Abstract
【解決手段】 反応混合物を、第一粒子よりも比重が大きく、第二粒子よりも比重が小さく、かつ第一粒子及び第二粒子からなる粒子複合体よりも比重が小さい液体、又は第一粒子よりも比重が小さく、第二粒子よりも比重が大きく、かつ第一粒子及び第二粒子からなる粒子複合体よりも比重が大きい液体と混合した後、沈降物及び浮上物が形成されるまで静置する。

Description
本態様によれば、磁性粒子を含む分子が反応溶媒中を様々な方向に移動するので、磁性粒子を含む分子と他の分子との遭遇確率がより一層向上し、磁性粒子を含む分子と他の分子との反応をより一層促進させることができる。
容器の外壁面の所定領域を通じた磁石の作用及びその解除を繰り返す回数は特に限定されるものではないが、第一プローブと第二プローブと標的物質との結合体が十分量形成される回数であることが好ましい。
容器は透明又は半透明であることが好ましい。これにより、容器内から発せられる光(例えば、蛍光、化学発光等)を容器外で検出することができる。透明又は半透明の材料としては、プラスチック、ガラス等が挙げられる。
図1は、本実施形態で使用される分注機の概略図であり、図2〜7は本実施形態の各ステップを表す概略断面図である。
磁性粒子MBの質量及び体積はオリゴヌクレオチドN1の質量及び体積よりも遥かに大きく、標識化非磁性粒子LBの質量及び体積はオリゴヌクレオチドN2の質量及び体積よりも遥かに大きく、磁性粒子MB及び標識化非磁性粒子LBの質量及び体積は標的核酸Tの質量及び体積よりも遥かに大きいので、プローブP1の比重は磁性粒子MBの比重に、プローブP2の比重は標識化非磁性粒子LBの比重に、ハイブリダイゼーション体C1の比重は磁性粒子LBの比重に、ハイブリダイゼーション体C2の比重は磁性粒子MB及び標識化非磁性粒子LBからなる粒子複合体の比重に近似することができる。したがって、液体L7を静置すると、未反応のプローブP1、ハイブリダイゼーション体C1及びハイブリダイゼーション体C2は沈降する一方、未反応のプローブP2は浮上する。このとき、ハイブリダイゼーション体C2には重力及び浮力以外の力は加わらないので、ハイブリダイゼーション体C2に破壊、分解等は生じない。このようにして、ハイブリダイゼーションC2に破壊、分解等が生じない穏やかな条件下、ハイブリダイゼーションC2は沈降物として分離される。
例えば、図8(a)に示すように、円筒状部材4の外壁面近傍に複数個の磁石3a〜3hを配置し、容器2dを回転させながら円筒状部材4の内壁面に沿って移動させることにより、容器2dの外壁面の所定領域の位置を容器2dの周方向に変化させながら、所定領域を通じて容器2d内の磁性粒子MBに各磁石を作用させることができる。この際、図8(b)に示すように、容器2dの外壁面に複数の凸部21を設けるとともに、円筒状部材4の内壁面に複数の凹部41を設け、凸部21と凹部41を順次嵌合させることにより、容器2dを回転させながら円筒状部材4の内壁面に沿って移動させることができる。なお、図8(a)及び(b)では、円筒状部材4の外壁面近傍に8個の磁石を配置したが、磁石の個数及び配置位置は適宜変更可能である。
本実施形態では、容器2dとして有底筒状容器を使用したが、ピペットチップ10のような無底筒状容器を使用してもよい。無底筒状容器内には、液体を吸引した状態で保持することができる。

Claims (14)
- 第一粒子及び前記第一粒子に固定された第一結合性物質を有する第一プローブと、前記第一粒子と比重が異なる第二粒子及び前記第二粒子に固定された第二結合性物質を有する第二プローブと、前記第一結合性物質が結合し得る第一部分及び前記第二結合性物質が結合し得る第二部分を有する標的物質とを反応させて得られる反応混合物から、前記第一プローブと前記第二プローブと前記標的物質との結合体を分離する方法であって、
前記反応混合物を、前記第一粒子よりも比重が大きく、前記第二粒子よりも比重が小さく、かつ前記第一粒子及び前記第二粒子からなる粒子複合体よりも比重が小さい液体、又は前記第一粒子よりも比重が小さく、前記第二粒子よりも比重が大きく、かつ前記第一粒子及び前記第二粒子からなる粒子複合体よりも比重が大きい液体と混合した後、沈降物及び浮上物が形成されるまで静置する工程を含む前記方法。 - 前記第一粒子及び/又は第二粒子が磁性粒子であり、
前記反応混合物が、前記第一プローブと前記第二プローブと前記標的物質とを容器内に収容された反応溶媒中で反応させた後、前記容器の外壁面近傍に磁石を配置した状態で前記反応溶媒を除去して得られたものである請求項1記載の方法。 - 前記第一粒子が非磁性粒子であり、前記第二粒子が磁性粒子である請求項2記載の方法。
