疼痛伝達能のために、ニューロンはこれまでに開発された全ての薬物療法の意図的な主要標的となってきた。概して、オピオイドはニューロンのオピオイド受容体で作用するだけで疼痛を変調し、かつオピオイドのアンタゴニストも同様にニューロン上でのみその効果を発揮すると考えられる。さらに、オピオイドの有害作用(例えば耐性、痛覚過敏、依存及び報酬等)及び有益作用(例えば鎮痛、咳止め等)はニューロンのオピオイド受容体に依存した、非常に類似した潜在的に切り離すことのできない機構により媒介されると従来より考えられている。
本発明の幾つかの態様は、式:

本発明の化合物を、利用可能な出発材料から容易に調製することができる。本発明の化合物における様々な置換基は、出発化合物に存在していても、既知の置換の方法又は変換反応により中間体のいずれか1つに付加しても、又は最終生成物の形成後に付加してもよい。例えば、ニトロ基をニトロ化により付加することができ、ニトロ基を他の基、例えば還元によりアミノ基、並びにアミノ基のジアゾ化及びジアゾ基のハロゲンへの置き換えにより、又は単純にハロゲン化反応によりハロゲンへと変換することができる。アシル基をフリーデル・クラフツアシル化により付加することができる。それからウォルフ・キッシュナー還元及びクレメンゼン還元を含む様々な方法により、アシル基を対応するアルキル基に変換することができる。アミノ基をアルキル化して、モノアルキルアミノ基及びジアルキルアミノ基を形成することができ、メルカプト基及びヒドロキシ基をアルキル化して、対応するエーテルを形成することができる。第1級アルコールを当該技術分野で既知の酸化剤により酸化して、カルボン酸又はアルデヒドを形成することができ、第2級アルコールを酸化して、ケトンを形成することができる。このようにして、置換反応又は修飾(alteration)反応を利用して、単離生成物を含む、出発材料、中間体、又は最終生成物の分子を通じて多種多様な置換基を得ることができる。

仮想スクリーニング法には2つの段階が含まれていた:静的タンパク質モデルへのドッキング及び動的タンパク質モデルを用いた精密化。静的タンパク質モデル及び動的タンパク質モデルへのドッキングはQuantumのソフトウェアユーティリティを利用して行った。静的タンパク質モデルへのドッキングには、最小結合エネルギーによる結合ポケットにおけるリガンド位置の特定と、結合エネルギーの推定とが含まれていた。分子動力学研究では、算出したタンパク質−リガンドの結合エネルギーをタンパク質柔軟性に関して精密化した。使用する精密化法は水性環境でのタンパク質−リガンド複合体全体に関する完全な自由エネルギー摂動分子動力学計算の実行であった。このためこの精密化法は、タンパク質及びリガンドの両方の柔軟性に関するものである。
全ての反応は乾燥窒素又はアルゴン雰囲気下で炉乾燥ガラス器具又は火炎乾燥ガラス器具中で行った。メタノールを単蒸留により蒸留し、4Åのモレキュラーシーブ上で保存した。アセトンは蒸留してから使用した。メチルアミン・HCl塩を乾燥剤(decadent)としてP2O5を使用して高真空下で一晩乾燥させた。他の試薬及び溶媒は全て、供給業者から受け取ったままのものを用した。フラッシュクロマトグラフィを32μm〜64μmのシリカゲルを使用して行った。1H NMRスペクトルを、内部標準として残留CHCl3(7.26ppm)を使用して、CDCl3中で300MHz、400MHz又は500MHzで記録した。13C NMRスペクトルを、内部標準として残留CHCl3(77.23ppm)を使用して、CDCl3中で75MHzで記録した。正確な質量をエレクトロスプレーイオン化を用いて決定した。
