Claims (1)
- 【特許請求の範囲】 1.植物細胞において発現可能なキメラ遺伝子であって: 植物細胞において構成的発現を与える能力により特徴付けられ、異種DNAコー ド配列に作動可能に連結された、配列番号2またはその連続するフラグメントと 少なくとも約80%の配列同一性を有するキイチゴdru1プロモーターを含む、キメ ラ遺伝子。 2.前記異種DNA配列が、ポリペプチドを発現する形質転換植物細胞に抗生物質 耐性を与える該ポリペプチドをコードする、請求項1に記載のキメラ遺伝子。 3.前記ポリペプチドが、ネオマイシンホスホトランスフェラーゼ、ハイグロマ イシンホスホトランスフェラーゼおよびブロモキシニル特異的ニトリラーゼから なる群から選択される、請求項2に記載のキメラ遺伝子。 4.前記異種DNA配列が、ポリペプチドを発現する形質転換植物細胞に除草剤耐 性を与える該ポリペプチドをコードする、請求項1に記載のキメラ遺伝子。 5.前記異種DNA配列がnptII遺伝子を含む、請求項1に記載のキメラ遺伝子。 6.前記プロモーターが、配列番号3と少なくとも約80%の配列同一性を有する 、請求項1〜5のいずれかに記載のキメラ遺伝子。 7.前記プロモーターが、配列番号4と少なくとも約80%の配列同一性を有する 、請求項1〜5のいずれかに記載のキメラ遺伝子。 8.前記プロモーターが配列番号3を含む、請求項1〜5のいずれかに記載のキ メラ遺伝子。 9.前記プロモーターが配列番号4を含む、請求項1〜5のいずれかに記載のキ メラ遺伝子。 10.請求項1〜9のいずれかに記載のキメラ遺伝子を含む植物細胞。 11.請求項1〜9のいずれかに記載のキメラ遺伝子を含む植物形質転換ベクタ ー。 12.請求項1〜9のいずれかに記載のキメラ遺伝子を含むトランスジェニック 植物。 13.植物細胞において構成的発現を与える能力により特徴付けられた、配列番 号2またはその連続するフラグメントと少なくとも約80%の配列同一性を有する 、キイチゴdru1プロモーター。 14.配列番号3と少なくとも約80%の配列同一性を有する、請求項13に記載 のプロモーター。 15.配列番号4と少なくとも約80%の配列同一性を有する、請求項13に記載 のプロモーター。 16.配列番号3を含む、請求項13に記載のプロモーター。 17.配列番号4を含む、請求項13に記載のプロモーター。 18.請求項13〜17のいずれかに記載のプロモーターを含むDNA構築物。 19.トランスジェニック植物を産生する方法であって、以下の工程を包含する 、方法: 請求項1〜9のいずれかに記載のキメラ遺伝子で植物の前駆細胞を形質転換し て、形質転換前駆細胞を生じさせる工程;および 該形質転換前駆細胞を成長させて、トランスジェニック植物を産生する工程。 20.トランスジェニック植物において選択可能なマーカー遺伝子の発現を提供 するための方法であって、以下の工程を包含する、方法: (a)請求項1に記載のキメラ遺伝子で植物の前駆細胞を形質転換して形質転 換前駆細胞を生じさせる工程であって、ここで前記DNA配列が、その発現が形質 転換植物細胞に抗生物質選択薬剤の存在下で増殖する能力を与える産物をコード する選択可能なマーカー遺伝子を含む、工程、 (b)該選択薬剤の存在下で増殖する能力により、該キメラ遺伝子を含む形質 転換植物細胞を選択する工程、 (c)(b)において選択された形質転換植物細胞を再生して、分化した植物を 提供する工程、および (d)該産物を発現する形質転換植物を選択する工程。 21.前記形質転換工程が、Agrobacteriumに基づく形質転換、エレクトロポレ ーション、マイクロインジェクション、およびマイクロプロジェクタイルボンバ ードメントからなる群から選択される形質転換方法により実施される、請求項2 0に記載の方法。 22.前記選択薬剤がハイグロマイシン、ゲネチシン、およびカナマイシンから なる群から選択される、請求項20または請求項21に記載の方法。 23.前記選択可能なマーカー遺伝子が、ネオマイシンホスホトランスフェラー ゼ(npt)遺伝子、ハイグロマイシンホスホトランスフェラーゼ(hpt)遺伝子、 およびブロモキシニル特異的ニトリラーゼ(bxn)遺伝子からなる群から選択さ れる、請求項20〜22のいずれかに記載の方法。 24.前記形質転換工程における前記キメラ遺伝子が、請求項14〜17のいず れかに記載のプロモーターを含む、請求項20〜23のいずれかに記載の方法。 25.請求項11に記載の植物形質転換ベクターを包含する、植物を形質転換す るためのキット。 26.請求項2に記載のキメラ遺伝子を含む植物形質転換ベクターを含む、トラ ンスジェニック植物において選択可能なマーカー遺伝子の発現を提供するための キット。 27.前記異種DNA配列がネオマイシンホスホトランスフェラーゼをコードする 、請求項26に記載のキット。 28.請求項14〜17のいずれかに記載のプロモーターを含む、請求項26ま たは請求項27に記載のキット。
