Claims (15)
- Alpha-amylase- (AMYS) varianter af geobacillus stearothermophilus med forbedrede egenskaber PATENTKRAV1. Polypeptidvariant, der har α-amylaseaktivitet, hvor polypeptidvarianten har mindst én ændret egenskab, der forbedrer enzymperformance, hvilken polypeptidvariant omfatter: en aminosyresekvens, der er mindst 90 % aminosyresekvensidentisk med et parentalt a-amylase-polypeptid, der er udvalgt fra AmyS og har sekvensen SEQ ID NO: 1, eller en trunkeret variant af AmyS, der har sekvensen SEQ ID NO: 2, og der har mindst følgende mutation ved en aminosyrerest, der svarer til den af det parentale a-amylase-polypeptid som bestemt ved alignment af polypeptidvarianterne med det parentale polypeptid, hvor mutationen ændrer aminosyreresten fra den for de parentale polypeptider, og hvor mutationen er en substitution, der introducerer én eller flere af aminosyreresteme, der er udvalgt fra gruppen bestående af 188P og 188D.
- 2. Variant ifølge krav 1, hvor substitutionen er 188P, og varianten har forbedret termostabilitet sammenlignet med det parentale polypeptid.
- 3. Variant ifølge krav 1, hvor substitutionen er 188D, og varianten udviser forøget aktivitet eller ekspression sammenlignet med det parentale polypeptid.
- 4. Variant ifølge krav 1, hvor substitutionen er E188P, og varianten udviser forøget viskositetsreduktion i en stivelsessmeltningsassay sammenlignet med det parentale polypeptid.
- 5. Variant ifølge et hvilket som helst af kravene 1-4, hvor mutationen er til stede i kombination med glutamin ved positionen 242, eller hvor mutationen er til stede i kombination med en deletion ved positionerne 179 og 180.
- 6. Variant ifølge et hvilket som helst af kravene 1-4, hvor det parentale polypeptid indbefatter en trunkering af C-terminale 29 aminosyrerester.

- 15. Anvendelse af en α-amylasevariant ifølge et hvilket som helst af kravene 1-7 i: (i) afsletning af tekstiler, stoffer og beklædningsgenstande eller (ii) i papirmasse- og papirproduktion.
