Abstract

Система селекции, лишенная генов устойчивости к антибиотикам, которая основана на применении гена araD в качестве маркера селекции, который несет вектор, встроенный в бактериальный штамм, дефицитный по гену araD. Ген araD из E.coli кодирует L-рибулозо-5-фосфат-4-эпимеразу. Способ селекции клеток, трансформированных плазмидой, которая содержит ген araD. Не связанный с антибиотиками маркер селекции делает данную систему подходящей для продукции терапевтических средств. Ген araD не существенен для роста хозяев, но манипуляция с ним влияет на рост в некоторых селективных условиях. Делеция araD ведет к накоплению вещества, токсичного для хозяина, но не для людей. Ген araD относительно мал, и поэтому может конструироваться малая плазмида, которая требует меньше энергии для репликации, и приводит к повышенным скорости роста и выходу.
