Abstract
Procedimiento de obtención de células del epitelio pigmentario de la retina (EPR), que comprende: cultivar una o más células madre en un sustrato que comprende una laminina, en el que la laminina es laminina-521 y/o laminina-511, en el que la laminina es una proteína intacta o un fragmento de proteína que contiene uno, dos o tres dominios funcionales que posee actividad de unión a otra molécula o receptor; exponer las células madre a un primer medio de cultivo celular que contiene un factor de crecimiento, en el que el factor de crecimiento es FGF2 en una cantidad mayor que cero a 3,9 ng/ml, siendo el primer medio de cultivo celular completamente definido químicamente y no xenogénico; después de un primer período de tiempo, eliminar el primer medio de cultivo celular y exponer las células madre a un segundo medio de cultivo celular que no contiene el factor de crecimiento, siendo el segundo medio de cultivo celular completamente definido químicamente y no xenogénico; y cambiar periódicamente el segundo medio de cultivo celular para obtener las células del EPR.

La fase líquida del medio de cultivo celular puede ser agua, suero o albúmina.
Muchos de los ingredientes o componentes enumerados anteriormente en la tabla 1 no son necesarios o pueden 5 utilizarse en concentraciones más bajas.
Se contempla que el medio de cultivo celular puede contener insulina o un sustituto de insulina. De forma similar, el medio de cultivo celular puede contener transferrina o un sustituto de transferrina.
10 En realizaciones más específicas, se contempla que el medio de cultivo celular puede no contener (1) albúmina, (2) insulina o sustituto de insulina, (3) transferrina o sustituto de transferrina o cualquier combinación de estos tres componentes.
Debe observarse que otros medios de cultivo celular pueden contener factores de crecimiento, tales como 15 interleucina-1 beta (IL-1β o catabolina), interleucina-6 (IL6) o factor derivado del epitelio pigmentario (PEDF). Dichos factores de crecimiento no están presentes en el medio de cultivo celular de la presente descripción.

La combinación del sustrato de laminina con el medio de cultivo celular de la presente divulgación da como resultado un sistema de cultivo celular que puede ser más barato, aunque da a conocer una mayor eficacia en el mantenimiento de células madre pluripotenciales. En esencia, todo lo que se requiere es una laminina y una 5 cantidad mínima de nutrición. Se contempla particularmente que la laminina utilizada en combinación con este medio de cultivo celular sea LN-511 o LN-521.
Los siguientes ejemplos son para fines ilustrativos adicionales la presente descripción. Los ejemplos son meramente ilustrativos y no están destinados a limitar dispositivos fabricados de acuerdo con la presente divulgación a los 10 materiales, condiciones o parámetros de proceso establecidos en los mismos.

seroalbúmina bovina adicional, se obtuvieron tres nuevas líneas de células hES que no son parte de la presente invención a partir de seis blastocitos cultivados. Cuatro días después de la siembra en placas, las masas internas de células produjeron excrecencias similares a células madre. La figura 24 muestra una derivación en una etapa temprana de una nueva línea celular del tronco embrionario humano HS841 que no es parte de la presente
5 invención sobre la matriz de laminina-521/E-cadherina. Se muestran células madre embrionarias humanas de morfología típica que crecen a partir de la masa celular interna de un blastocisto del día seis cuatro días después del aislamiento mecánico de la masa celular interna y de la siembra en placas sobre la matriz de laminina-521/E-cadherina.
10 Inesperadamente, el sistema y el procedimiento de cultivo celular dieron líneas celulares hES estables en 3 de 6 embriones (50%), una tasa de derivación más alta que la de los procedimientos estándar.
La utilización de una matriz de laminina-521/E-cadherina y medio TeSR2 con rHSA adicional, un entorno completamente definido químicamente no xenogénico, produjo resultados similares en la derivación de nuevas
15 líneas celulares hES que no son parte de la presente invención.
La tabla 5 a continuación da a conocer la formulación para el medio mTeSR1 utilizado en este ejemplo. Las cantidades de cada ingrediente pueden variar hasta en un 20%.
20 En realizaciones particulares, la cantidad de seroalbúmina humana (HSA) se puede variar desde una concentración de 0,195 mM a 1 mM, incluyendo de 0,3 mM a 1 mM o de 0,3 mM a aproximadamente 0,4 mM. La cantidad de bFGF también se puede variar de 0 a aproximadamente 105 ng/ml o de 0 a 3,9 ng/ml o de 0,5 ng/ml a 3,5 ng/ml.
