Abstract

Изобретение относится к области медицины, в частности к клинической микробиологии, и может быть использовано для преодоления устойчивости грамотрицательных микроорганизмов вида Acinetobacter baumannii, продуцирующих металло-β-лактамазу (МβЛ), к карбапенемам. Указанный технический результат достигается тем, что клодроновая или этидроновая кислоты из группы бисфосфонатов ингибируют стандартный реактив - металло-β-лактамазу P.aeruginosa, рекомбинантную, экспрессированную в Escherichia coli. Достигнутый результат позволил выявить антимикробный эффект комбинации клодроновой или этидроновой кислот с карбапенемами (имипенемом или меропенемом) в отношении референс-штамма A.baumannii ATCC BAA-747, резистентного к карбапенемам, в присутствии стандартного реактива МβЛ, инактивирующего эти антибиотики. Предложенное техническое решение позволяет моделировать получение штаммов грамотрицательных микроорганизмов с различным уровнем резистентности к карбапенемам и оценивать эффективность ингибирования металло-β-лактамаз.The invention relates to the field of medicine, in particular to clinical microbiology, and can be used to overcome the resistance of Gram-negative microorganisms of the species Acinetobacter baumannii, producing metallo-β-lactamase (МβЛ), to carbapenems. The achieved result allowed to reveal the antimicrobial effect of the combination of clodronic or etidronic acids with carbapenems (imipenem or meropenem) with respect to the reference A.baumannii ATCC BAA-747 resistant antibiotic The proposed solution allows to simulate the production of strains of gram-negative microorganisms with different levels of resistance to carbapenems and to assess the effectiveness of inhibition of metallo-β-lactamase.
