マンゴスチン果皮(Pericarp)1kgを、25℃の換気ドライオーブンで乾燥させて水分含有量10%以下の乾燥物250gにした後、これを破砕機を用いて粉砕して、粒子サイズ400mesh〜500meshのマンゴスチン果皮粉砕物230gを得た。

分析の結果、マンゴスチン果皮400mesh〜500meshの粉砕物中、アルファ−マンゴスチンの含有量は3.0±1.0%であり、ガンマ−マンゴスチンの含有量は0.2±0.05%と確認された。
前記製造例1によるマンゴスチン果皮粉砕物の製造工程で得られたマンゴスチン果皮粉砕物1kgを、20L反応槽で10Lのエタノールを用いて、60℃の条件で4時間抽出した後、1.0μmのろ過紙を用いてろ過し、そのろ液を40℃で減圧濃縮して、150gの濃縮物を得た。
下記表2に記載された配合比率に応じてマンゴスチン抽出物が可溶化された水溶性マンゴスチン分子カプセルを製造した。具体的には、<製造例2>の方法で得られた脂溶性マンゴスチン抽出物150gにガンマ−シクロデキストリン45gを混合して、70℃の温度、0.4気圧の真空下、撹拌速度1000rpmで2時間撹拌して、マンゴスチン抽出物を抱接させた。抱接されたマンゴスチン抽出物をさらに安定化するためにアルギニン22gを添加して、4℃以下で6時間保管してカプセルの相安定化を行う。その後、この溶液を凍結乾燥(−40℃から40℃昇温、昇温条件:1℃/hr)により粉末化した。
製造例3で製造された水溶性マンゴスチン粉末430gに、プロポリス抽出物210g(液状)、大豆不けん化物120g、塩化リゾチーム150g、ビタミンC50g、ビタミンE10g、キシリトール30gを入れて、粉末混合器に入れてよく混合した後、真空乾燥機(60℃、0.4気圧、8時間)でよく乾燥後、粉砕して粉末に製造する。
一般に、新規の抗菌剤が開発された時、その抗菌剤の有用性を判定する基準となる試験方法として、最小抑制濃度(minimum lnhibitory concentration:以下、MICという)試験と時間対比の殺菌力(time−kill assay)試験とがある。
ストレプトコッカスミュータンス(Streptococcus mutans ATCC25175)に対するMIC試験は、米国の臨床試験標準方法であるNCCLS(National Committee for Clinical Laboratory Standards)により進行させた。マンゴスチン抽出物がストレプトコッカスミュータンス菌に対する抗菌性を比較試験するために、それぞれの分析試料を、次の表5のように、各分析試料の濃度が0、1、2、3、4、5、6、7、8、9および10ulとなり、使用するストレプトコッカスミュータンス菌の濃度が50重量%の比率として最終的に1×105となるようにし、分析試料溶解剤として使用されたDMSO(ジメチルスルホキシド)の濃度が最終的に1重量%となるようにし、残りの40重量%をBHI培地(Brain heart infusion:Peptic Digest of Animal tissue、Sodium Chloride、Dextrose、Pancreatic Digest of Gelation、Disodium Phosphate、Final PH7.4+−0.2;Difco社)を加えて製造した後、試験に使用した。
ストレプトコッカスミュータンス(Streptococcus mutans ATCC25175)に対する分析試料のTime−kill assay実験は、NCCLS(National Committee for Clinical Laboratory Standards)に基づく方法のうち、McFarland Standardにより最初O.D550nm値対比の菌数を換算する方法によって施行した。
ポルフィロモナスジンジバリス(Porphyromonas gingivalis CCARM0145)に対するMIC実験は、NCCLS(National Committee of Clinical Laboratory Standards、1997)の固体培地希釈法(Agar dilution method)により行われた。
