Abstract

Dispositivo (10) para la administración de un fármaco contra la artritis reumatoide a través de una barrera dérmica, comprendiendo el dispositivo: una matriz de microagujas (116) y una pluralidad de nanoestructuras (104) conformadas sobre una superficie de, como mínimo, una de las microagujas (12; 22; 120; 330; 230), estando las nanoestructuras (104) dispuestas en un patrón predeterminado; y caracterizado por que comprende adicionalmente: un depósito (112; 206; 306) que está en comunicación fluida con la matriz de microagujas (116); y un fármaco contra la artritis reumatoide retenido dentro del depósito (112; 206; 306).

Se prepararon varios moldes diferentes utilizando técnicas de fotolitografía similares a las empleadas en el diseño y conformación de circuitos eléctricos. Las etapas individuales del proceso se conocen de forma general en la técnica y han sido descritas.
Inicialmente, los sustratos de silicio se prepararon mediante lavado con acetona, metanol y alcohol isopropilíco, y posteriormente se recubrieron con una capa de 258 nanómetros (nm) de dióxido de silicio según un proceso de deposición química en fase vapor.
A continuación, se conformó un patrón sobre cada sustrato a través de un proceso de formación de patrones por litografía de haz de electrones, tal como se conoce en la técnica, utilizando un sistema JEOL JBX-9300FS EBL. Las condiciones de procesamiento fueron las siguientes:
Corriente del haz = 11 nA Tensión de aceleración = 100 kV Periodo de tiro = 14 nm Dosis = 260 µC/cm2 Resina fotosensible = ZEP520A, ~330 nm de espesor Revelador = acetato de n-amilo Revelado = 2 min de inmersión, seguido de enjuague de 30 s con alcohol isopropílico.
A continuación, se llevó a cabo un grabado de dióxido de silicio con un STS Advanced Oxide Etch (AOE). El tiempo de grabado fue de 50 segundos utilizando 55 centímetros cúbicos estándar por minuto (sccm) de He, 22 sccm de CF4, 20 sccm de C4F8 a 4 mTorr, bobina de 400 W, RIE de 200 W y un DC Bias de 404-411 V.
A continuación, se realizó un grabado con silicio con un grabado con óxido de silicio STS (SOE). El tiempo de grabado fue de 2 minutos utilizando 20 sccm de Cl2 y 5 sccm de Ar a 5 mTorr, bobina de 600 W, RIE de 50 W y un DC Bias de 96-102 V. La profundidad de grabado de silicio fue de 500 nanómetros.
Se utilizó una solución de grabado de óxido tamponada (BOE) para la eliminación del óxido restante, que consistió en una inmersión de BOE de tres minutos seguida por un enjuague con agua desionizada.
Se utilizó una nanoimpresora Obducat NIL-Eitre® 6 para formar patrones nanométricos sobre una variedad de sustratos poliméricos. Se utilizó agua externa como refrigerante. El módulo UV utilizó una sola lámpara pulsada a una longitud de onda comprendida entre 200 y 1.000 nanómetros a 1,8 W/cm2. Se utilizó un filtro UV de 250-400 nanómetros. El área de exposición fue de 6 pulgadas con una temperatura máxima de 200°C y a 80 bar. La nanoimpresora incluía una unidad de separación semiautomática y desmoldeo controlado automático.
Para facilitar la liberación de las películas nanoimpresas desde los moldes, los moldes se trataron con trideca-(1,1,2,2 tetrahidro)octactriclorosilano (F13-TCS). Para tratar un molde, el molde de silicio se limpió primero con un lavado de acetona, metanol y alcohol isopropílico, y se secó con nitrógeno gaseoso. Se colocó una placa de Petri sobre una placa caliente en una atmósfera de nitrógeno y se añadieron 1-5 ml de F13-TCS a la placa de Petri. Se colocó un molde de silicio en la placa de Petri y se cubrió durante 10-15 minutos para permitir que el vapor de F13-TCS humedeciera el molde de silicio antes de retirar el molde.
Se utilizaron cinco polímeros diferentes, según se indican en la siguiente tabla 1, para formar diversos diseños de nanotopografía.

Se formaron diferentes patrones de nanotopografía, cuyas representaciones esquemáticas se ilustran en las figuras 24A-24D. El patrón de nanotopografía ilustrado en la figura 12E era una superficie de un sustrato plano adquirido a NTT Advanced Technology, de Tokio, Japón. Los patrones se designaron DN1 (figura 24A), DN2 (figura 24B), DN3 (figura 24C), DN4 (figura 24D) y NTTAT2 (figura 24E). Las imágenes de SEM de los moldes se muestran en las figuras 24A, 24B, y 24C, y las imágenes de las películas se muestran en las figuras 24D y 24E.
