Abstract
Description
本発明のさらなる目的は、身体外で容易に分解するポリマーを提供することである。
のポリ[ポリ[α,ω−ジヒドロキシ−[オリゴ(3−(R)−ヒドロキシブチレート)−stat−グリコリド)−エチレン−オリゴ−(3−(R)−ヒドロキシブチレート−stat−グリコリド)]alt−2,2,4−トリメチルヘキサメチレン−1,6−ジイソシアネート]−co−ポリ[ジヒドロキシ[オリゴ−グリコリド−ran−ε−カプロラクトン)−エチレン−(オリゴ−グリコリド−ran−ε−カプロラクトン)]alt−2,2,4−トリメチレンヘキサエチレン−1,6−イソシアネート]である。さらに好ましいポリマーは、ポリマーのグリコリド単位が対応するラクチド又はカプロラクトンで置換されることを除いて、上記と同じポリマーである。
該薬理学的物質は、ジオール又はブロックコポリマーへの吸着又は共有化学結合のどちらかによって保持されている。同様に、該移植片材料は、血管又は他の生物学的管状構造体(食道、胆道、尿道)用のステント(硬質、拡張性、自己拡張性)を製造するために好適である。
・同様に、本発明のジオール又はブロックコポリマーを用いて製造できるシート様構造体(外傷カバー、膜型人工肺、代用角膜ベース等)。
・外科的縫合材料として、そして編物、ブレ−ド又はニット構造に加工するためのスレッド様構造体。
・微小血管を結紮し、そして閉鎖に対して熱可塑性を利用するステープラー又はクランプ用のクリップ様又はクランプ様構造体。
・局所的な外傷の治療に用いる単一の又は複合のin vitro生物組織(in vivo再生医療)を製造するためのマトリックスとしてのゼラチン状又は多孔質の構造体に対する固体。
・代用肌、脂肪組織、腱、軟骨及び骨、神経等用の事前調整されたプレースホルダー。
・物理的又は生物学的な装填特性と、物理的構造(発泡体(schaume)、ゲル、ミクロスフェア及びナノスフェア)と、表面構造とのために、体内の解剖学的構造又は肌を経由して、治療物質(ホルモン、薬剤)又は美容物質(リポソーム、たんぱく質、ビタミン)を供給できるポリマー構造体。
・静脈節瘤、脚部の静脈瘤(食道静脈瘤)又は胃腸源の出血(内視的に又は血管を通して)を硬化させるための本発明の材料の組成物。
・生物活性物質を伴う好適な装填及び形状において、閉塞(卵管、精管)によって可逆的又は不可逆的なの避妊を可能とする造形品。
・人工耳小骨及び人工心臓弁、人工大動脈及び人工循環器系血管。
下記例を用いて、さらに本発明を具体的に説明する。
ポリ[(R)−3−ヒドロキシブチレート]をエチレングリコールとエステル交換させることによるα,ω−ジヒドロキシ[オリゴ(3−(R)−ヒドロキシブチレート)−エチレン−オリゴ(3−(R)−ヒドロキシブチレート)]の調製
該沈殿物をろ過し、乾燥させた。
収率=86%のオリゴマー(Mn=2450)
α,ω−ジヒドロキシ[オリゴ−3−(R)−ヒドロキシブチレート−stat−グリコリド)−エチレン−オリゴ−(3−(R)−ヒドロキシブチレート−stat−グリコリド)]の合成
グリコレート単位に対するエステル交換中で用いられる3(R)−ヒドロキシブチレート単位の比の値が約3より低くなると、わずかな濁りが該反応生成物内の該エステル交換の終端方向に進み、そして該濁りの理由は、不溶性のオリゴグリコリド生成物にありうる。該ポリマーを、これらの部分、触媒(DBTL)から、そしてジグリコリドから、次の手段で精製させることができる。
6時間、18℃に冷却させた冷却ジャケットを有するソックスレー内で、メタノールを用いて25gの粗ポリマーを抽出し、次いで、減圧下で乾燥させた。次いで、無水メチレンクロリドを用いて、同じ冷却したソックスレー中で該ポリマーを抽出し、そして5倍量の無水メタノールを用いて沈殿させ、そして減圧下で乾燥させた。
収率:86%の粗ポリマー。
