Abstract

Раскрываются процессы получения устойчиво трансформируемой высокопродуктивной пшеницы и система трансформации пшеницы с помощью Agrobacterium. Настоящее изобретение представляет способы трансформации и различные эксплантаты, например свежевыделенные либо предварительно культивированные незрелые зародыши, зародышевый каллюс и суспензионные клетки. Раскрывают также способы выделения трансгенных растений после трансформации в течение непродолжительного периода времени при условии, если эксплантаты регенерируются во время трансформации. Таким образом, существенно снижается частота сомаклональных изменений, связанных с продолжительным периодом культивирования in vitro. Частота трансформации с использованием этой системы сопоставима либо превосходит опубликованные способы с использованием других систем, например бомбардировки микроносителями.Отчет о международном поиске опубликован 1998.03.19.The processes of obtaining stably transformed highly productive wheat and the system of wheat transformation using Agrobacterium are revealed. The present invention provides transformation methods and various explants, such as freshly isolated or pre-cultured immature embryos, germ callus and suspension cells. Methods for isolating transgenic plants after transformation for a short period of time are also disclosed, provided the explants are regenerated during transformation. Thus, the frequency of somaclonal changes associated with a long period of in vitro cultivation is significantly reduced. The frequency of transformation using this system is comparable or exceeds published methods using other systems, such as microcarrier bombardment.
