Abstract

L'invention est un procédé d'estimation d'un volume représentatif de particules d'intérêt (10i) baignant dans un échantillon, l'échantillon s'étendant selon au moins un plan, dit plan de l'échantillon (P10), l'échantillon comportant un agent sphérisant, apte à modifier la forme des particules, le procédé comportant les étapes suivantes : a) illumination de l'échantillon à l'aide d'une source de lumière (11), la source de lumière émettant une onde lumineuse incidente (12) se propageant vers l'échantillon (10) selon un axe de propagation (Z) ; b) acquisition, à l'aide d'un capteur d'image (16), d'une image (I0) de l'échantillon (10), formée dans un plan de détection (P0), l'échantillon étant disposé entre la source de lumière (11) et le capteur d'image (16), chaque image étant représentative d'une onde lumineuse (14) dite d'exposition, à laquelle est exposé le capteur d'image (16) sous l'effet de l'illumination ; c) utilisation de l'image de l'échantillon (I0), acquise lors de l'étape b), et d'un opérateur de propagation holographique, pour calculer une expression complexe (A(x,y, z)) de l'onde lumineuse d'exposition (14) en différentes positions par rapport au plan de détection; le procédé comportant une étape d'estimation du volume représentatif (V) des particules d'intérêt (10i) en fonction des expressions complexes calculées lors de l'étape c)
