Claims (7)
- 生細胞および死細胞を含み、前記生細胞と前記死細胞とで励起光に対する蛍光の発生態様を互いに異ならせた蛍光標本を準備する準備工程と、
前記蛍光標本を蛍光撮像した蛍光画像を取得する蛍光画像取得工程と、
前記蛍光標本を明視野撮像した明視野画像を取得する明視野画像取得工程と、
前記蛍光画像から前記蛍光標本中の生細胞および死細胞それぞれの位置を、前記生細胞および前記死細胞それぞれが発する特有の蛍光色に対応する領域を検出する画像処理によって特定する位置特定工程と、
前記明視野画像に含まれる画像オブジェクトを画像処理により検出し、検出された前記画像オブジェクトのうち前記位置特定工程において特定された生細胞の位置に対応する位置にある前記画像オブジェクトを生細胞オブジェクトとして、また前記位置特定工程において特定された死細胞の位置に対応する位置にある前記画像オブジェクトを死細胞オブジェクトとしてそれぞれ抽出する抽出工程と、
前記生細胞オブジェクトおよび前記死細胞オブジェクトとして抽出された前記画像オブジェクト各々の特徴ベクトルを求め、前記特徴ベクトルおよび生死の特定結果を含む教師データを用いて機械学習を実行し分類器を構成する学習工程と
を備える、細胞の生死判定のための分類器構成方法。 - 前記蛍光画像取得工程では、蛍光を励起する励起光が照射された前記蛍光標本を撮像手段が撮像する一方、前記明視野画像取得工程では、照明光としての可視光が照射された前記蛍光標本を前記蛍光画像取得工程と同一の撮像手段、同一の撮像視野で撮像する請求項1に記載の分類器構成方法。
- 前記準備工程では、緑色蛍光タンパク質を導入して培養した細胞に死細胞染色試薬としての赤色蛍光色素を添加した前記蛍光標本を作製し、
前記蛍光画像取得工程では、前記蛍光標本のカラー画像を取得し、
前記位置特定工程では、前記カラー画像を分版したグリーン単色画像において所定以上の輝度を有する領域を前記生細胞とみなす一方、前記カラー画像を分版したレッド単色画像において所定以上の輝度を有する領域を前記死細胞とみなす請求項1または2に記載の分類器構成方法。 - 前記特徴ベクトルは、前記画像オブジェクトの輪郭線の形状に関連する輪郭線特徴量、および、前記画像オブジェクト内のテクスチャーに関連する前記テクスチャー特徴量の少なくとも一方を要素として含む請求項1ないし3のいずれかに記載の分類器構成方法。
- 前記テクスチャー特徴量は、高次局所自己相関特徴に関連する特徴量を含む請求項4に記載の分類器構成方法。
- 前記学習工程では、前記画像オブジェクトが分類される分類カテゴリとして、生細胞に対応する分類カテゴリと、死細胞に対応する分類カテゴリと、生細胞および死細胞のいずれでもないものに対応する分類カテゴリとを含む前記分類器を構成する請求項1ないし5のいずれかに記載の分類器構成方法。
- 細胞を含む試料を明視野撮像した処理対象画像を取得する原画像取得工程と、
前記処理対象画像から前記細胞に対応する画像オブジェクトを検出する検出工程と、
請求項1ないし6のいずれかに記載の分類器構成方法により構成された分類器を用いて、前記検出工程で検出された前記画像オブジェクトを分類し、分類結果に基づいて当該画像オブジェクトに対応する前記細胞の生死を判定する判定工程と
を備える細胞の生死判定方法。
