Claims (17)
- 細胞増殖システムにおいて複数の細胞を投入する方法であって、該方法は、
細胞増殖システムのバイオリアクター内の中空糸の内部及び前記バイオリアクターの中空糸の内部を流れる流体の循環路であるICループ内において第1レートで循環する流体に、複数の細胞を付加するステップと、
前記流体の循環レートを、所定の期間、前記第1レートに維持して、前記流体において前記複数の細胞を均一に懸濁させるステップと、
前記複数の細胞を均一に懸濁させた後、前記流体の循環レートを、前記第1レートよりも低いレートに減らし、所定の期間で、前記複数の細胞のうちの第1のパーセンテージである80%以下の量を前記バイオリアクター内に沈殿させ、前記複数の細胞のうちの第2のパーセンテージの量を前記バイオリアクターの外側に沈殿させるステップと、
前記バイオリアクター内において前記第1のパーセンテージの量の細胞を増殖するステップと、
前記第2のパーセンテージの量の細胞を廃棄するステップと、
を有する方法。 - 細胞増殖システムにおいて複数の細胞を投入する方法であって、該方法は、
細胞増殖システムのバイオリアクター内の中空糸の内部及び前記バイオリアクターの中空糸の内部を流れる流体の循環路であるICループ内において第1レートで循環する流体に、複数の細胞を付加するステップと、
付加した後、前記ICループ及びバイオリアクター内を循環する前記流体の循環レートを、前記第1レートから、前記第1レートよりも高い第2レートに増やすステップと、
前記流体の循環レートを、所定の期間、前記第2レートに維持して、前記流体において前記複数の細胞を均一に懸濁させるステップと、
前記複数の細胞を均一に懸濁させた後、前記流体の循環レートを、前記第2レートよりも低いレートに減らし、所定の期間で、前記複数の細胞のうちの第1のパーセンテージである80%以下の量を前記バイオリアクター内に沈殿させ、前記複数の細胞のうちの第2のパーセンテージの量を前記バイオリアクターの外側に沈殿させるステップと、
前記バイオリアクターにおいて前記第1のパーセンテージの量の細胞を増殖するステップと、
前記第2のパーセンテージの量の細胞を廃棄するステップと、

- 請求項1〜8のいずれか1項に記載の方法において、前記維持するステップにおいて、
前記バイオリアクターを、第1の姿勢である水平姿勢から、第1の姿勢から270°回転した第2の姿勢へ、回転軸を中心として回転させ、
前記バイオリアクターを、前記第2の姿勢で、第1の期間、休止させ、
前記バイオリアクターを、前記回転軸を中心として回転させて、前記第1の姿勢に戻し、
前記バイオリアクターを、前記第1の姿勢で、第2の期間、休止させる、
ことを特徴とする方法。 - 請求項9記載の方法において、前記第2の期間は、前記第1の期間と略等しいことを特徴とする方法。
- 請求項9記載の方法において、前記バイオリアクターを回転させる際の前記バイオリアクターの角速度(rad/秒)をωとした場合、前記第2の期間は、2ω-1に略等しいことを特徴とする方法。
- 請求項9記載の方法において、前記バイオリアクターを、前記第1の姿勢と前記第2の姿勢との間で回転させる際は、第1角速度が維持されることを特徴とする方法。
- 請求項12記載の方法において、前記バイオリアクターを、前記回転軸を中心として回転させて、前記第1の姿勢に戻す際は、第2角速度が維持され、前記第2角速度は、前記第1角速度と略等しいことを特徴とする方法。
- 複数の細胞を増殖するためのシステムであって、該システムは、
少なくとも両端部を有する第1流体流路を備えるバイオリアクターと、
前記第1流体流路と流体的に関連付けられた流体入口路と、
前記バイオリアクターの前記第1流体流路において流体を循環させる第1ポンプと、
前記第1ポンプの動作を制御するコントローラと、
を備え、
前記第1流体流路の第1端部は、中空糸膜の第1ポートと流体的に関連付けられ、前記第1流体流路の第2端部は、前記中空糸膜の第2ポートと流体的に関連付けられ、前記第1流体流路は、前記中空糸膜の毛細管内側部分を有し、
前記流体入口路を介して、前記第1流体流路には、複数の細胞が導入され、
前記コントローラは、前記第1ポンプを制御することによって、
前記第1流体流路内において、流体を、第1レートにて循環させ、
前記流体の循環レートを、所定の期間、前記第1レートに維持して、前記流体において前記複数の細胞を均一に懸濁させ、
前記複数の細胞を均一に懸濁させた後、前記流体の循環レートを、前記第1レートよりも低いレートに減らし、前記複数の細胞のうちの第1のパーセンテージである80%以下の量を前記バイオリアクター内に沈殿させ、前記複数の細胞のうちの第2のパーセンテージの量を前記バイオリアクターの外側に沈殿させ、
成長培地を有する流体を循環させて、前記バイオリアクターにおいて前記第1のパーセンテージの量の細胞を増殖させ且つ前記第1流体流路から前記第2のパーセンテージの量の細胞の大部分を廃棄することを特徴とするシステム。 - 請求項14記載のシステムにおいて、前記第1のパーセンテージは、前記複数の細胞の約65%以上であることを特徴とするシステム。
- 請求項14又は15に記載のシステムにおいて、前記第1レートは、約145ml/分以上であることを特徴とするシステム。
- 請求項14〜16のいずれか1項に記載のシステムにおいて、前記第1レートは、約250ml/分以下であることを特徴とするシステム。
