Abstract

Chemical fixation of organic substances contg. a sugar residue to a support contg. >=1 reactive NH2 gp. is effected by oxidising >=1 CH2OH gp. in the sugar residue to CHO, and reacting the resulting CHO gps. with NH2 gps. carried by side chains fixed to an insoluble solid support. When the NH2 gp. is a prim. amine function, the resulting Schiff's base is stabilised by redn. Process can be used to immobilise glycoproteins such as antibodies, antigens, hormones and enzymes and also glycolipids and polysaccharides. The immobilised antibodies and antigens are esp. suitable for use in agglutination-type immunoassay tests. The organic substances are fixed in highly stable form and in good yields without loss of their specific activity. In an example, equine immunogammaglobulins were oxidised with aq. NaIO4 and coupled with a support obtd. by reacting carboxylated polystyrene spheres with hexamethylenediamine. The prod. was then reduced with NaBH4.
