Abstract
Description
酸溶液0.01〜1重量%を含む癒着防止用高分子組成物を提供する。
温度を3〜7℃に下げた後、化1で表されるポリエチレングリコール−ポリプロピレングリコール−ポリエチレングリコール(PEG−PPG−PEG)ブロック共重合体24〜50重量%を添加し、1〜10時間の間攪拌する段階と、を含む癒着防止用高分子組成物の製造方法を提供する。
酸溶液0.01〜1重量%を含む癒着防止用高分子組成物に関する。
(S2)温度を3〜7℃に下げた後、化1で表されるポリエチレングリコール−ポリプロピレングリコール−ポリエチレングリコール(PEG−PPG−PEG)ブロック共重合体24〜50重量%を添加し、1〜10時間の間攪拌する段階と、を通じて製造されてもよい。
比較例1.
ゼラチン0.05重量%、キトサン0.3重量%を滅菌蒸留水に入れ、55〜60℃で150rpm、30分湯煎攪拌した。その後、グリセリン10重量%を入れて徐々に温度を4〜6℃に下げて同じ条件で攪拌した。温度が下がると、ポロキサマー188 17重量%、ポロキサマー407 15重量%を入れ、100rpm、3時間の間攪拌して癒着防止用高分子組成物を製造した。
ゼラチン0.05重量%、キトサン0.3重量%、100%酢酸0.01重量%を滅菌蒸留水に入れ、55〜60℃で150rpm、30分湯煎攪拌した。グリセリン10重量%を入れ、徐々に温度を4〜6℃に下げて同じ条件で攪拌した。温度が下がると、ポロキサマー188 17重量%、ポロキサマー407 15重量%を入れ、100rpm、3時間撹拌して癒着防止用高分子組成物を製造した。
酢酸0.01重量%の代わりに酢酸0.1重量%を使用したことを除いては、実施例1と同じ方法で癒着防止用高分子組成物を製造した。
酢酸0.01重量%の代わりに酢酸0.5重量%を使用したことを除いては、実施例1と同じ方法で癒着防止用高分子組成物を製造した。
酢酸0.01重量%の代わりに酢酸1重量%を使用したことを除いては、実施例1と同じ方法で癒着防止用高分子組成物を製造した。
酢酸0.01重量%の代わりに酢酸3重量%を使用したことを除いては、実施例1と同じ方法で癒着防止用高分子組成物を製造した。
酢酸0.01重量%の代わりに100%塩酸0.01重量%を使用したことを除いては、実施例1と同じ方法で癒着防止用高分子組成物を製造した。
酢酸0.01重量%の代わりに100%リン酸0.01重量%を使用したことを除いては、実施例1と同じ方法で癒着防止用高分子組成物を製造した。
酢酸0.01重量%の代わりに100%クエン酸0.01重量%を使用したことを除いては、実施例1と同じ方法で癒着防止用高分子組成物を製造した。
酢酸0.01重量%の代わりに100%乳酸0.01重量%を使用したことを除いては、実施例1と同じ方法で癒着防止用高分子組成物を製造した。
酸添加によるキトサン安定化は、実施例及び比較例で製造された癒着防止用高分子組成物を4℃、25℃及び40℃で1日、7日、28日、60日の間保管し、キトサンの析出の有無を観察する方法で評価した。
様々な酸の添加によって製品の特性である粘度に影響を及ぼすのか確認した。
癒着防止用高分子組成物に酸が添加されることにより、物質のpHに及ぼす影響を確認し、体内の使用に適したpHの範囲を設定するため、実施例及び比較例で製造された癒着防止用高分子組成物のpHを測定した。
様々な酸の添加により、癒着防止用高分子組成物の付着性能に影響を及ぼすのか確認した。
ラットの盲腸表面と腹膜に人為的な傷をつけて強制癒着を起こし、比較例1と実施例1によって製造した本発明の癒着防止用高分子組成物を傷部位に処理した後、1週間後にラットの盲腸と腹膜の癒着防止効果を観察した。癒着の程度は、0から5まで区分して評価(癒着の程度を示すスコア;0=癒着が発生しない場合、1=点状(focal)の癒着が一部発生した場合、2=点状の癒着が多い場合、3=面状の癒着が発生した場合、4=面状の癒着が深く発生した場合、5=面状の癒着とともに血管が形成された場合)し、その結果を図2に示した。
Claims (8)
- PEG−PPG−PEGブロック共重合体は、2種以上のPEG−PPG−PEGブロック共重合体の混合物であるものである、請求項1に記載の癒着防止用高分子組成物。
- 2種のPEG−PPG−PEGブロック共重合体を使用するとき、
一つのPEG−PPG−PEGブロック共重合体において、x及びzは、それぞれ独立して90〜110の整数であり、yは、40〜70の整数であり、
もう一つのPEG−PPG−PEGブロック共重合体において、x及びzは、それぞれ独立して75〜90の整数であり、yは、20〜40の整数であるものである、請求項2に記載の癒着防止用高分子組成物。 - 2種のPEG−PPG−PEGブロック共重合体を使用するとき、1つのPEG−PPG−PEGブロック共重合体及びもう一つのPEG−PPG−PEGブロック共重合体の含量は、それぞれ12〜25重量%であるものである、請求項2に記載の癒着防止用高分子組成物。
- 酸溶液において、酸は、塩酸、酢酸、アスコルビン酸、硝酸、硫酸、フッ酸、リン酸、クエン酸、乳酸、及びギ酸からなるグループから選ばれた一つ以上を含むものである、請求項1に記載の癒着防止用高分子組成物。
- 成長因子をさらに含み、
前記成長因子は、上皮細胞成長因子(Epidermal growth factors;EGF、beta−urogastrone)、ヘパリン結合性EGF類似成長因子(Heparin−binding EGF−like growth factor; HB−EGF)、形質転換成長因子−α(transforming growth factor−α;TGF−α)、及び繊維芽細胞成長因子(Fibroblast growth factors; FGFs)からなるグループから選ばれた一つ以上を含むものである、請求項1に記載の癒着防止用高分子組成物。 - 安定化剤をさらに含み、
前記安定化剤は、グリセロール、グリセリド類及びグリコール類からなるグループから選ばれた一つ以上を含むものである、請求項1に記載の癒着防止用高分子組成物。
