Abstract
1,250,719. Substituted N-benzoylpiperidene derivatives. LOVENS KEMISKE FABRIK PRODUKTIONSAKTIESELSKAB. 16 March, 1970 [19 March, 1969], No. 14529/69. Heading C2C. Novel compounds of formula wherein R 1 is a C 3-12 aliphatic hydrocarbon chain optionally attached directly or through O or S to the ring and optionally being substituted by phenyl, phenoxy or phenylthio, R 2 is alkyl and R 3 is C 5-8 cycloalkyl or R 2 and R 3 with N represents an optionally alkyl substituted heterocyclic ring, n 1 is 2 to 4, n 2 is 1 to 5 and n3 is 1 to 3, and pharmaceutically acceptable acid addition salts, are prepared by reacting a compound of Formula III with an acid halide, an anhydride, a mixed anhydride with an alkylcarbonic, carboxylic, sulphonic or inorganic acid, of an acid of Formula II or a reactive derivative thereof formed by reaction with a carbodiimide or a N,N<SP>1</SP>-carbonyldiimidazole. Intermediates of Formula II above are prepared by either diazotization of an aminobenzoic acid followed by reaction with potassium xanthogenate and an alkyl halide or by reaction of an ethyl hydroxybenzoate and an R 1 - bromide followed by hydrolysis of the ester. Piperidino alkylptperidines of Formula III above are prepared by reduction of the corresponding piperidino alkylpyridines which are prepared by reaction of 4-chloromethylpyridine or 4-vinylpyridine with the appropriate piperidine or alkylpiperidine respectively. 4 - (2 - N - Methylcyclohexylamino - ethyl)- piperidine is prepared by reaction of N-methylcyclohexylamine and 4-vinylpyridine followed by reduction. Pharmaceutical compositions in conventional forms for oral or parenteral administration and useful in treating Parkinsonism and having anticholinergic action comprise an above novel compound or salt and one or more pharmaceutical carriers therefor.
