Abstract
本发明提供一种新型灵芝营养液及其制备方法,其特征是将菌种在培养基滤液中进行培养。本发明的营养液清亮呈棕色果冻状,口感纯正,灵芝多糖,维生素、氨基酸和微量元素的含量丰富,是一种优良的增强体质营养液。
Description
本发明涉及一种广泛使用的增强体质的新型灵芝培养液及其制备方法。
灵芝(Ganoderma Lucidum)历来视为中华医药宝库的奇葩异草。千百年来其药用价值已得到广泛地认同,它可以提高人体的免疫机能,是WHO确认的理想免疫增强剂。灵芝多糖的抗衰老作用机理,已被证实。因此,将其以合理的制剂形式加以利用,更好地为增强人们的体质服务,是当前的主要课题。
迄今,灵芝制剂多以子实体或固体培养菌丝为原料,生产周期长,易受环境污染,当然对其品质产生影响。液体培养技术在此领域的应用,实现了灵芝的大规模工业化培养。但遣憾的是,现行的液体培养技术中,培养基全部入培养罐,培养基含有不溶性固形物,在液体培养过程中释放出酵素,致使培养液有异味,而且引起菌丝、不溶性固形物和培养液的分离困难。
本发明正是基于此而提出的,为解决上述问题,本发明在液体培养工艺中增设配料罐,培养基经配料罐处理后,将不溶性的固形物分去,将滤液输入培养罐,接种菌种后进行液体培养。经过本发明人的多次反复研究,确定了培养基滤液配伍量与灵芝菌种接种量的最佳比例。经本发明方法制得的灵芝培养液含有所有菌丝中的有效成分,以及丰富的维生素,多糖、氨基酸和微量元素。产品口感纯正、酸甜微苦,无异味。
本发明的灵芝培养液的培养基的配置如下(W/V):麦麸粉3—8%,豆饼粉2—5%,玉米粉2-5%,蔗糖4—8%,磷酸二氢钾0.03-0.1%,硫酸镁0.01-0.%,硫酸锌5-10PPM,硫酸亚铁10-20PPM,硫酸锰10-20PPM,余为水。
本发明使用的菌种为灵芝Ganoderma Lucidum(Leysserex Fries)Karsten保存菌种。
本发明的灵芝营养液是由上述灵芝菌种在所述的培养基滤液中培养而获得的。具体地,本发明的灵芝培养液的制备方法如下:
(1)用保存菌种[灵芝Ganoderma Lucidum(Leysserex Fries)Karsten]在PDA斜面培养基上接种,在25—30℃下培养10—15天,得到斜面菌种;
(2)以麦麸粉3—8%(W/V,下同),豆饼粉2—5%,玉米粉2—5%,蔗糖4—8%,磷酸二氢钾0.03-0.1%,硫酸镁0.01-0.1%,硫酸锌5—10PPM,硫酸亚铁10—20PPM,硫酸锰10—20PPM的量进行配制(配料罐),升温到90℃,保持2小时,滤去不溶物,得到不含不溶物固形物的液养基滤液。
(3)将(1)制得的斜面菌种接种到(2)所得的培养基滤液内培养,温度为25—30℃,时间为2—3天,液体培养菌种采用1—3级摇瓶和1—2级种子罐逐级扩大培养。摇瓶培养基装量40%,接种量10—20%,旋转式摇瓶机转速150—200RPM,然后于培养罐中培养,培养罐中培养基装量50—70%,接种量15—30%,温度25—28℃,时间为2—3天,通气量1∶0.8-1.5,搅拌速度130—2DDRPM,保持罐压0.5KG/平方厘米。
(4)培养终止后,培养液升温至60℃,保持1—2小时,菌丝自溶,分离去掉不溶固形物,在≤6D℃下,真空减压蒸发浓缩20倍。
以下用具体实施例说明本发明。
实施例1:
用2000毫升三角瓶,按下列组成配制培养基:麦麸粉30克,豆饼粉20克,玉米粉20克,蔗糖30克,磷酸二氢钾0.3克,硫酸镁0.1克,硫酸亚铁5毫克,硫酸锌3毫克,硫酸锰2毫克,加水至1000毫升,将三角瓶放在加热器上,升温90℃,保持1小时,冷却至40℃,过滤,将不溶性固形物分离,将滤液PH调至5.5,分装在500毫升三角瓶中,每瓶100毫升。按常规高压灭菌后,每瓶接种灵芝Gano derma Lucidum(Leysser ex Fries)karsten斜面菌种10立方厘米,置于旋转摇瓶机上,转速150RPM,温度25—28℃,培养5天,得到一级摇瓶菌种;用2000毫升三角瓶,每瓶装上述同样培养基1000毫升,灭菌后接种一级摇瓶菌种150毫升,在同样条件下培养4天,得到二级摇瓶菌种;用5000毫升三用瓶,每瓶装上述同样培养基2000毫升,灭菌后接种二级摇瓶菌种500毫升,在同样条件下培养3天,得到三级摇瓶菌种。
