Description
実施例に入る前に、蛋白質溶液の吐出が困難であることの、より一層の理解のため、蛋白質のみをサーマルインクジェット方式で吐出させた場合の吐出量を示す。蛋白質溶液はウシ血清アルブミン(BSA)をリン酸バッファー(PBS)に溶解させたものを用い、各濃度にてバブルジェット(登録商標)プリンター(商品名PIXUS950i;キヤノン(株)社製)を溶液が回収できるよう改造した液体吐出装置を用いて吐出した。純水を同様に吐出したときの吐出量(1液滴量)を100%として、各アルブミン溶液の吐出量(1液滴量)を表した。結果を図6に示す。
(サーマルインクジェット方式の原理に基づいた蛋白質溶液の吐出)
吐出用液体の作製手順を以下に示す。予め適切な濃度の蛋白質を超純水に溶解させ、さらに攪拌しながら1分子中にグアニジン基とカルボキシル基又はスルホン酸基を同時に持つ化合物を添加した。0.1M NaOH又は0.1M HClでpH7.0に調製した後、超純水にて所定のインスリン濃度となるように定容した。
(吐出周波数と吐出量)
実施例1〜7と比較例4の処方について、実施例1と同様の条件で吐出し、吐出周波数を500Hzから1000Hzに変化させて吐出発数を比較した。実施例1〜7に対応する実施例を実施例15〜21とし、比較例4に対応する比較例を比較例15として結果を図8に示す。
(各種蛋白質への効果と添加物の濃度)
続いて、1分子中にグアニジン基とカルボキシル基又はスルホン酸基を同時に持つ化合物として3−グアニジノプロピオン酸、2−グアニジノエタンスルホン酸を選択し、各種蛋白質にある濃度にて添加した。これら吐出用液体を実施例1と同様の吐出実験により評価を行った。吐出周波数は1000Hzとした。120秒間吐出実験を行い、正常に吐出されたものを○、吐出が途中で途切れたものを×として表示した。なお実施例、比較例で検討した処方、及び結果を下記表2に列挙した。
(1分子中にグアニジン基とカルボキシル基又はスルホン酸基を同時に持つ化合物と界面活性剤による相乗効果)
蛋白質及びペプチド溶液に3−グアニジノプロピオン酸を添加し、さらに界面活性剤を加え、吐出用液体を調製した。これらの吐出用液体を実施例22と同様の吐出実験により評価を行った。なお、本実施例で検討した処方、及び結果を下記表3に列挙した。
3μmのノズル径を持つサーマルインクジェット方式による液体吐出ヘッドを用意し、これに接続したタンク内に30%エタノール水溶液を充填した。液体吐出ヘッドに電気的に接続したコントローラにより吐出ヘッドを駆動して液体を吐出口から吐出させ、得られた液滴(噴霧)の粒径及び粒度分布を、レーザー回折式粒度分布測定装置(スプレーテック、マルバーン社製)により測定し、確認した。その結果、約3μmにシャープな粒度分布を持つ液滴として検出された。
(インクジェットプリンターを用いた抗体チップの作製及びセンシング)
図9に本実施例のモデル図を示す。Human IL2モノクローナル抗体、Human IL4モノクローナル抗体及びHuman IL6モノクローナル抗体をそれぞれ0.1〜500μg/mLの濃度でPBSを用いて調製した。ここに2―グアニジノエタンスルホン酸を0.5%(w/w)となるように添加して吐出用液体とした。この液体をインクジェットプリンター(商品名PIXUS950i;キヤノン(株)社製)のヘッドに充填し、Poly−L−Lysinコートスライドガラス上に吐出した。

Claims (4)
- 前記一般式(1)に示す化合物が、2−グアニジノ酢酸、3−グアニジノプロピオン酸、4−グアニジノブタン酸、α−グアニジノグルタミン酸、グアニジノメタンスルホン酸、2−グアニジノエタンスルホン酸、3−グアニジノプロパンスルホン酸、又はこれらの塩からなる群より選択される一種又は二種以上の化合物である請求項1に記載の吐出用液体。
- 界面活性剤を更に含有する請求項1または2に記載の吐出用液体。
- 前記界面活性剤がポリオキシエチレンソルビタン脂肪酸エステルである請求項3に記載の吐出用液体。
