- 【特許請求の範囲】 1. 共有結合された多座リガンドを含むヌクレオシドであって、前記リガンドが 前記ヌクレオシドの2'位又は3'位で結合したことを特徴とするヌクレオシド。 2. 前記結合が2'位である請求の範囲第1項に記載のヌクレオシド。 3. 前記結合が3'位である請求の範囲第1項に記載のヌクレオシド。 4. 共有結合された多座リガンドを含むホスホルアミジトヌクレオシドであって 、前記リガンドが前記ヌクレオシドの2'位で結合したことを特徴とするホスホル アミジトヌクレオシド。 5. 共有結合された多座リガンドを含む第一ヌクレオシドを含む組成物であって 、前記リガンドが前記ヌクレオシドの2'位で結合し、前記ヌクレオシドが調節細 孔ガラス(CPG)に共有結合したことを特徴とする組成物。 6. 調節細孔ガラス(CPG)に共有結合されたオリゴヌクレオチドを含む組成物で あって、前記オリゴヌクレオチドの少なくとも一つのヌクレオシドが多座修飾ヌ クレオシドであることを特徴とする組成物。 7. 前記多座修飾ヌクレオシドが前記オリゴヌクレオチドの5'末端にある請求の 範囲第6項に記載の組成物。 8. 前記多座修飾ヌクレオシドが前記オリゴヌクレオチドの内部ヌクレオシドに ある請求の範囲第6項に記載の組成物。 9. 前記オリゴヌクレオチドに共有結合された第二2’−アミノ修飾ヌクレオチ ドを更に含む請求の範囲第6項に記載の組成物。 10.前記多座ヌクレオシドにキレート化された遷移金属を更に含む請求の範囲第 1項、第4項、第5項、又は第6項に記載の組成物。 11.少なくとも一つの電子ドナー部分及び少なくとも一つの電子アクセプター部 分を含む一本鎖核酸であって、前記電子ドナー部分及び前記電子アクセプター部 分が前記核酸に共有結合され、かつ前記電子ドナー部分及び前記電子アクセプタ ー部分の少なくとも一つが多座ヌクレオシドに結合されることを特徴とする一本 鎖核酸。 12.前記電子ドナー部分及び電子アクセプター部分の両方が多座ヌクレオシドに より結合される請求の範囲第11項に記載の核酸。 13.前記電子ドナー部分及び電子アクセプター部分の他方が末端塩基により結合 される請求の範囲第11項に記載の核酸。 14.前記電子ドナー部分及び電子アクセプター部分の他方がリボースホスフェー ト主鎖のリボースの2'位又は3'位により結合される請求の範囲第11項に記載の核 酸。 15.前記電子ドナー部分及び前記電子アクセプター部分の少なくとも一つが遷移 金属錯体である請求の範囲第11項、第12項、第13項、又は第14項に記載の核酸。 16.前記電子ドナー部分及び前記電子アクセプター部分の少なくとも一つが電極 である請求の範囲第11項、第13項、第14項、又は第15項に記載の組成物。 17.前記電子ドナー部分及び前記電子アクセプター部分の少なくとも一つが有機 電子ドナー又はアクセプターである請求の範囲第11項、第12項、第13項、第14項 、第15項、又は第16項に記載の組成物。 18.少なくとも一つの共有結合された電子ドナー部分を含む第一一本鎖核酸と少 なくとも一つの共有結合された電子アクセプター部分部分を含む第二一本鎖核酸 とを含む組成物であって、前記電子ドナー部分及び電子アクセプター部分の少な くとも一つが多座ヌクレオシドに結合されることを特徴とする組成物。 19.核酸サンプル中の標的配列の検出方法であって、 a)少なくとも一つの共有結合された電子ドナー部分及び少なくとも一つの共有 結合された電子アクセプター部分を含む一本鎖核酸を前記標的配列にハイブリッ ド形成してハイブリダイゼーション錯体を形成し(前記電子ドナー部分及び電子 アクセプター部分の少なくとも一つが多座ヌクレオシドに結合される)、そして b)前記標的配列の存在又は不在のインジケーターとしてハイブリダイゼーショ ン錯体中の前記電子ドナー部分及び電子アクセプター部分の間の電子伝達を検出 することを特徴とする検出方法。 