4−エトキシフェノール(0.25g、1.81mmol)、無水炭酸カリウム(0.50g、3.62mmol)及びエピクロロヒドリン(0.57ml、7.24mmol)をアセトン(4.52ml)に添加し、得られた不均一溶液を16時間還流した。混合物を室温まで冷却し、セライトパッドで濾過して、濾液を減圧下で濃縮した。得られた油状物質(oil)をトルエン(20mL)に溶解し、水(15mL)、5%NaOH水溶液(20mL)及び再び水(20mL)で連続して洗浄した。有機層をMgSO4を用いて乾燥させ、減圧下で濃縮し、白色固体として0.292g(83%)の2−((4−エトキシフェノキシ)メチル)オキシランを得た(mp=41℃)。1H NMR(400MHz、CDCl3)δ 6.91〜6.77(m、4H)、4.17(dd、J=11.0、3.2、1H)、3.98(q、J=7.0、2H)、3.91(dd、J=11.0、5.6、1H)、3.34(m、1H)、2.90(dd、J=4.9、4.1、1H)、2.75(dd、J=5.0、2.7、1H)、1.39(t、J=6.98、3H)。13C NMR(75MHz、CDCl3)δ 153.72、152.78、115.90、115.90、115.59、115.59、69.71、64.18、50.49、44.98、15.15。HRMS(m/z):[MNa]+ C11H14O3Na+に関する計算値 217.08;実測値 217.0826。
水酸化カリウム粉末(1.751g、31.2mmol)を3,5−ジメチルピラゾール(2g、20.81mmol)の無水DMSO溶液(10.40ml)に添加し、得られた不均一溶液を80℃で1.5時間攪拌した後、室温まで冷却した。それから2−クロロベンジルクロリド(2.64ml、20.81mmol)を15分かけて6MのDMSOに添加し、溶液を更に1.5時間攪拌した。TLCにより完了を観察した後、反応物を水に注ぎ、得られた水相を2つの20mLのCHCl3で抽出した。合わせた有機層を100mLの水で洗浄し、無水MgSO4で乾燥させ、減圧下で濃縮し、透明な液体として4.55g(99%)の1−(2−クロロベンジル)−3,5−ジメチル−1H−ピラゾールを得た。1H NMR(300MHz、CDCl3)δ 7.41〜7.31(m、1H)、7.24〜7.09(m、2H)、6.59〜6.50(m、1H)、5.90(s、1H)、5.31(s、2H)、2.26(s、3H)、2.15(s、3H)。13C NMR(75MHz、CDCl3)δ 148.32、139.96、135.46、131.96、129.42、128.76、127.72、127.48、105.84、50.12、13.80、11.15。HRMS(m/z):[MNa]+ C12H13ClN2Na+に関する計算値 243.07;実測値 243.0651。
調製され、試験される代表的な化合物の一部を以下に挙げる(塩及び鏡像異性的に濃縮された異性体も一部調製されたが、別個には示していない):

ドッキング研究を、AutoDock 4.0. Lamarckian Generic Algorithm(LGA)を用いて行い、リガンドのねじれ角はAUTOTORSを用いて変化させた。ドッキング実験に関する他の手順は全て、AutoDock 4.0プログラムのユーザーマニュアルに記載されている通りに従った。ドッキングさせた立体構造を、力場スコアリング関数を用いてAutoDock 4.0プログラムにより自動的にランク付けした。総数100個の異なる立体構造クラスタを、1.0Åのrmsd耐性を用いて100回の実行から見出した。これらの中で、最高のランクを得た3つのドッキング構造のうちの1つを分子の可視化に使用した。
HeLa細胞を、10%ウシ胎児血清(FBS)を添加したダルベッコ変法イーグル培地(DMEM)中で8×106個/mlの密度まで成長させ、ダルベッコPBS/1.25% DMSO中の20μgのFlag−sTLR4及び10μgのMD−2−Flag−Hisを用いてエレクトロポレーション(250V、960μF)によりトランスフェクトした。プラスミドはファビオ研究所から無償で提供された。回収及び細胞接着が可能となるように、細胞を、FBSを含有するDMEMの入った10cmプレートで再び平板培養した。6〜8時間後、細胞培地を血清無含有培地293 SFM2(Invitrogen, CA, USA)に交換した。培地を24時間後に回収した。Hisタグ付けタンパク質複合体を捕捉するために、濾過した培地をProBondニッケル樹脂(Invitrogen, CA, USA)とともに一晩インキュベートした。それから樹脂をリン酸緩衝生理食塩水(PBS)で洗浄し、レムリ試料バッファー中で再懸濁して、沸騰させ、SDS−PAGE及び抗Flag mAbを用いた免疫ブロット法により分析した。TLR4/MD−2相互作用に対する小分子の効果を、DMSOに溶解した化合物(対照では同量のDMSO)を細胞に添加した後、一晩インキュベートすることにより評価した。
