Claims (16)
1. Fremgangsmåde til reduktion afvandaktiviteten i et bakterieholdigt koncentrat omfattende følgende trin: a) tilvejebringelse af et første bakterieholdigt koncentrat; b) tilsætning af en vandig opløsning til det første bakterieholdige koncentrat for at tilvejebringe en bakterieholdig suspension, hvor volumenet af den tilsatte vandige opløsning ligger i området fra 0,3 liter til 10 liter pr. liter af det første bakterieholdige koncentrat; c) koncentrering af den bakterieholdige suspension for at tilvejebringe et andet bakterieholdigt koncentrat; d) frysetørring af det andet bakterieholdige koncentrat ved et tryk på 0,5 til 2,0 mBar.
2. Fremgangsmåde ifølge krav 1, der yderligere omfatter ét eller flere af de følgende trin: b1) blanding og c1) tilsætning af et kryobeskyttelsesmiddel og/eller en stabilisator.
3. Fremgangsmåde ifølge krav 1 til 2, hvor den vandige opløsning omfatter fra 1 til 300 g/l af et carbohydrat, hvor carbohydratet fortrinsvis er saccharose eller trehalose.
4. Fremgangsmåde ifølge krav 1, hvor volumenet af den vandige opløsning ligger i området fra 0,5 liter til 6 liter pr. liter af det første bakterieholdige koncentrat, såsom i området fra 1 liter til 4 liter, i området fra 1,5 liter til 3,5 liter, i området fra 1,0 liter til 3,0 liter, i området fra 1,0 liter til 2,0 liter, i området fra 1,0 liter til 1,5 liter, i området fra 1,5 liter til 2,0 liter eller i området fra 2 liter til 3 liter.
5. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor koncentreringen i trin c) udføres som et centrifugeringstrin, og centrifugeringstrinnet fortrinsvis udføres ved en centrifugeringskraft fra ca. 400 til ca. 65000 x g, fortrinsvis fra ca. 4000 til ca. 20000 x g eller fra ca. 8000 til ca. 15000 x g; eller hvor koncentreringen i trin c) udføres som et filtreringstrin, hvor filtreringstrinnet omfatter mikrofiltrering, fortrinsvis under anvendelse af en filtreringsmembran med en porestørrelse på 0,1 til 10 mikrometer, eller filtreringstrinnet omfatter ultrafiltrering, fortrinsvis under anvendelse af en filtreringsmembran med en molekylvægtsgrænse på ca. 5 til ca. 200 kDa.
6. Fremgangsmåde ifølge krav 5, hvor filtreringstrinnet omfatter tangentiel filtrering.
7. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor det første bakterieholdige koncentrat koncentreres 5 til 25 gange, såsom 10-20 gange, baseret på fermenteringsvæskens volumen.
8. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor det bakterieholdige koncentrat er et mælkesyrebakterieholdigt koncentrat.
9. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor det bakterieholdige koncentrat omfatter en bakterie valgt fra gruppen bestående af Acetobacter, Bifidobacterium, Carnobacterium, Enterococcus, Lactococcus, Lactobacillus, Leucono-stoc, Pediococcus, Oenococcus, Propionibacterium og Streptococcus.
10. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor det bakterieholdige koncentrat omfatter mindst én mælkesyrebakterieslægt, der fortrinsvis er valgt fra gruppen bestående af Lactococcus, Lactobacillus, Leuconostoc, Carnobacterium, Pediococcus og Streptococcus, og mere fortrinsvis mindst én art, der er valgt fra gruppen bestående af Leuconostoc spp., Bifidobacterium ssp, Lactococcus lactis, Lactococcus cremoris, Lactobacillus acidophilus, Lactobacillus casei, Lactobacillus kefir, Lactobacillus bifidus, Lactobacillus brevis, Lactobacillus helveticus, Lactobacillus para-casei, Lactobacillus rhamnosus, Lactobacillus salivarius, Lactobacillus curvatus, Lactobacillus bulgaricus, Lactobacillus sake, Lactobacillus reuteri, Lactobacillus lactis, Lactobacillus delbreuckii, Lactobacillus plantarum og Streptococcus thermophilus.
11. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor bakterierne er fra en stamme, der er valgt fra gruppen bestående af BB-12®, som er deponeret hos Deutsche Sammlung von Mikroorganismen und Zellkulturen (DSMZ) under deponeringsnummer DSM15954, BB-12® free, der er deponeret hos Deutsche Sammlung von Mikroorganismen und Zellkulturen (DSMZ) under deponeringsnummer DSM17281, LA-5®, der er deponeret hos Deutsche Sammlung von Mikroorganismen und Zellkulturen (DSMZ) under deponeringsnummer DSM13241, ST6008, der er deponeret hos Deutsche Sammlung von Mikroorganismen und Zellkulturen (DSMZ) under deponeringsnummer DSM18111, LGG®, der er deponeret hos American Tissue Type Collection Center under deponeringsnummer ATCC53103, og en mutant/variant af en hvilken som helst af disse.
12. Bakterieholdigt koncentrat, som er opnået ved fremgangsmåden ifølge et hvilket som helst af de foregående krav.
13. Bakterieholdigt koncentrat ifølge krav 12, hvor koncentratet er i form af et frysetørret koncentrat, som har lavere vandaktivitet (aw) end et frysetørret koncentrat, som er fremstillet under identiske betingelser, men med et uvasket koncentrat.
14. Bakterieholdigt koncentrat ifølge et hvilket som helst af kravene 12-13, hvor koncentratet er frysetørret ved et højere vakuum end muligt for det uvaskede koncentrat, og hvor koncentratet fortrinsvis er frysetørret ved et tryk i området 0,2-1,2 mBar eller 0,3-0,9 mBar.
15. Bakterieholdigt koncentrat ifølge et hvilket som helst af kravene 12-14, hvor koncentratet er i form af et frysetørret koncentrat, der (a) har en højere renhed og/eller hvidere farve end det frysetørrede koncentrat, der er fremstillet under identiske betingelser, men med et uvasket koncentrat, eller (b) er lettere at formale end det frysetørrede koncentrat, der er fremstillet under identiske betingelser, men med et uvasket koncentrat.
16. Bakterieholdigt koncentrat ifølge et hvilket som helst af kravene 12-15, hvor koncentratet omfatter fra 109 til 1013 cfu/g mælkesyrebakterieceller.
