Abstract

Dibenzopyran derivs. of formula (I) and their pharmaceutically acceptable acid addn. salts when R1 is NR4R5 and/or W is pyridyl, are new. In the formula R is H or 1-5C alkanoyl, R1 is H 1-5C alkanoyl or CO(CH2)pNR4R. p is 0-4. R4 and R5 are H or 1-4C alkyl, or NR4R5 is piperidino, pyrrolo, pyrrolidino, morpholino or N-(1-4C alkyl) piperazino. R2 and R3 are H or CH3. Z is 1-10C alkylene or (alk1)m-O-(alk2)n-. Each of alk1 and alk2 are 1-10C alkylene with not >10C in total, and m and n are 0 or 1. W is H, pyridyl or 4-W1-phenyl. W1 is H, F or Cl. Also new are intermediates of formula (II); where W' is as W but not H. (I) have analgesic and tranquillising activities in humans and other mammals. They are not narcotic, do not induce dependence and the analgesic effect is not lost during prolonged use. The pref. oral dose is 0.1-50 mg/day and the pref. parenteral dose 0.01-20 mg/day. A typical cpd. is dl-6 alpha beta, 7,8,9,10,10 alpha-hexahydro-1-hydroxy-6,6-dimethyl-9 beta-hydroxymethyl-3- 2-(5-phenylpentyloxy -6H-dibenzo b,d pyran.