TLR4シグナル伝達により、3つの並行した細胞内シグナル伝達経路の同時活性化がもたらされる。これらのうちの2つ(NF−κB及びMAPKを介する)は、TLR4活性化により誘導される炎症誘発性応答に主に関与すると考えられ、第3の並行経路(PI3K/Akt1)は細胞生存、アポトーシス及び細胞運動性との関連がより強いと考えられる。3つの経路は全てTLR4でのアゴニズムにより活性化されるので、これらのいずれか1つをTLR4活性化の反映として使用することができる。緑色蛍光タンパク質(GFP)タグ付けAkt1を発現するために安定してトランスフェクトしたRAW264.7マウスマクロファージ細胞株が利用可能であることを考慮して、GFP−Akt1の動員及びサイトゾルクリアランスをTLR4活性化の指標として使用した。リポ多糖(LPS、大腸菌、血清型:0111:B4)をSigma(St. Louis, MO, USA)から入手した。細胞を10% FBS、10×ペニシリン/ストレプトマイシン及び10×l−グルタミンを添加した成長培地中で成長させ、それから35mm MatTekガラス底皿(Ashland, MA, USA)上の成長培地中で2×105細胞/mLの密度で18時間平板培養した後、画像化した。画像化の直前、成長培地をプレートから取り出し、pH7.4へと緩衝させた25mMのHEPESを添加したハンクス緩衝生理食塩水溶液(HBSS)1mLで2回洗浄し、加温しコンディショニングした画像化ハンクス緩衝培地(培地はRAW264.7細胞との24時間のインキュベートによりコンディショニングした)1mLに交換した。画像化を、60×油浸対物レンズと、対応する単一バンドの励起フィルター及び発光フィルターを有するGFP/RFPダイクロイックミラー(Chroma Technology, VT, USA)と、CoolSNAP ESカメラ(Photometrics,Tucson, AZ, USA)とを備えるNikonの倒立顕微鏡(Melville, NY, USA)で行った。水銀灯により励起が与えられた。画像を7.5秒ごとに取り込んだ。ベースラインの蛍光を5フレームで取り込んだ後、ビヒクル又はアンタゴニストを200μlで添加した。画像化を更に20フレーム続け、この時点でLPS又は試験アゴニスト(200μl)を適用し、更に20フレームでモニタリングした。目で見える応答が得られなければ、C5a(200μl)をプレートに添加し、細胞が応答性であるかを確認した。GFP−Akt1サイトゾルクリアランスを、ImageJを用いて定量化し、経時的な細胞質の蛍光の正規化変化として表した。
RAW細胞を10% FBS、ペニシリン(100U/ml)、ストレプトマイシン(100mg/ml)及びL−グルタミン(2mM)を添加したDMEM中で成長させた。それからRAW細胞を1ウェル当たり100000個の細胞で96ウェルプレートに植え、先に記載の培地中で24時間成長させた。24時間後に培地を取り出し、マクロファージ−SFM(Invitrogen, CA, USA)に交換した。レーンを適切なTLR特異的リガンドでドープした:LPS(リポ多糖)、ポリ(I:C)(ポリイノシン酸−ポリシチジル酸)、FSL−1((S,R)−(2,3−ビスパルミトイルオキシプロピル)−Cys−Gly−Asp−Pro−Lys−His−Pro−Lys−Ser−Phe)、R848(4−アミノ−2−(エトキシメチル)−α,α−ジメチル−1H−イミダゾ[4,5−c]キノリン−1−エタノール)及びPam3CSK4(N−パルミトイル−S−[2,3−ビス(パルミトイルオキシ)−(2RS)−プロピル]−[R]−システイニル−[S]−セリル−[S]−リシル−[S]−リシル−[S]−リシル−[S]−リシン.3HCl)を使用し、それぞれTLR4、TLR3、TLR2/6、TLR7及びTLR2/1を選択的に活性化した。