用100立升不锈钢配料罐,按下列组成配制培养基:麦麸粉9公斤,豆饼粉6公斤,玉米粉6公斤,蔗糖9公斤,磷酸二氢钾90克,硫酸镁30克,硫酸锌2.4克,硫酸亚铁3克,硫酸锰2克。加水至70立升,升温90℃,保持2小时,降温至40℃,将物料输入离心机,将不溶性固形物分离,将滤液输入400立升不锈钢搅拌式种子罐,加水至220立升,PH调至5.5,以1公斤/平方厘米压力蒸汽灭菌30分钟,待罐内温度降至30℃以下,接种三级摇瓶菌种60立升,保持罐压0.5公斤/平方厘米,温度25-28℃搅拌速度140RPM,通气量1∶0.8,培养60小时,得到一级种子液。
用400立升不锈钢配料罐,按下列组成配制培养基:麦麸粉18公斤,豆饼粉12公斤,玉米粉12公斤,蔗糖18公斤,磷酸二氢钾180克,硫酸镁60克,硫酸锌5克,硫酸亚铁6克,硫酸锰4克。加水至280立升,升温90℃,保持2小时,保持2小时,降温至40℃,将物料输入离心机,将不溶性固形物分离,将滤液输入1000立升不锈钢搅拌式种子罐,加水至450立升,PH调至5.5,以1公斤/平方厘米压力蒸汽灭菌30分钟,待罐内温度降至90℃以下,接种一级种子液260立升,保持罐压0.5公斤/平方厘米,温度25-28℃,搅拌速度14DRPM,通气量1∶0.8,培养55小时,得到二级种子液。
用2000立升不锈钢配料罐,按下列组成配料:麦麸粉49.5公斤,豆饼粉33公斤,玉米粉33公斤,蔗糖100公斤,磷酸二氢钾495克,硫酸镁165克,硫酸锌13.2克,硫酸亚铁16.5克,硫酸锰15克。加水至1400立升,升温90℃,保持2小时,降温至40℃,将物料输入离心机,将不溶性固形物分离,将滤液输入5000立升不锈钢搅拌式培养罐,加水至2800立升,PH调至5.5,以1公斤/平方厘米压力蒸汽灭菌40分钟,待罐内温度降至30℃以下,接种二级种子液700立升,保持罐压0.5公斤/平方厘米,温度25—28℃,通气量1∶1,搅拌速度130RPM,培养50小时,终止培养,罐内升温至60℃,保持2小时,菌丝自溶,降温至40℃,将培养液输入离心机,将菌丝分离,将纯净培养液输入真空泵经蒸发器减压浓缩20倍,得到棕色果冻状浓缩液。
实施例1得到的营养液经北京市理化分析测试中心以高效液相色谱法、离子色谱法、1CP—AES全测,其内含维生素(mg/100g):Vc 0.83,Vb1 0.99,Vb6 1.05,Va 0.61,Ve 2.52,Vd3 0.642,Vpp 1.31,总糖(g/100g)41.95,其中多糖4.49%,二糖11.361,葡萄糖18.821,果糖7.278。水解氨基酸17种,总含量(mg/100g)0.6806,元素23种,离子4种。
灵芝营养液中重要成分Ve、Va、Vd3含量高,多糖含量达4.49%,微量元素铬、锗、硒含量高。有害元素铅、铜含量低于国家标准,砷、汞未检出。
灵芝营养液经北京市卫生防疫站卫生检验合格。
Claims (2)
1、新型灵芝营养液,其特征是它是以保存菌种[灵芝Ganoderma Lucidum(Leysser ex Fries)Karsten]在下述培养基滤液中培养获得的。
培养基(W/V):麦麸粉3-8%,豆饼粉2-5%,玉米粉2-5%,蔗糖4-8%,磷酸二氢钾0.03-0.1%,硫酸镁0.01-0.1%,硫酸锌5-10PPM,硫酸亚铁10-20PPM,硫酸锰10-20PPM,余为水
2、权利要求1灵芝营养液的制备方法,其特征是:
(1)用保存菌种[灵芝Ganoderma Lucidum(Leysser exFries)Karsten]在PDA斜面培养基上接种,在25-30℃下培养10-15天,得到斜面菌种;
(2)以麦麸粉3-8%(W/V,下同),豆饼粉2-5%,玉米粉2-5%,蔗糖4-8%,磷酸二氢钾0.03-0.1%,硫酸镁0.01-0.1%,硫酸锌5-10PPM,硫酸亚铁10-20PPM,硫酸锰10-20PPM的量进行配制,升温到90℃,保持2小时,滤去不溶物,得到培养基滤液。
(3)将(1)制得的斜面菌种接种在(2)所得的培养基滤液内培养,温度25-30℃,时间为2-3天,液体培养菌种采用1-3级摇瓶和1-2级种子罐逐级扩大培养。摇瓶培养基装量40%,接种量10-20%,旋转式摇瓶机转速150-200RPM,然后于培养罐中培养,培养罐中培养基装量50-70%,接种量15-30%,温度25-28℃,时间为2-3天,罐压0.5KG/平方厘米。
(4)培养终止后,培养液升温至60℃,保持1-2小时,菌丝自溶,分离去掉不溶固状物,在≤60℃下,真空减压蒸发浓缩20倍。
