Abstract
Esillä oleva keksintö koskee lääkeaineen vapauttamiseen suolistoon tarkoitettuja lääkekoostumuksia, jotka koostuvat pelletistä, jolle suoritetaan myöhemmin kalvopäällystys enteerisellä lateksin tai pseudolateksin vesidispersiolla tai pH-herkkien polymeerien liuoksella. Enteropäällysteiset pallomaiset rakeet (pelletit) valmistetaan edullisesti mikrokiteisen selluloosan ja amylopektiinimaissitärkkelyksen kostearakeistetusta jauheseoksesta. Pellettikoostumus, joka sisältää amylopektiiniä keratäyteaineena, saa aikaan jopa erittäin vesiliukoisen aktiivisen aineen huomattavan vähäisen ennenaikaisen läpäisevyyden simuloidun mahanesteen happamassa ympäristössä. Lääkeaineen vapautuminen tapahtuu luotettavasti ja nopeasti simuloidun suolinesteen korkeammassa pH:ssa. Enteropäällysteisissä pelleteissä aluspäällystemateriaalina käytettynä amylopektiini myös parantaa pellettien haponkestävyysominaisuuksia simuloidussa mahanesteessä.&sr;The present invention relates to pharmaceutical compositions for the release of a drug into the intestine, consisting of a pellet, which is subsequently film-coated with an enteric latex or pseudolatex aqueous dispersion or with a solution of pH-sensitive polymers. Enteric-coated spherical granules (pellets) are preferably prepared from a wet-granulated powder mixture of microcrystalline cellulose and amylopectin corn starch. The pellet composition, which contains amylopectin as a co-filler, provides a significantly reduced premature permeability of even a highly water-soluble active substance in the acidic environment of simulated gastric fluid. The release of the drug occurs reliably and rapidly at a higher pH of simulated gastric fluid. When used as a base coating material in enteric-coated pellets, amylopectin also improves the acid resistance properties of the pellets in simulated gastric fluid.&sr;