それぞれのリガンドに関して2つのレーン(1つはリガンドのみを含有し、もう1つはリガンドと300nMの化合物A−1とを有する)を作製した。それからプレートを24時間インキュベートした。インキュベーション後、培地100μLを取り出し、平らなブラック96ウェルマイクロフルオロプレート(Thermo scientific, MA, USA)に添加した。2,3−ジアミノナフタレン(0.62MのHCl中、0.05mg/ml)10μLをそれぞれのウェルに添加し、15分間インキュベートした。反応を3MのNaOH 5μLの添加によりクエンチし、プレートを、Beckman CoulterのDTX880リーダー(Beckman Coulter,CA, USA)で、365nmでの励起及び450nmでの発光により読み取った。亜硝酸(一酸化窒素の安定した代謝産物)濃度を亜硝酸の標準曲線から決定した。
SEAPアッセイの材料をApplied Biosystems(CA, USA)から入手し、製造業者の仕様書に従って利用した。TLR4及び分泌型アルカリホスファターゼ(SEAP)レポーター遺伝子を安定してトランスフェクトしたヒト胚腎臓293(HEK293)細胞をInvivogen(CA, USA)から入手した。細胞を10%ウシ胎児血清、10×ペニシリン/ストレプトマイシン、10×l−グルタミン、1×ノルモシン(ant−nr−1)及び1×HEK Blue(hb−sel)を添加したDMEM培地中で培養した。細胞を96ウェルプレートにおいて37℃で24時間植え付け後、薬物処理を行った。処理日に、培地を96ウェルプレートから取り出し、0.5% DMSOとともに1ng/mL〜20ng/mLのLPS及び0.2μM〜50.0μMの薬物又は3ng/mL〜400ng/mLのLPS−RSを含有する脳脊髄液(CSF)バッファー(124mMのNaCl、5mMのKCl、0.1mMのCaCl2、3.2mMのMgCl2、26mMのNaHCO3及び10mMのグルコース(pH7.4))に交換した。
ヒト胚腎臓293(HEK293)細胞に、TLR4及び必要なアセンブリ及びシグナル伝達タンパク質(MD2、CD−14、LPSBP等)を安定してトランスフェクトした。細胞を、10% FBSと、ペニシリン(100U/ml)と、ストレプトマイシン(100mg/ml)と、L−グルタミン(2mM)と、0.1mg/mlのノルモシン(InvivoGen, CA, USA)と、1×HEK Blue選択試薬(InvivoGen,CA, USA)とを添加したDMEM中で培養した。細胞を6cmプレートに植え付け、37℃でインキュベートすることにより65%〜75%の密集度まで成長させた後、薬物処理した。処理日に、培地を6cmプレートから取り出し、薬物処理を加えた脳脊髄液(CSF)バッファーに交換した。37℃で24時間のインキュベーション後、CSFを取り出し、細胞を0.05% トリプシン+0.2g/lのEDTA(Invitrogen, CA, USA)でかき混ぜ、新たなDMEMを添加した培地中で再懸濁した。再懸濁後、試料100μlをそれぞれの6cmプレートから採取し、0.4% Trypan Blue(Sigma, MO, USA)100μlと穏やかに混合し、5分間静置した。それから青色染色した細胞数と総細胞数との比を、NikonのTMS光学顕微鏡(Nikon Instrumentals, CA USA)下で深さが0.1mmのBright Line血球計(VWR, PA, USA)を使用して定量化した。
ネズミの小膠細胞株、BV−2を、Primaria処理したフラスコ(Falcon, BD Bioscience, CA, USA)内の10% FBSを添加したDMEM培地中で成長させた。約80%の密集度に到達したら、細胞をトリプシン消化によりフラスコから引き剥がした。SMARTpoolのsiRNA(Dharmacon, Lafayette, CO, USA)ストック溶液(50μM)6μLをD−PBS 14μlで希釈し、リポフェクタミンLTX(Invitrogen, Carslbad , CA, USA)8μlをD−PBS 12μlで希釈した。続いてTLR4 siRNAとリポフェクタミンLTX溶液とを6ウェルプレートのウェル内で穏やかに混合し、室温で30分間インキュベートした。