Abstract
提供了一种以抗体Fc区为骨架的融合蛋白二聚体,包括第一和第二多肽链,其中所述第一多肽链包括第一抗体Fc区和与所述第一抗体Fc区融合的一个或多个单域抗体;所述第二多肽链包括第二抗体Fc区和与所述第二抗体Fc区融合的一个或多个单域抗体;所述第一多肽链和/或第二多肽链还包括与各自抗体Fc区融合的细胞因子。还提供了该融合蛋白二聚体在制备治疗肿瘤的免疫治疗药物中的应用。所述Fc融合蛋白异源二聚体,不仅提升了单域抗体的活性,而且明显改善了细胞因子的生物学活性;并利用抗体的靶向特异性,有效增强了细胞因子的靶向运输,减弱了细胞毒性,从而具有更佳的抗肿瘤潜力。