- 第一粒子及び前記第一粒子に固定された第一結合性物質を有する第一プローブと、第二粒子及び前記第二粒子に固定された第二結合性物質を有する第二プローブと、前記第一結合性物質が結合し得る第一部分及び前記第二結合性物質が結合し得る第二部分を有する標的物質とを反応させる方法であって、
前記第一粒子及び/又は前記第二粒子が磁性粒子であり、
前記第一プローブ、前記第二プローブ、前記標的物質及び反応溶媒を容器に収容する第一工程、
前記容器の外壁面の所定領域を通じて前記容器内の前記磁性粒子に磁石を作用させる第二工程、
前記磁石の作用を解除する第三工程、及び
前記所定領域の位置を変化させながら前記第二工程及び前記第三工程を繰り返す第四工程を含む前記方法。 - 前記第二工程において、前記所定領域と前記磁石とを近接させることにより前記磁石を作用させ、
前記第三工程において、前記所定領域と前記磁石とを離反させることにより前記磁石の作用を解除し、
前記第四工程において、前記所定領域の位置を変化させながら前記所定領域と磁石との近接及び離反を繰り返す請求項4記載の方法。 - 前記所定領域の位置を前記容器の周方向又は長手方向に変化させる請求項4又は5記載の方法。
- 第一粒子及び前記第一粒子に固定された第一結合性物質を有する第一プローブと、第二粒子及び前記第二粒子に固定された第二結合性物質を有する第二プローブとを用いて、前記第一結合性物質が結合し得る第一部分及び前記第二結合性物質が結合し得る第二部分を有する標的物質を検出する方法であって、
前記第一粒子として標識化粒子を選択し、前記第二粒子として前記第一粒子よりも比重が大きい粒子を選択する第一工程、
前記第一プローブと前記第二プローブと前記標的物質とを反応させ、反応混合物を得る第二工程、
前記反応混合物を、前記第一粒子よりも比重が大きく、前記第二粒子よりも比重が小さく、かつ前記第一粒子及び前記第二粒子からなる粒子複合体よりも比重が小さい液体と混合した後、沈降物及び浮上物が形成されるまで静置する第三工程、及び
前記沈降物に含まれる前記第一プローブと前記第二プローブと前記標的物質との結合体を、前記第一粒子の標識に基づき検出する第四工程を含む前記方法。 - 第一粒子及び前記第一粒子に固定された第一結合性物質を有する第一プローブと、第二粒子及び前記第二粒子に固定された第二結合性物質を有する第二プローブとを用いて、前記第一結合性物質が結合し得る第一部分及び前記第二結合性物質が結合し得る第二部分を有する標的物質を検出する方法であって、
前記第一粒子として標識化粒子を選択し、前記第二粒子として前記第一粒子よりも比重が小さい粒子を選択する第一工程、
前記第一プローブと前記第二プローブと前記標的物質とを反応させ、反応混合物を得る第二工程、
前記反応混合物を、前記第一粒子よりも比重が小さく、前記第二粒子よりも比重が大きく、かつ前記第一粒子及び前記第二粒子からなる粒子複合体よりも比重が大きい液体と混合した後、沈降物及び浮上物が形成されるまで静置する第三工程、及び
前記浮上物に含まれる前記第一プローブと前記第二プローブと前記標的物質との結合体を、前記第一粒子の標識に基づき検出する第四工程を含む前記方法。 - 前記第一工程において、前記第一粒子及び/又は前記第二粒子として磁性粒子を選択し、
前記第二工程において、前記第一プローブと前記第二プローブと前記標的物質とを容器内に収容された反応溶媒中で反応させた後、前記容器の外壁面近傍に磁石を配置した状態で前記反応溶媒を除去し、前記反応混合物を得る請求項7又は8記載の方法。 - 前記第一工程において、前記第一粒子として非磁性粒子を選択し、前記第二粒子として磁性粒子を選択する請求項9記載の方法。
- 前記第一工程において、前記第一粒子及び/又は前記第二粒子として磁性粒子を選択し、
前記第二工程において、請求項4〜6のいずれかに記載の方法により前記第一プローブと前記第二プローブと前記標的物質とを反応させる請求項7又は8記載の方法。 - 前記第二工程において、前記第一プローブと前記第二プローブと前記標的物質とを反応させた後、前記容器の外壁面近傍に磁石を配置した状態で前記反応溶媒を除去し、前記反応混合物を得る請求項11記載の方法。
- 前記第一工程において、前記第一粒子として非磁性粒子を選択し、前記第二粒子として磁性粒子を選択する請求項12記載の方法。
- 前記第一工程において、前記第二粒子として磁性粒子を選択し、
前記第二工程において、前記標的物質を含有する試料と前記第二プローブとを混合し、磁力制御により前記第二プローブと前記標的物質との結合体を回収し、前記第二プローブと前記標的物質との結合体を前記第一プローブと反応させる請求項7〜13のいずれかに記載の方法。
