Abstract

Crude urokinase extracts from human and urine, are purified by (a) two stages of exclusion chromatography on DEAE cellulose, the first at pH 4-6 and conductivity 15000-25000 mu m hos/cm, and the second at pH 6-7 and conductivity 15000-18000 mu m hos/cm initially, lowered in stages to 9000 mu m hos/cm before elution. The quantity of adsorbent must be proportional to the urokinase, at 70-60g. dry resin/107 CTA units, (b) further chromatographic purificn. (opt.) (c) fractional pptn. at pH 3-4.5, conductivity 15000-25000 mu m hos/cm and concn. 1.5-3 mg/ml, using a 25-35% satn. of (NH4)2SO4, and to remove pyrogens, (d) complex formation with heparin to stabilize the product.
