Abstract
Description
エチジウム部分は核酸ポリマーに結合し、それによってニッケル−パラジウムナノ粒子を核酸ポリマーに付ける。
1.100mmのTRIS、1.5%のデキストラン硫酸及び0.2%のSDS
2.イソプロピルアルコール(IPA)
3.90%のIPA、1%のドデシルベンゼン硫酸、9%の水
4.90%のIPA、1%のポリアクリル酸ナプタラム、9%の水
レーンは以下のとおり。
レーン1:EDTA(エチレンジアミン四酢酸)を有するTE(トリス(ヒドロキシメチル)アミノメタン)
レーン2:10mMトリス−(ヒドロキシメチル)アミノメタン
レーン3:500mMリン酸ナトリウム
レーン4:50mMリン酸ナトリウム
レーン5:60mMクエン酸ナトリウム
レーン6:3%塩化ナトリウム
を除去する。
Claims (15)
- 使い捨てカートリッジであって、
内側円筒表面を画定し、且つ、シリンジポートを介して分析流体を流動させる動作をなすシリンジ、リリーフポート及び反応チャンバを有するカートリッジ本体と、
前記カートリッジ本体の前記内側円筒表面内に回転可能に接続されて配置された円筒状インサートと、を有し、
前記円筒状インサートは、第一のポートに流動的に接続された破砕チャンバと、第二のポートに流動的に接続された少なくとも1つのチャンバと、前記円筒状インサートの底面に沿って配置された第一及び第二の細長チャネルと、を有し、
前記第一及び第二の細長チャネルの各々は、前記円筒状インサートの前記底面の少なくとも一部分を横断し、
前記第一の細長チャネルは、前記第二の細長チャネルから流動的に隔離され、
前記第一の細長チャネルは、第一の回転位置において前記シリンジを前記反応チャンバに流動的に接続するチャネルポートの第一の対に流動的に接続され、
前記第二の細長チャネルは、第二の回転位置において前記シリンジを前記リリーフポートに流動的に接続するチャネルポートの第二の対に流動的に接続され、
前記円筒状インサートは、前記底面の少なくとも一部分を横断する細長洗浄チャネルを更に有し、
前記細長洗浄チャネルは、第一の幅を有する第一の部分と、前記第一の幅よりも大きい第二の幅を有する第二の部分と、からなり、
前記第二の部分は、洗浄ステップ中に磁性ナノ粒子を濃縮する場所を提供することを特徴とする使い捨てカートリッジ。 - 前記第一のポートは、前記円筒状インサートの垂直側面に配置され且つ前記シリンジポートに整列するように回転され、前記破砕チャンバ及び前記シリンジの間の流体の移動を可能とすることを特徴とする請求項1に記載の使い捨てカートリッジ。
- 前記第二のポートは、前記円筒状インサートの垂直側面に配置され且つ前記シリンジポートに整列するように回転され、前記少なくとも1つのチャンバ及び前記シリンジの間の流体の移動を可能とすることを特徴とする請求項1に記載の使い捨てカートリッジ。
- 前記第一のポート及び前記第二のポートは、前記円筒状インサートの垂直面に沿った所定の高さにあることを特徴とする請求項1に記載の使い捨てカートリッジ。
- 前記リリーフポートは、前記カートリッジ本体の前記内側円筒表面に沿って所定の高さで配置され、
前記リリーフポートの前記所定の高さは、前記第一のポート及び前記第二のポートの各々の所定の高さよりも垂直方向において高いことを特徴とする請求項4に記載の使い捨てカートリッジ。 - カラムチャンバを更に有することを特徴とする請求項1に記載の使い捨てカートリッジ。
- 前記カラムチャンバは脱塩母材を有することを特徴とする請求項6に記載の使い捨てカートリッジ。
- バーコードを更に有することを特徴とする請求項1に記載の使い捨てカートリッジ。
- 生物学的プローブを有するチップを更に有することを特徴とする請求項1に記載の使い捨てカートリッジ。
- 核酸試料を調整するシステムであって、
前記システムは使い捨てカートリッジと検出装置とを有し、
前記使い捨てカートリッジは、内側円筒表面を画定し、且つ、シリンジポートを介して分析流体を流動させる動作をなすシリンジ、リリーフポート及び反応チャンバを有するカートリッジ本体と、前記カートリッジ本体に回転可能に接続された円筒状インサートと、生物学的プローブを有するチップと、を有し、
前記円筒状インサートは、第一のポートに流動的に接続された破砕チャンバと、第二のポートに流動的に接続された少なくとも1つのチャンバと、前記円筒状インサートの底面に沿って配置された第一及び第二の細長チャネルと、を有し、
前記第一及び第二の細長チャネルの各々は、前記円筒状インサートの前記底面の少なくとも一部分を横断し、
前記第一の細長チャネルは、前記第二の細長チャネルから流動的に隔離され、
前記第一の細長チャネルは、第一の回転位置において前記シリンジを前記反応チャンバに流動的に接続するチャネルポートの第一の対に流動的に接続され、
前記第二の細長チャネルは、第二の回転位置において前記シリンジを前記リリーフポートに流動的に接続するチャネルポートの第二の対に流動的に接続され、
前記検出装置は、
前記チップを受容するセンサ載置部と、
前記チップからの電気信号を処理するマイクロプロセッサと、
超音波力を前記破砕チャンバに送る破砕器と、
前記円筒状インサートを回転させるカートリッジ駆動部と、
前記シリンジに接続するプランジャ駆動部と、
前記円筒状インサートの中心から外れて且つ前記円筒状インサートに近接して固定的に搭載されている磁石と、
を有し、
前記カートリッジ駆動部の作動時に、前記円筒状インサートが前記磁石に関して回転するように構成されていることを特徴とするシステム。 - 前記検出装置はバーコードリーダを更に有することを特徴とする請求項10に記載のシステム。
- 前記検出装置と通信するコンピュータネットワークを更に有することを特徴とする請求項10に記載のシステム。
- 前記検出装置は全地球測位システムを更に有することを特徴とする請求項10に記載のシステム。
- 前記検出装置は、各々独立して制御される第一のヒータ及び第二のヒータを有し、
前記第一のヒータ及び第二のヒータは、前記カートリッジ駆動部の作動時に、前記円筒状インサートが前記第一のヒータの作動時に前記第二のヒータに関して回転するように、前記円筒状インサートに近接して固定的に搭載されていることを特徴とする請求項10に記載のシステム。 - 前記検出装置は携帯可能な検出装置であることを特徴とする請求項10に記載のシステム。
