された生体組織をDNaseI(ロシュ・ダイアグノスティックス社製)0.2mg/mL、MgCl2(和光純薬工業社製)0.05Mを含む生理食塩水に入れ、4℃の雰囲気で7日間振とうし、DNAを分解させた。
工程(S2)により得られたDNAが分解した生体組織を、エタノール80体積%を含む生理食塩水に入れ、4℃の雰囲気で3日間振とうさせた後、生理食塩水に入れ、4℃の雰囲気で1日間振とうさせ、脱細胞化組織を得た。
工程(S2)で、4℃の雰囲気で、それぞれ5日間、3日間振とうさせた以外は、実施例1と同様にして、脱細胞化組織を得た。
ジメチルエーテル60mLの代わりに、ジメチルエーテル57mLおよび水3mLを用いた以外は、実施例1と同様にして、脱細胞化組織を得た。

Claims (9)
- 液化ジメチルエーテルを含む液体を用いて、生体組織の細胞を破壊する工程と、
核酸分解酵素を用いて、前記破壊された細胞に含まれる核酸成分を分解させる工程を含む脱細胞化組織の製造方法。 - 前記液体は、溶媒をさらに含む請求項1に記載の脱細胞化組織の製造方法。
- 前記溶媒は、水および/またはエタノールである請求項2に記載の脱細胞化組織の製造方法。
- 前記液化ジメチルエーテルの温度が1~40℃の範囲内である請求項1から3のいずれか一項に記載の脱細胞化組織の製造方法。
- 前記液化ジメチルエーテルの圧力が0.2~5MPaの範囲内である請求項1から4のいずれか一項に記載の脱細胞化組織の製造方法。
- 前記核酸成分が分解した生体組織を洗浄する工程をさらに含む請求項1から5のいずれか一項に記載の脱細胞化組織の製造方法。
- 前記核酸成分が分解した生体組織を、水、生理食塩水、エタノール水溶液、または、前記液化ジメチルエーテルを含む液体で洗浄する請求項6に記載の脱細胞化組織の製造方法。
- 前記核酸成分が分解した生体組織を、エタノール水溶液、または、前記液化ジメチルエーテルを含む液体で洗浄した後に、水、または、生理食塩水で洗浄する請求項7に記載の脱細胞化組織の製造方法。
- 液化ジメチルエーテルを含む液体を用いて、生体組織の細胞を破壊する手段と、
核酸分解酵素を用いて、前記破壊された細胞に含まれる核酸成分を分解させる手段を含む脱細胞化組織の製造装置。