ポリ[ポリ[α,ω−ジヒドロキシ[オリゴ−3−(R)−ヒドロキシブチレート−stat−グリコリド)−エチレン−オリゴ−(3−(R)−ヒドロキシブチレート−stat−グリコリド)]−alt−2,2,4−トリメチルヘキサメチレン1,6−ジイソシアネート]−co−ポリ[α,ω−ジヒドロキシ−[オリゴ−グリコリド−ran−ε−カプロラクトン)−エチレン−(オリゴ−グリコリド−ran−ε−カプロラクトン)]−alt−2,2,4−トリメチルヘキサメチレン1,6−ジイソシアネート]の調製
オイル加熱式でジャケットをかぶせた1000mlの反応器(温度センサー、保護ガスとしての窒素用キャピラリ、及び均圧を有する滴下漏斗上の還流冷却器を備える)内で重合させた。該滴下漏斗にA4のモレキュラーシーブスを詰めた。該反応器を、400mlの1,2−ジクロロエタンと、31.3gのジヒドロキシ[オリゴ−3−(R)−ヒドロキシブチレート−stat−グリコリド)−エチレン−オリゴ−(3−(R)−ヒドロキシブチレート−stat−グリコリド)](Mn=2440、E7由来の生成物)とで充填し、そして該溶媒が冷却器内に達し、モレキュラーシーブス上を還流するまで加熱した。該溶媒が20ppm未満になるように乾燥するまで、還流を続けた。
参照ポリマーであるポリ[ポリ[α,ω−ジヒドロキシ−[オリゴ−3−(R)−ヒドロキシブチレート)−エチレン−オリゴ−(3−(R)−ヒドロキシブチレート]−alt−2,2,4−トリメチルヘキサメチレン1,6ジイソシアネート]−co−ポリ[α,ω−ジヒドロキシ[オリゴ−グリコリド−ran−ε−カプロラクトン)−エチレン−(オリゴ−グリコリド−ran−ε−カプロラクトン)]−alt−2,2,4−トリメチルヘキサメチレン1,6−ジイソシアネート]と比較したポリ[ポリ[α,ω−ジヒドロキシ−[オリゴ−3−(R)−ヒドロキシブチレート−stat−グリコリド)−エチレン−オリゴ−(3−(R)−ヒドロキシブチレート−stat−グリコリド)]−alt−2,2,4−トリメチルヘキサメチレン1,6−ジイソシアネート]−co−ポリ[α,ω−ジヒドロキシ[オリゴ−グリコリド−ran−ε−カプロラクトン)−エチレン−(オリゴ−グリコリド−ran−ε−カプロラクトン)]−alt−2,2,4−トリメチルヘキサメチレン1,6−ジイソシアネート]の加水分解
グリコリド/ε−カプロラクトン=1/1モル;例1由来のPHB/グリコリドジオール
分解速度に対するグリコリド変性PHBジオールの影響を、未変性PHBジオールを有する構造的に類似であるポリマーに関して測定した。フィルム及び連続気泡発泡体(孔サイズ、約50〜300μm)にあらかじめ加工したポリマー試料上と、粉末状の粗ポリマー上で、分解例を実施した。

- ジイソシアネート、二塩基酸ハロゲン化物又はホスゲンの存在下で、ジオールを、該同一のジオール、α,ω−ジヒドロキシポリエステル又はα,ω−ジヒドロキシポリエーテルの群から選択される成分と線状重縮合させることによって得られる、少なくとも2種の化学的に異なるブロック単位を有する生体適合性ブロックコポリマーであって、
該ジオールが、α,ω−ジヒドロキシ−[オリゴ(3−(R)−ヒドロキシブチレート)−エチレン−オリゴ−3−(R)−ヒドロキシブチレート)を、ジグリコリド及び/若しくはジラクチド及び/若しくはカプロラクトン又はそれらの混合物とエステル交換させることによって得られ、
該α,ω−ジヒドロキシポリエステルが、ポリ−(R)−ヒドロキシ吉草酸又はそれらのコポリマーを、エチレングリコールを有する3−ヒドロキシ吉草酸とエステル交換させることによって得られ、
該α,ω−ジヒドロキシポリエーテルが、α,ω−ジヒドロキシポリ(オキシテトラメチレン)、α,ω−ジヒドロキシポリ(オキシエチレン)及びエチレングリコールとプロピレングリコールとのコポリマーの群から選択される、

- 触媒の存在下で該反応を実施する、請求項16に記載の方法。
- 精製のため、該ジオールをメチレンクロリド中に溶解させ、そして不純物を取り除くことを特徴とする、請求項16又は17に記載の方法。