20.核酸中の標的配列の検出方法(前記標的配列は第一標的ドメインと前記第一 標的ドメインに隣接する第二標的ドメインとを含む)であって、前記方法が a) 少なくとも一つの共有結合された電子ドナー部分を含む第一核酸を前記第一標的 ドメインにハイブリッド形成し、 b)少なくとも一つの共有結合された電子アクセプター部分を含む第二核酸を前 記第二標的ドメインにハイブリッド形成し(前記電子ドナー部分及び電子アクセ プター部分の少なくとも一つが多座リボヌクレオシドに結合される)、そして c)前記第一核酸及び第二核酸が前記核酸サンプル中の前記標的配列の存在又は不 在のインジケーターとして前記第一標的ドメイン及び第二標的ドメインにハイブ リッド形成される間に、前記電子ドナー部分と電子アクセプター部分の間の電子 伝達を検出することを特徴とする検出方法。 21.前記電子ドナー部分及び電子アクセプター部分の両方が多座ヌクレオシドに 結合される請求の範囲第19項又は第20項に記載の方法。 22.前記電子ドナー部分及び電子アクセプター部分の他方が末端塩基により結合 される請求の範囲第19項又は第20項に記載の方法。 23.前記電子ドナー部分及び電子アクセプター部分の他方がリボースホスフェー ト主鎖のリボースの2'位又は3'位で結合される請求の範囲第19項又は第20項に記 載の方法。 24.前記電子ドナー部分及び前記電子アクセプター部分の少なくとも一つが遷移 金属錯体である請求の範囲第19項及び第20項に記載の方法。 25.前記電子ドナー部分及び前記電子アクセプター部分の少なくとも一つが電極 である請求の範囲第19項及び第20項に記載の方法。 26.前記電子ドナー部分及び前記電子アクセプター部分の少なくとも一つが有機 電子ドナー又はアクセプターである請求の範囲第19項及び第20項に記載の方法。 27.核酸の3'末端で結合された電子伝達部分を有する前記核酸の生産方法であっ て、前記方法が a)多座ヌクレオシドを調節細孔ガラスに結合し、そして b)ホスホルアミジトヌクレオシドを前記修飾ヌクレオシドの5'末端に付加して 核酸を形成することを特徴とする核酸の生産方法。 28.核酸の5'末端で結合された電子伝達部分を有する前記核酸の生産方法であっ て、前記方法が a)ヌクレオシドを調節細孔ガラスに結合し、 b)ホスホルアミジトヌクレオシドを前記修飾ヌクレオシドの5'末端に付加して 核酸を形成し、そして c)多座ヌクレオシドを前記核酸の5'末端に付加することを特徴とする核酸の生 産方法。 29.内部ヌクレオシドで結合された電子伝達部分を有する核酸の生産方法であっ て、前記方法が a)ヌクレオシドを調節細孔ガラスに結合し、 b)必要によりホスホルアミジトヌクレオシドを前記ヌクレオシドの5'末端に付 加して核酸を形成し、 c)多座ヌクレオシドを前記核酸の5'末端に付加し、そして d)ホスホルアミジトヌクレオシドを前記多座ヌクレオシドの5'末端に付加する ことを特徴とする核酸の生産方法。 30.核酸の3'末端及び5'末端で結合された電子伝達部分を有する前記核酸の生産 方法であって、前記方法が a)第一多座ヌクレオシドを調節細孔ガラスに結合し、 b)ホスホルアミジトヌクレオシドを前記多座ヌクレオシドの5'末端に付加して 核酸を形成し、そして c)第二多座ヌクレオシドを前記核酸の5'末端に付加することを特徴とする核酸 の生産方法。 31.前記多座ヌクレオシドが結合された遷移金属を更に含む請求の範囲第27項、 第28項、第29項、又は第30項に記載の方法。 32.前記核酸を前記担体から開裂する工程を更に含む請求の範囲第27項、第28項 、第29項、又は第30項に記載の方法。
