Abstract
Description
E.ROZENGURT "Biochemical basis of the early events stilated by serum and mitogenic factors in cultures of quiescent ceps, Cold Spring Harbor Conferences on Cell
Proliferation, vol. 6A & 6B, Cold Spring Harbor Laboratory, N.Y., 1978, pp. 773-788
M. PRUNIERS "Recent Advances in Epidermal Cell Cultures
Arch. Dermatol. Res., 1979, 164 : 2, 243-248. The main patent cited relates to "New biological extracts for use in Cosineology and the processes for obtaining these extracts". These extracts contain growth factors, peptidic constituents of low molecular weight (500 to 50,000 daltons), heat-resistant up to 800 C and stable in a pH range of between 2 and 9. They are obtained by thermal degradation, digestion enzymatic and molecular sieving of the starting substances. The characteristics of these extracts: demonstrable activity in vitro, on cell cultures, safety and stability are particularly advantageous for use in cosmetics. The main patent (claim 9) claims the use of products having a at least partial activity of "growth factors
This certificate of addition is more generally concerned with the use in cosmetology of growth factors, as defined in biochemistry and cell biology, since about twenty years. years and of which we find summary reviews in the following articles, for example
E.ROZENGURT "Biochemical basis of the early events by serum and mitogenic factors in cultures of quiescent ceps, Cold Spring Harbor Conferences on Cell
Proliferation, vol. 6A & 6B, Cold Spring Harbor Laboratory, NY, 1978, pp. 773-788
M. PRUNIERS Recent Advances in Epidermal Cell Cultures
Arch. Dermatol. Res., 1979, 164: 2, 243-248.
It is recalled that the growth factors in biochemistry and cell biology are substances that are capable of stimulating the proliferation of cells (ie, which have a mitoyl action). In a more practical way, they are substances which may at least partly replace the calf serum, or else have a synergy therewith, for the survival and growth of cell lines cultured in a minimum nutrient medium, for example the MEM medium according to
HANKS. The growth factors may optionally have specificities for certain classes of cells.
The inventors have studied for a cosmetic use of growth factors, those of them which exhibit mitogenic activity on cell lines of skin in culture: epidermal cells, fibroblasts and chondrocytes. The table in the appendix presents examples of defined growth factors for which the cosmetic use has been studied, without this list being exhaustive.
The inventors have demonstrated a satisfactory parallelism between the in vitro properties of these growth factors and the chemical properties of the cosmetic preparations containing these. They discovered that these substances incorporated in finished cosmetic preparations, at low doses (0.001% to 0.1% of active ingredients dry in relation to the finished preparation) exhibit remarkable cosmetological activities.
Biological extracts containing growth factors not yet fully identified have also been examined. These extracts show very similar mitogenic properties on cultured cell lines of cutaneous origin. These extracts come inter alia from 1 animal or human serum 2 - embryonic skin of animal origin 3- milk or human or bovine whey
These three sources are already covered by the main patent.
Conditioned culture media (i.e., where cell cultures have been grown beforehand, especially human skin fibroblast cultures.) The growth of the first culture releasing in the medium of growth, assets in view of subsequent crops.
5- des extraits protéolytiques d'organes tels que :
- cartilages animaux
- peau animale, porcine ou bovine
- rate de bovins
- foie de bovins
- duodénum de porc
- embryons de bovins entiers sans que cette liste ne soit limitative.5- proteolytic extracts of organs such as:
- animal cartilage
- animal skin, swine or beef
- spleen of cattle
- liver of cattle
- pig duodenum
whole bovine embryos without this list being exhaustive.
These extracts are obtained in the same manner as described in the main patent, namely: specific enzymatic hydrolyses, heat treatments and molecular sieving, in order to collect growth factors which generally have low molecular weights (500 to 50 000 dalton) and to remove high molecular weight components which are sometimes responsible for cellular growth inhibition phenomena.
Alternatively, the hydrolysis of the above organs can also be obtained by bacterial fermentation.
Incubation of organ stock, especially animal skin tissue, was incubated with cultures of selected bacterial strains so that the resulting extract exhibited active growth factor properties on cell lines. of epidermal or dermal origin, human or animal.
After hydrplyse, the proteolyzed extracts undergo one or more heat treatments, then if necessary, molecular sieving, so as to recover low molecular weight fragments (from 500 to 50,000 daltons, as above).
The biological extracts are already known and for some, used in cosmetology. The present invention is therefore their use as a growth factor and to do this - first, to adjust the mode of preparation to promote the formation and release of these growth factors according to the previous descriptions
second, to adjust the use doses to optimum values of between 0.01% and 1% of dry organ extract relative to the finished cosmetic product.
