Description

【発明の詳細な説明】 背景技術及び発明の要約 この発明は、自然の老化過程や、太陽放射線への過度
の曝露により促進される同様の過程により影響を及ぼさ
れる皮膚の外観を改善させるための組成物に関するもの
である。特に、本発明は、皮膚の局部的なすじ（皺）を
減少させるために効果的であると認められる局部的な処
方に関するものである。
皮膚は、上層、つまりおよそ20細胞層または約厚さ0.
1mmの表皮と、下層、つまり厚さ約１〜約4mmであり毛細
血管、コラーゲン、弾性素、そして繊維胚を含有する真
皮から成る。真皮は、表皮に対する支持構造及び栄養源
として機能する。老化は、表皮層の細胞質の異成分の増
加により現れるが、その層の厚さにはほとんど影響しな
い。しかしながら、支持真皮は、加齢や太陽への曝露に
伴って薄くなることが知られている。真皮層は、表皮を
支持するとともに表皮への血液の供給機能を有している
ので、皮膚の弾力及び外観を維持するのに決定的に重要
なものである。支持真皮層の崩壊は、表皮の弛みを生
じ、必然的にすじ、つまり皺形成の直接的な原因とな
る。
深い皺は、真皮と表皮の継続的な伸張や萎縮により生
じる。このようにして形成された深い皺は、形成外科ま
たは衰えた箇所下への直接のコラーゲンの注入のみによ
り、除去可能である。しかしながら、あまり伸張されな
い部位の皮膚上の、加齢によって（そして、太陽に何度
も長く曝露されることにより）生ずる細い皺は、支持真
皮層の劣化が直接的に原因するものである。このよう
に、老化過程において、入射放射線により過程が促進さ
れる場合、表皮を結束して支持するコラーゲンの崩壊が
生じる。
コラーゲンは、常態では密集した組織化された形で存
在している。老化過程において、コラーゲンは組織を乱
され、表皮の支持性能が悪化する。さらに、弾性素が真
皮から喪失し、さらに若い皮膚においては多数であり表
面に近くまで達している毛細血管からの血液循環による
支持機能も序々に低下する。入射放射線により促進され
るか否かかわらず、皮膚の老化の結果、真皮層の悪化、
繊維胚の減少、ゴラーゲンの減少、弾性素の減少、循環
による支持機能の低下が生じる。従って、皮膚の常態の
伸張や萎縮によって、たやすく矯正出来ない真皮の損傷
となり皺となる。
皮膚病専門医と美容術専門家により、同様に老化の皺
により損なわれる皮膚の外観を改善させる努力がなされ
てきた。非常によく研究された方法の一つは、表皮細胞
の成長及び増殖を刺激することが知られている薬剤によ
る治療である。新たに増殖した細胞は、さらに多くの組
織を増殖し、多くの水分を保持するので、皮膚は、より
若い外観を呈する。これは、刺激性物質または皮膚を剥
離する化学物質を使用して行われている。表皮の最上層
は、脱皮し新しい表皮細胞と入れ替わる。このような治
療は、何らかの美顔上の改善を提供すると認められる
が、それは主要な原因となる要素、つまり損なわれた支
持真皮層を改善することを意図するものではない。
本発明は、支持真皮層の細胞に照射される太陽による
損傷または老化した皮膚の為の治療に関するものであ
る。我々は、局部的塗布に適した賦形剤に配合されるア
スコルビン酸、チロジン、及び無毒性亜鉛塩、好ましく
は亜鉛硫酸の化合物が、老化／太陽曝露による細かい皺
を示す部位に塗布されると、細かい皺構造をたやすくか
つ目に見えて減少させることを発見した。本発明の組成
物の作用操作は完全に解明されていないが、成分は繊維
束の増殖を刺激し、それらのコラーゲンと弾性素の製造
を促進するように作用し合い、それにより交感真皮組織
の支持の役割を助長している。
本発明は、老化／光による老化した皮膚に対して、現
在最も多く行われている治療、すなわちトレチノイン
（tretinoin）（レティン−A:Retin−Ａ）の治療とは異
なる。トレチノインは、表皮細胞を刺激するが、真皮に
おいてなんら注目すべき変化には至らない［ワイツ（We
iss），ジャマ（JAMA）、p.531（1988）］。トレチノイ
ンは明らかに、普通は若い皮膚と同じように頻繁に脱皮
しない細胞を成長を刺激することにより、表皮の代謝時
間を短縮させる。トレチノインによる表皮細胞の増進さ
れた増殖は、老化による皺を有する皮膚の外観を改善す
るものとして示されているが、表皮に存在する老化に関
連したメラニン細胞の欠乏を補うわけではない。表皮細
胞の増進された増殖は、メラニン細胞の急激な減少を導
き、これが太陽による損傷に対する過敏性と関係してい
ることは既に知等れている。従って、トレチノイン治療
を利用する人は、太陽に不必要にさらすことを避けるよ
う十分に注意しなければならない。
発明の詳細な説明 本発明は、内因性の老化または光による老化に起因し
て表皮に生じる皺を減少する局部的塗布のための組成物
に関するものである。組成物は、約２〜約20％の、さら
に好ましくは約５〜約15％のアスコルビン酸、約１〜約
10％の、さらに好ましくは約３〜約８％のチロジン、そ
して約0.5〜約５％の、さらに好ましくは約１〜約２％
の硫酸亜鉛から成る。これらの組成物は、好ましくは親
水性ローション状、軟膏状、クリーム状またはゲル状の
賦形剤等のような組織と適合性を有する賦形剤に配合さ
れる。このような薬学的に許容される賦形剤は、当業者
において周知であり、局部的塗布に適する形での有効成
分の処方のために、市販されている。このような賦形物
の例としては、市販のデルマベース（Dermabase）とユ
ニベース（Unibase）処方がある。
本発明組成物の必須成分は、無毒性、水溶性亜鉛塩で
あり、好ましくは、硫酸亜鉛である。本発明の処方にお
ける硫酸亜鉛なくしては、我々は有益な効果を見い出せ
なかった。
異常な皮膚状態を引き起している老齢の人々には、亜
鉛が欠乏しているということは認められているが、最近
の研究により、見かけの低血清亜鉛不足は実際には亜鉛
それ自体の欠乏ではなく、亜鉛の担体タンパク質である
血清アルブミンの減少によるということが示唆されてい
る。亜鉛の補給を受けた被験者においては、血漿亜鉛の
増加は検出されるものの、なんの改善も認められなかっ
た。
まず初めに、本発明による局所製剤の調合には３％の
硫酸亜鉛が配合され、45から70才までの５人の女性被験
者により毎日施薬された。一人の被験者は、皮膚刺激に
より研究の間試薬の使用を中止した。その被験者は、後
に１％のみの硫酸亜鉛を含む同種の試薬を使用し続け
た。すべての５人の被験者について、一週間以内で細か
い皺、特に口のまわりの皺、に僅かな改善がみられた。
三カ月使用後、試験に参加し続けた４人の被験者は、全
て素晴らしい結果を報告した。
予備試験の成功によって、二重盲試験が美容術専門家
により行われた。二つの処方が試験された。試験処方製
剤は、表１に示されている割合で成分を混合することに
より、調合された。コントロールクリームは、チロジン
及びアスコルビン酸を除いた同じ成分で調合された。
一人または二人の無関係の美容術専門家が、50人の女
性被験者それぞれの細かい皺の位置または皺のパターン
を記録した。研究においてそれぞれの参加者は、クリー
ムをほんの少し使用し、それを顔全体にではなく皮膚の
皺が寄っている範囲にのみ使用するよう指示された。参
加者は、試験処方製剤は額、口やあごのまわりには使用
可能であるが、目のまわりは避けるべきであることを告
げられた。また、参加者は、試験処方製剤の使用後数分
以上不快なひりひりする感覚を経験しラ場合には、試験
処方製剤の使用前に非常にかるい保湿クリームを使用す
べきであることが支持された。また、被験者は、クリー
ムは朝と夜に使用するべきであることが告げられた。
被験者は、四週間毎日二回使用する二つのクリームサ
ンプルのうち一つを無作為に渡された。被験者及び監督
する美容術専門家により、テストされている皺の状態の
改善が認められた場合、これは記録され、当該被験者は
テストすべき他のクリームサンプルは与えないもとのし
た。しかしながら、与えられた最初のサンプルが何の改
善も示さなかった場合、叉はその使用により何らかの刺
激が起こった場合、第二のサンプルが「ウォッシュアウ
ト（wash−out）」の為の一週間後、参加者に与えられ
た。それぞれのサンプルに対して四週間後、それぞれの
被験者の皺／皺パターンが、試験の初めに記録されたパ
ターンと比較された。50人の被験者は、クリームサンプ
ルの一つ叉は両方を使用し、主観を含む向上が記録され
た後、コード表に示されたテストクリーム及びコントロ
ールクリームは、廃棄された。50人の被験者のうち48人
は、四週間の試験期間の第一または第二週目でテストク
リームによる改善が認められたということが、わかっ
た。一人の被験者は、テストクリームは効果がなかった
と報告しており（彼女の皺は、美容術専門家により非常
に深いと判断を下された）、第二の被験者は、テストサ
ンプルは非常に刺激性があるとし、コントロールクリー
ムの方を好んだ。この研究は、明白に本発明の処方の効
能を確証している。さらに、使用中止後すぐにその効能
が失われるトレレチノインとは違い、、ここに記載され
た組成物の効能は、試験処方製剤の使用中止後少なくと
も二週間持続することが認められた。

