Abstract

1,139,164. Heterocyclic acetic acids. PARKE, DAVIS & CO. 17 Nov., 1967 [5 Dec., 1966, 13 Oct., 1967], No. 52529/67. Heading C2C. Novel compounds of the general Formula I where R is hydrogen, a pharmaceuticallyacceptable cation, a lower alkyl radical, or a lower dialkylaminoalkyl radical, R 1 is phenyl, m- or p-halophenyl, m- or p-tolyl, m- or panisyl and Z is oxygen or sulphur, are prepared by hydrolysing a compound containing a group hydrolysable to a carboxyl group or by reacting the hydrazone of a heterocyclic glyoxylic acid with a base, and, if desired, esterifying the resulting acid formed. The 5-aryl-2-furan (or thiophene) acetonitrile is prepared by reacting sodium cyanide with the corresponding bromide which is derived from the corresponding aldehyde via the corresponding alcohol by methods known per se. The 5-aryl-2-furan (or thiophene) aldehyde is prepared by reacting furaldehyde (or thiophenealdehyde) with the corresponding diazotized aniline. The 5-aryl-2-furan (or thiophene) glyoxylic acid is prepared by reacting 2-arylfuran (or 2-arylthiophene) with ethyl oxalyl chloride. Pharmaceutical compositions containing as active ingredients compounds of Formula I together with a suitable carrier are administered orally or parenterally and have anti-inflammatory activity.
