Claims (21)

1. Gær eller trådformet svampeværtscelle, der er transformeret med en nukleinsyrekonstruktion, der omfatter en nukleotidsekvens, der koder for en xyloseisomerase, hvilken xyloseisomerase omfatter en aminosyresekvens, der har mindst 95 % sekvensidentitet med aminosyresekvensen ifølge SEQ ID NO: 2, hvor nukleinsyrekonstruktionen ved transformation af værtscellen bibringer værtscellen evnen til direkte isomerisering af xylose til xylulose, hvor værtscellen omfatter en genmodifikation, der øger strømningen via pentosephosphat-reaktionsvejen, og hvor genmodifikationen omfatter overekspression af mindst ét gen i den ikke-oxidative del af pentosephosphat-reaktionsvejen sammenlignet med en værtscelle, der er genetisk identisk bortset fra genmodifikationen.
2. Værtscelle ifølge krav 1, hvor nukleotidsekvensen koder for en xyloseisomerase, der kan opnås fra en bakterie fra klassen Bacteroides.
3. Værtscelle ifølge krav 1 eller 2, hvor genet i den ikke-oxidative del af pentosephosphat-reaktionsvejen er udvalgt fra gruppen, der består af generne, der koder for ribulose-5-phosphatisomerase, ribulose-5-phosphatepimerase, transketolase og transaldolase.
4. Værtscelle ifølge krav 3, hvor genmodifikationen omfatter overekspression af mindst generne, der koder for en transketolase og en transaldolase.

6. Værtscelle ifølge krav 5, hvor genmodifikationen omfatter overekspression af et gen, der koder for en xylulosekinase.
7. Værtscelle ifølge et hvilket som helst af kravene 1-6, hvor genet, der overudtrykkes, er endogent for værtscellen.
8. Værtscelle ifølge et hvilket som helst af kravene 1-7, hvor værtscellen omfatter en genmodifikation, der reducerer uspecifik aldosereductaseaktivitet i værtscellen.
9. Værtscelle ifølge krav 8, hvor genmodifikationen reducerer ekspressionen af eller inaktiverer et gen, der koder for en uspecifik aldosereductase.
10. Værtscelle ifølge krav 9, hvor genet er inaktiveret ved deletion af mindst en del af genet eller ved destruktion af genet.
11. Værtscelle ifølge krav 8 eller 9, hvor ekspressionen af hvert gen i værtscellen, der koder for en uspecifik aldosereductase, er reduceret eller inaktiveret.
12. Værtscelle ifølge et hvilket som helst af ovennævnte krav, hvor værtscellen er en gær, fortrinsvis en gær, der tilhører en af slægterne: Saccharomyces, Kluyveromyces, Candida, Pichia, Schizosaccharomyces, Hansenula, Kloeckera, Schwanniomyces og Yarrowia.
13. Værtscelle ifølge krav 12, hvor gæren tilhører en af arterne: S. cerevisiae, S. bulderi, S. barnetti, S. exiguus, S. uvarum, S. diastaticus, K. lactis, K. marxianus og K. fragilis.
14. Værtscelle ifølge et hvilket som helst af kravene 1-11, hvor værtscellen er en trådformet svamp, fortrinsvis en trådformet svamp, der tilhører en af slægterne: Aspergillus, Trichoderma, Humicola, Acremonium, Fusarium og Penicillium.
15. Værtscelle ifølge et hvilket som helst af ovennævnte krav, hvor værtscellen udtrykker et eller flere enzymer, der bibringer værtscellen evnen til at danne mindst ét fermenteringsprodukt udvalgt fra gruppen, der består af ethanol, mælkesyre, 3-hydroxypropionsyre, acrylsyre, eddikesyre, ravsyre, citronsyre, aminosyrer, 1,3-propandiol, ethylen, glycerol, β-lactam-antibiotika og cephalosporiner.
16. Fremgangsmåde til fremstillng af ethanol, hvilken fremgangsmåde omfatter trinene: (a) fermentering af et medium, der indeholder en kilde af xylose, med en modificeret værtscelle ifølge et hvilket som helst af kravene 1-14, hvor værtscellen fermenterer xylose til ethanol, og eventuelt (b) indvinding af ethanol.
17. Fremgangsmåde ifølge krav 16, hvor mediet også indeholder en kilde af glucose.
18. Fremgangsmåde ifølge krav 16 eller 17, hvor den volumetriske produktivitet af ethanol er mindst 0,5 g ethanol pr. liter pr. time.
19. Fremgangsmåde ifølge et hvilket som helst af kravene 16-18, hvor ethanoludbyttet er mindst 50 %.
20. Fremgangsmåde til fremstilling af et fermenteringsprodukt udvalgt fra gruppen, der består af mælkesyre, 3- hydroxypropionsyre, acrylsyre, eddikesyre, ravsyre, citronsyre, aminosyrer, 1,3-propandiol, ethylen, glycerol, β-lactam-antibiotika og cephalosporiner, hvilken fremgangsmåde omfatter trinene: (a) fermentering at et medium, der indeholder en kilde af xylose, med en modificeret værtscelle ifølge krav 15, hvor værtscellen fermenterer xylose til fermenteringsproduktet, og eventuelt (b) indvinding af fermenteringproduktet.
21. Fremgangsmåde ifølge krav 20, hvor mediet også indeholder en kilde af glucose.