本発明に係るマンゴスチン抽出物の抗菌性をより詳細に測定するために、韓国人の口腔から分離同定された歯周疾患原因菌であるPrevotella intermedia KCOM1107、Fusbacterium nucleatum subsp.polymorphum KCOM1232、Aggregatibacter actinomycetemcomitans KCOM1306およびPorphyromonas gingivalis ATCC33277Tを対象に最小殺菌濃度(Minimum bactericidal concentration(MBC))を測定して、マンゴスチン果皮抽出物の抗菌性を測定した。
Claims (13)
- マンゴスチン抽出物、プロポリス、塩化リゾチーム、大豆不けん化物、ビタミンC、キシリトール、ビタミンE及び分子カプセルコーティング剤としてガンマ−シクロデキストリンを含み、
前記マンゴスチン抽出物が、マンゴスチン果皮の、水、炭素数1〜6のアルコール、および水と炭素数1〜6のアルコールとの混合溶媒からなる群より選択された1種以上の溶媒の抽出物であり、
アルファ−マンゴスチンを7.5〜9.0mg/1投与単位の含量で含む、
1日1回投与用のプレボテラインターメディア及び/又はフゾバクテリウムヌクレアタムによる歯周疾患の予防または治療用薬学組成物。 - 前記炭素数1〜6のアルコールは、エタノールであることを特徴とする請求項1に記載の歯周疾患の予防または治療用薬学組成物。
- 前記マンゴスチン抽出物は、全組成物の重量に対して30〜50重量%含まれるものである請求項1に記載の歯周疾患の予防または治療用薬学組成物。
- 全組成物の重量に対して、プロポリス15〜25重量%、塩化リゾチーム10〜20重量%、大豆不けん化物7〜13重量%、ビタミンC3〜10重量%、キシリトール1〜7重量%およびビタミンE0.5〜2重量%を含むことを特徴とする請求項1に記載の歯周疾患の予防または治療用薬学組成物。
- 海藻粉末、乾燥酵母およびクロム酵母からなる群より選択される1種以上の成分を追加的に含むものである請求項1に記載の歯周疾患の予防または治療用薬学組成物。
- 薬学的に許容可能な担体をさらに含むものである請求項1に記載の歯周疾患の予防または治療用薬学組成物。
- マンゴスチン抽出物、プロポリス、塩化リゾチーム、大豆不けん化物、ビタミンC、キシリトール、ビタミンE及び分子カプセルコーティング剤としてガンマ−シクロデキストリンを含み、
前記マンゴスチン抽出物が、マンゴスチン果皮の、水、炭素数1〜6のアルコール、および水と炭素数1〜6のアルコールとの混合溶媒からなる群より選択された1種以上の溶媒の抽出物であり、
アルファ−マンゴスチンを7.5〜9.0mg/1投与単位の含量で含む、
1日1回投与用のプレボテラインターメディア及び/又はフゾバクテリウムヌクレアタムによる歯周疾患の予防または改善用食品組成物。 - 前記炭素数1〜6のアルコールは、エタノールであることを特徴とする請求項7に記載の歯周疾患の予防または改善用食品組成物。
- 前記マンゴスチン抽出物は、全組成物の重量に対して30〜50重量%含まれるものである請求項7に記載の歯周疾患の予防または改善用食品組成物。
- 全組成物の重量に対して、プロポリス15〜25重量%、塩化リゾチーム10〜20重量%、大豆不けん化物7〜13重量%、ビタミンC3〜10重量%、キシリトール1〜7%およびビタミンE0.5〜2重量%を含むことを特徴とする請求項7に記載の歯周疾患の予防または改善用食品組成物。

アルファ−マンゴスチンを7.5〜9.0mg/1投与単位の含量で含む、
1日1回投与用のプレボテラインターメディア及び/又はフゾバクテリウムヌクレアタムによる歯周疾患の予防または改善用口腔衛生組成物。 - 前記組成物の剤形が、歯磨き粉、口腔清浄剤、口腔洗浄剤または歯茎マッサージクリームである請求項12に記載の歯周疾患の予防または改善用口腔衛生組成物。
