Abstract
【解決手段】酸性成分としてピロリドンカルボン酸、塩基性成分としてL−アルギニン、L−リジン、L−ヒスチジンの塩基性アミノ酸、トリスヒドロキシメチルアミノメタン、BIS-Trisの中から選択される少なくとも1種類からなる緩衝剤と、浸透圧調整剤として糖アルコール、およびビグアニド系消毒剤を含有することを特徴とするコンタクトレンズ用溶液である。

1.消毒剤としての有効性試験
2.安全性試験(細胞毒性試験)
3.レンズ適合性試験(イオン性高含水レンズのサイズ変化測定)
試験菌株として、緑膿菌(Pseudomonas aeruginosa:ATCC 9027)、黄色ブドウ球菌 (Staphylococcus aureus:ATCC6538)、霊菌(Serratia marcescens:ATCC13880)、の細菌3種類、および カンジダ(Candida albicans:ATCC 10231)、フザリウム(Fusarium solani:ATCC36031)の真菌2種類を用いた。細菌3種類については、ソイビーンカゼイン寒天培地の斜面培地上で35℃×24時間培養した菌原体に、滅菌済みダルベッコリン酸緩衝生理食塩水を加えて回収した懸濁液(108〜109cfu/mL)を試験菌原液とした。また真菌2種類については、ブドウ糖ペプトン寒天培地の斜面培地上で35℃×24時間培養した菌原体に、滅菌済みダルベッコリン酸緩衝生理食塩水を加えて回収した懸濁液(108〜109cfu/mL)を試験菌原液とした。
初発菌数(対数値)−接種後試験液の生菌数(対数値)
毒時間における対数差が、細菌3種類については対数差3.0以上、真菌2種類については対数差1.0以上であることとした。なお、消毒時間は4時間にて試験を実施した。
本発明におけるコンタクトレンズ用溶液の安全性を評価するために「医療用具及び医用材料の基礎的な生物学的試験のガイドライン1995解説」における第3章「細胞毒性試験」を参考とし、コロニー法を用いて評価を行った。
(試験液の各添加濃度におけるコロニー数/コントロールのコロニー数)×100
試験レンズ1枚を生理食塩水(ISO10344(Optics and optical instruments-Contact lenses-Saline solution for contact lens testing)4mLの入ったレンズケースに浸漬して、25℃±1℃の恒温水槽にて24時間以上放置した。レンズを取り出し、初期レンズサイズとして、ISO生理食塩水中のレンズサイズ(直径)を測定した。レンズサイズの測定は読み取り顕微鏡を用いて、レンズを200倍に拡大し、25℃±1℃の生理食塩水20mL中のレンズの直径を測定した。レンズ1枚当たり3回測定し、その平均値を実測値とした。次に同一の試験レンズを25℃±1℃のコンタクトレンズ用溶液20mL中に入れ、5分浸漬後後、10分浸漬後のレンズサイズをレンズ1枚当たり3回測定し、その平均値を実測値とした。測定後のレンズをコンタクトレンズ用溶液4mLの入ったレンズケースに浸漬して、25℃±1℃の恒温水槽にて14日間浸漬保存した。途中、1日浸漬後、7日浸漬後、そして14日浸漬後のレンズサイズを、上記と同様にして、コンタクトレンズ用溶液中において測定した。なお、試験用レンズとしては、ジョンソン・エンド・ジョンソン社製商品名 アキュビュー2(イオン性高含水レンズ:グループ4、表示値(規格):ベースカーブ8.7mm、頂点屈折率−3.00D、直径14.0mm、含水率58%)を用いた。
Claims (2)
- 酸性成分としてピロリドンカルボン酸、塩基性成分としてL−アルギニン、L−リジン、L−ヒスチジンの塩基性アミノ酸、トリスヒドロキシメチルアミノメタン、BIS-Trisの中から選択される少なくとも1種類からなる緩衝剤と、浸透圧調整剤として糖アルコール、およびビグアニド系消毒剤を含有することを特徴とするコンタクトレンズ用溶液。
- 糖アルコールがマンニトール、キシリトール、グルシトール(ソルビトール)、エリトリトール、トレイトール、ガラクチトール、ラクチトール、マルチトール、アラビトール、リビトール、イジトール、ボレミトールから選択される少なくとも1種類を含有することを特徴とする請求項1記載のコンタクトレンズ用溶液。