Claims (6)

    (57)【特許請求の範囲】
  1. 【請求項１】内因性の老化または光に誘発された老化に
    よる被験者の皮膚の皺の深さを減少させるための美容方
    法であって、薬学的に許容される担体中に２〜20％のア
    スコルビン酸、１〜10％のチロジン、及び0.5〜５％の
    無毒性水溶性亜鉛塩を含有する組成物を、前記皺の部位
    に局部的に塗布するようにしたことを特徴とする内因性
    の老化または光に誘発された老化による被験者の皮膚の
    皺の深さを減少させるための美容方法。
  2. 【請求項２】皺の深さの減少を達成しまたそれを維持す
    るために、前記組成物が毎日一度または二度、局部的に
    適用される請求の範囲第１項に記載の方法。
  3. 【請求項３】薬学的に許容される担体中に２〜20％のア
    スコルビン酸、１〜10％のチロジン、及び0.5〜５％の
    無毒性水溶性亜鉛塩を含有する、老化過程による真皮性
    の皺を減少させるための局部塗布のための組成物。
  4. 【請求項４】亜鉛塩が硫酸亜鉛である請求の範囲第３項
    に記載の組成物。
  5. 【請求項５】１〜３％の硫酸亜鉛を含有する請求の範囲
    第３項に記載の組成物。
  6. 【請求項６】親水性クリームまたは軟膏ベースにおいて
    ５〜15％のアスコルビン酸、５〜10％のチロジン、及び
    １〜２％の硫酸亜鉛からなる請求の範囲第３項に記載の
    組成物。