それから細胞を1ml当たり5×104個の細胞の細胞密度で6ウェルプレートに植えた。48時間のRNAi後、200μMのモルヒネを添加した。それからプレートを更に24時間インキュベートした。その後、細胞を回収し、M−PER哺乳動物タンパク質抽出試薬(Thermo Scientific, Rockford, IL, USA)で溶解した。バックグラウンド炎症性因子に対するMD−2又はTLR4を下方調節する効果を研究するために、細胞を、72時間のRNAiの後に回収した。IL−1β及びTNF−αのレベルを製造業者の取扱説明書に従ってELISA(BD Biosceince, San Diego, CA, USA)により分析した。
BV−2細胞をPrimaria処理したフラスコ(Falcon, BD Bioscience, CA, USA)内の10% FBSを添加したDMEM培地中で成長させた。約80%の密集度に到達したら、細胞をトリプシン消化によりフラスコから引き剥がした。それから細胞を1ウェル当たり4×105個の細胞で6ウェルプレートに植え、24時間成長させた。24時間後、培地を取り出し、1% FBSを添加したDMEMに交換し、モルヒネ(200μM)を添加した。加えて、化合物1、化合物2又は化合物3(10μM)をモルヒネと同時に又は単独で投与した。それからプレートを更に24時間インキュベートした。その後、細胞を回収し、M−PER哺乳動物タンパク質抽出試薬(Thermo Scientific, Rockford, IL, USA)で溶解した。IL−1βを製造業者の取扱説明書に従ってELISA(BD Biosceince, San Diego, CA, USA)により分析した。
病原体を含まない成体スプラーグ・ドーリー雄ラット(それぞれの実験で1群当たりラット数n=5〜6、300gm〜375gm、Harlan Labs, Madison, WI, USA)を全ての実験で使用した。ラットを、標準的な齧歯動物の食餌及び水を自由に入手することができる、温度(23+3℃)及び光(12時間:12時間 明:暗サイクル、0700に点灯)を制御した部屋に収容した。群の割り当てに関しては全ての試験をブラインドで行った。ラットは適切な試験環境に対して60分間の馴化を少なくとも3回行った後、行動試験を実施した。尾に適用した熱刺激に対する行動応答の閾値を、改良ハーグリーブス試験を用いて評価した。要するに、ベースライン引き抜き(withdrawal)値を、15分間隔で測定した尾の2つの連続した引き抜き潜時の平均から算出した。ベースラインの熱刺激に対する潜時は2秒〜3秒の範囲であり、10秒のカットオフ時間は組織損傷を避けるために与えられた。ベースライン引き抜き潜時の評価を薬物投与前、及び更に薬物投与後に経時的に行った。供試薬物と同量のビヒクルを投与した。
炎症の全血モデルにおけるサイトカインIL−6、IL−8、(TNFα)及びIL1βの分泌に対する影響に関して本発明の化合物によるTLR4阻害の試験を、(i)制限構成における2126−HClの2つの変異体を比較するためのLPSと、(ii)TLR4/MD2、TLR2の両方を伴う先天性免疫応答、及び補体系(抗CD14及びコンプスタチンと比較する)を開始させるためのグラム陰性細菌とを使用して、Mollnes et al. (Blood, 2002, 100, 1869-1877)により記載された手順を用いて行った。

- Toll様受容体(TLR)活性化と関連する臨床病態に関して被験体の治療剤であって、請求項1に記載の化合物を該被験体に投与することを含む、Toll様受容体(TLR)活性化と関連する臨床病態に関して被験体の治療剤。
- 臨床病態が神経因性疼痛、急性オピオイド鎮痛、若しくは望ましくないオピオイド副作用、又はそれらの組合せを含む、請求項7に記載の治療剤。
- 臨床病態が、慢性疼痛、侵害受容、急性オピオイド鎮痛、若しくは望ましくないオピオイド副作用、胃腸病変、心血管疾患、糖尿病、免疫関連病態、全身性病変、神経変性、陣痛誘発、発熱、発作、癲癇、癲癇発生、又はそれらの組合せを含む、請求項7に記載の治療剤。
