Description
先行技術の針の欠点を考慮して、本発明の目的は、経皮的流体投与に用いられ得る、マイクロマシニングされた構造物およびその作製方法を開発することである。針は、流体抵抗が低く高度の構造強さを有し、組織への薬剤露出の領域が大きく、また、閉塞の危険性が低いことが必要である。これらの事項を考慮し、針の頂部にではなく側方に開口部を有する新しいタイプの面外マイクロ針アレイを提供する。したがって、鋭利なマイクロ針尖端は組織に押し込まれるとき、組織片を打ち抜くのではなく組織を切開する。側方開口部の寸法はプロセスパラメータにより制御されることができる。側方開放型針の薬剤露出の面積は、針の液体チャネルの直径が同じである尖端開放型針と比較して、より大きい。
針の設計および作成
側方開放型マイクロ針は、以前に我々のグループが提示した3部分構成のDRIE(ディープ反応性イオンエッチング)(Deep Reactive Ion Etching)プロセス(上記のグリス(Griss)らによる記事を参照されたい)に基づいている。このプロセスにおいて、DRIEを施された高アスペクト比のシリコン構造物の垂直な壁部が、等方性プラズマエッチング中に、図2a)に示されているように垂直に維持されることが観察された。さらに、二酸化シリコンマスクの下のアンダーエッチングを行い、次いで異方性エッチングを行うと、得られる構造物の断面がマスクに対応することが観察された(図2b)参照)。これらの観察を、裏側からの高アスペクト比の垂直穴と組合せて、側方開放型面外マイクロ針の作製のための、DRIEをベースとしたプロセスを確立した。プロセスの原理を示す簡略図が図2c)に示されている。
流体抵抗の測定は、図4に示されているように、側方開放型マイクロ針アレイを含むチップを、流体チューブへの連結を可能にするキャリア上に組み付けることを必要とする。キャリアは黄銅(真鍮)でできており、慣用のマシニング方法を用いて製造される。3x3mm2のチップをキャリアに紫外線硬化性エポキシ(Epotek OG198)により固定する。チップの正方形の形状およびキャリアの通し穴の円形の形状により、チップコーナーにある幾つかのマイクロ針が閉塞される。21個のマイクロ針は閉塞されずにデバイスを通る流れをもたらす。
Claims (20)
- 支持体から突出するマイクロ針の製造方法であって、前記針が、本体部と、閉じた尖端部と、前記支持体を通って前記突出針に延在する内腔と、を含み、前記本体部が、前記内腔と連通する側方開口部を有し、前記方法が、
エッチング可能な材料から構成され、かつ前側および後側を有するウェハを設けるステップと、
前記ウェハに止り穴をウェハの後側からつくるステップと、
ウェハの前側にマスクを、前記マスクの垂直投影が少なくとも部分的に前記穴の範囲を覆うように設けるステップと、
第1の等方性エッチングを前記マスク下に施してウェハ材料を除去するステップと、
異方性エッチングをウェハに施して突出構造物を形成するステップと、
第2の等方性エッチングを前記突出構造物に選択的に施して止り穴を露出させるステップと、を含み、
穴の位置および寸法に対するマスクの位置および範囲が、前記異方性エッチングまたは前記第2等方性エッチング中に前記側方開口部が形成される位置および範囲である方法。 - 側方開口部は形成せずに針を延長するために最終的な異方性エッチングを行うことを含む請求項1に記載の方法。
- 前記マスクと前記穴がほぼ同じ幾何学的形状を有し、前記マスクが穴の断面よりも大きく、穴の中央点とマスクの中央点は互いに対して位置がずれている請求項1または2に記載の方法。
- 前記マスクが穴の断面よりも大きく、マスクの形状と穴の形状が互いに異なる請求項1または2に記載の方法。
- 前記マスクが穴よりも大きく、穴の中央点とマスクの中央点とが一致する請求項4に記載の方法。
- 前記マスクが少なくとも1つの領域において穴に重なる請求項1または2に記載の方法。
- 前記止り穴がエッチングにより形成される請求項1〜6のいずれか1項に記載の方法。
- 前記止り穴が高いアスペクト比を有する請求項1〜7のいずれか1項に記載の方法。
- マスクの形状および穴の断面が、それぞれ、円形、矩形、三角形または多角形のいずれかである請求項1〜8のいずれか1項に記載の方法。
- 支持部材から突出するマイクロ針であって、前記針が、
針本体部と、
前記支持部材を通って前記突出針へと延在する内腔と、
先端領域において前記内空を閉鎖する閉じた尖端部と、
を含み、前記針本体部が、前記内腔と連通する少なくとも1つの側方開口部を有し、前記本体に少なくとも1つの側方開口部を作るように、針構造物の外側の外囲面が少なくとも一つの決められた領域における針構造物の範囲内で内腔の外囲面と交差し、前記側方開口部が、前記尖端部より下の或る点から前記支持部材より上の或る点まで延在するマイクロ針。 - 前記側方開口部が、前記尖端部より下の或る点から前記支持部材に向って延在する請求項10に記載のマイクロ針。
- 前記マイクロ針の断面が、複数のウィングを有するほぼ十字形状を有し、前記側方開口部が、前記ウィングが互いに連結されている十字形の角部に配置された請求項10に記載のマイクロ針。
- 前記内腔が前記針本体部に対して偏心している請求項10に記載のマイクロ針。
- 前記本体部と前記内腔が互いにほぼ同じ幾何学的形状を有するが、前記内腔の断面が本体部の断面よりも小さい請求項13に記載のマイクロ針。
- 前記幾何学的形状が、円形、矩形、三角形または多角形である請求項14に記載のマイクロ針。
- 前記内腔が前記針本体部に対して同心である請求項10に記載のマイクロ針。
- 前記内腔と前記針本体部とが異なる幾何学的形状を有する請求項16に記載のマイクロ針。
- 支持部材上に設けられた請求項10に記載のマイクロ針アレイと、前記マイクロ針アレイに連結可能な液体貯蔵容器と、液体を、針を通して患者の皮膚を横切って輸送するためのポンプと、を含む経皮的液体輸送装置。
- ポンプが、前記液体を前記液体貯蔵容器から前記マイクロ針アレイに供給するように適合されている請求項18に記載の装置。
