Abstract

1424571 3,4-Dihydrocarbostyril derivatives OTSUKA PHARMACEUTICAL CO Ltd 22 Jan 1973 [13 April 1972 (2) 14 Sept 1972 (3) 2 Dec 1972 14 Dec 1972 21 Dec 1972] 3251/73 Heading C2C The invention comprises 3,4-dihydrocarbostyril derivatives of Formula (I) and Formula (III) in which R<SP>1</SP> is hydrogen, a C 1 -C 2 alkyl group, a C 2 -C 4 alkenyl group or an aralkyl group, R<SP>2</SP> is hydrogen or a group -COR<SP>3</SP> in which R<SP>3</SP> is a C 1 -C 4 alkyl group, a phenyl group or a 3,4,5- trimethoxyphenyl group; and R' and R" are the same or different and separately represent hydrogen, a C 1 -C 4 alkyl group, a phenalkyl group or a cycloalkyl group or together with the nitrogen atom to which they are attached form a heterocyclic group which may have an oxygen atom or an additional nitrogen atom as a ring member and their acid addition salts. Those compounds of Formula III in which R<SP>1</SP> and R' are both hydrogen are excluded from the scope of the invention. Those compounds of Formula (I) in which R<SP>2</SP> is H may be prepared by reacting a compound of Formula (II) in which Z is -CHOHCH 2 Y (Y is halogen) or with an amine of formula HNR'R" or (b) hydrogenating a compound of the general Formula (III) in the presence of a metal catalyst, e.g. nickel, palladium or platinum, and an organic solvent. When R<SP>2</SP> is a group -COR<SP>3</SP> the compounds may be obtained by acylating the hydroxy compound with an acylating agent R<SP>3</SP>COCl. The compounds of Formula III may be prepared by reacting a compound of Formula (VIIIa) in which X is a halogen, or a compound of Formula VIIIb with an amine of formula HNR'R". The products of Formulae (I) and (III) and their acid addition salts may be used as active agents in pharmaceutical compositions which may be produced in the form of tablets, powders or injectable solutions.
