Abstract
Dispositivo compartimentado y método de captura y doble amplificación enzimática de secuencias diana de ácidos nucleicos en un solo dispositivo compartimentado cerrado. El dispositivo compartimentado (1) consta de un tubo de reacción que comprende dos compartimentos (2) y (3), que configuran dos zonas físicamente separadas y destinadas a contener reactivos, y una tapa (4). El método incluye el empleo de dicho dispositivo y comprende la captura, específica o no, de una secuencia diana, su transcripción reversa (en su caso), una primera amplificación enzimática y una segunda amplificación enzimática de una diana presente en los productos resultantes de la primera amplificación enzimática, en un solo dispositivo que contiene todos los reactivos necesarios para dichas reacciones y sin necesidad de abrir el dispositivo una vez iniciada la reacción de transcripción reversa (en su caso) o la primera reacción amplificación enzimática. The method includes the use of said device and comprises the capture, specific or not, of a target sequence, its reverse transcription (if any), a first enzymatic amplification and a second enzymatic amplification of a target present in the products resulting from the first enzymatic amplification, in a single device containing all the reagents necessary for said reactions and without opening the device once the reverse transcription reaction (if applicable) or the first enzymatic amplification reaction is initiated. The method has application in the detection of nucleic acid targets.
