Description
(a)1以上のバイオリアクター2、32、52と、
(b)保持液入口4、34、54と、保持液出口5、35、55と、1以上の透過液出口ポート6、36、56とを備える1以上のフィルターユニット3、33、53と、
(c)保持液入口に流体接続されている1以上の往復ポンプ7、37、57とを備える。
(a)上記のシステム1、31、51を準備するステップ、
(b)細胞培地と細胞をバイオリアクター2、32、52に添加するステップ、
(c)撹拌下バイオリアクター内で細胞を培養するステップ、
(d)培養中、往復ポンプ7、37、57を運転して、(i)バイオリアクターから液体をチューブの所定の長さ部分11、41、61、分岐点10、40、60、及び入口逆止弁8、38、58を介してフィルターユニット3、33、53の保持液入口4、34、54へと引き出し、(ii)液体をフィルターユニットの保持液出口5、35、55から出口逆止弁9、39、59、分岐点10、40、60、及びチューブの所定の長さ部分11、41、61を介してバイオリアクター2、32、52に戻すステップ、及び
(e)透過液を透過液出口ポート6、36、56から回収するステップを含む。
(a)1以上のバイオリアクター(2、32、52)と、
(b)1以上の往復ポンプ(7、37、57)と、
(c)1以上の無菌コネクタを介して該往復ポンプに流体接続され、該バイオリアクターに流体接続される1以上のフィルターユニット(3)とを備える。
Claims (10)
- 細胞の灌流培養システム(1、31、51)であって、当該システムが、
(a)1以上のバイオリアクター(2、32、52)と、
(b)保持液入口(4、34、54)と、保持液出口(5、5、55)と、1以上の透過液出口ポート(6、36、56)とを備える1以上のフィルターユニット(3)と、
(c)保持液入口に流体接続されている1以上の往復ポンプ(7、37、57)と
を備えており、
保持液入口が、バイオリアクターから保持液入口への方向の流れを許容するが逆方向の流れを防ぐように構成された入口逆止弁(8、38、58)を介してバイオリアクターに流体接続され、保持液出口が、保持液出口からバイオリアクターへの方向の流れを許容するが逆方向の流れを防ぐように構成された出口逆止弁(9、39、59)を介してバイオリアクターに流体接続されており、
入口逆止弁及び出口逆止弁が各々チューブ分岐点(10、40、60)に流体接続され、分岐点がさらに、一本のチューブの所定の長さ部分(11、41、61)を介してバイオリアクターに接続されている、システム。 - 往復ポンプがダイヤフラムポンプであり、システムが、往復ポンプ(37)内の駆動流体圧力を測定するように構成された駆動流体圧力変換器(46)を備える、請求項1記載のシステム。
- さらに、入口逆止弁(8、38、58)と分岐点(10、40、60)との間に入口制御弁(42、63)及び/又は出口逆止弁(9、39、59)と分岐点(10、40、60)との間に出口制御弁(43、64)を備える、請求項1又は請求項2記載のシステム。
- さらに、保持液入口の圧力を測定するように構成された入口圧力変換器(44)を備え、保持液出口の圧力を測定するように構成された出口圧力変換器(45)を備える、請求項1乃至請求項3のいずれか1項記載のシステム。
- さらに、入口圧力変換器(44)及び出口圧力変換器(45)のうちの少なくとも1つから受信した圧力データにより、往復ポンプ(37)、入口制御弁(42)及び出口制御弁(43)のうちの少なくとも1つを制御するように構成された、1以上の制御ユニット(47)を備えており、制御ユニットが、入口圧力変換器から受信した圧力データにより、入口制御弁及び往復ポンプのうちの少なくとも1つを制御するように構成されている、請求項4記載のシステム。
- さらに、1以上の制御ユニット(47)に接続された、透過液ポンプ(50)及び透過液圧力変換器(49)を備える、請求項1乃至請求項5のいずれか1項記載のシス
テム。 - 細胞の灌流培養方法であって、
(a)請求項1乃至請求項6のいずれか1項記載のシステム(1、31、51)を準備するステップ、
(b)細胞培地と細胞をバイオリアクター(2、32、52)に添加するステップ、
(c)撹拌下、バイオリアクター内で細胞を培養するステップ、
(d)培養中、往復ポンプ(7、37、57)を運転して、(i)バイオリアクターから液体を一本のチューブの所定の長さ部分(11、41、61)、分岐点(10、40、60)及び入口逆止弁(8、38、58)を介してフィルターユニット(3、33、53)の保持液入口(4、34、54)へと引き出し、(ii)液体をフィルターユニットの保持液出口(5、35、55)から出口逆止弁(9、39、59)、分岐点(10、40、60)及び一本のチューブの所定の長さ部分(11、41、61)を介してバイオリアクター(2、32、52)に戻すステップ、及び
(e)透過液を透過液出口ポート(6、36、56)から回収するステップ
を含む方法。 - フィルターユニットが、精密濾過膜を備えていて透過液が細胞により発現された目的物質を含んでいるか、或いは限外濾過膜を備えていて細胞により発現された目的物質を保持液中に保持したままバイオリアクターに戻す、請求項7記載の方法。
- さらに、i)往復ポンプ(37、57)及び入口制御弁(42、63)のうちの少なくとも1つを制御して、往復ポンプ(37、57)の外側ストローク中、入口圧力変換器(44)の圧力読み値を所定範囲内に維持すること、及び/又はii)入口圧力変換器(44)及び出口圧力変換器(45)から得られるデータから膜差圧を計算し、往復ポンプ(57)及び/又は入口制御弁(42)及び出口制御弁(43)のうちの少なくとも1つを制御して、膜差圧を予め設定した上限値と下限値との間に維持することを含む、請求項7又は請求項8記載の方法。
- さらに、透過液ポンプ(50)を逆方向に運転してフィルター膜の逆フラッシュを生じさせ、透過液圧力変換器(49)からの入力データを用いて制御ユニット(47)で制御することを含む、請求項7乃至請求項9のいずれか1項記載の方法。
