Abstract
本发明涉及一种消化内镜消毒液及其制备方法,其中将邻苯二甲醛、异丙醇、三氯羟基二苯醚与厚朴酚复配,采用不同机制的杀菌消毒成分直接或间接作用于致病微生物的生物蛋白质,增强对致病微生物如真菌、细菌、芽孢等的穿透作用,进而增强所述消毒液对上述微生物的杀灭作用,实现了消化内镜等精密医疗器械的高水平消毒。同时,本发明的消毒液还具有稳定性好,明显缩短所的浸泡时间,对消化内镜等医疗器械的腐蚀性小等优点。

技术领域
本发明涉及一种消化内镜消毒液及其制备方法,属于消毒液领域。
背景技术
消化内镜是一种重复性使用的精密检查器械,是消化领域重要的诊疗手段。但是由于消化内镜价格昂贵,且需要进入人体内部进行检查和治疗,因此,其消毒问题已经成为保障患者健康和预防医源性感染的重要问题之一。
目前,消化内镜的消毒主要为化学药液和气体两种,其中化学药液浸泡是更为常用的消毒方法。化学消毒剂种类繁多,如苯扎溴铵(新洁尔灭)、含氯消毒剂、戊二醛、过氧乙酸等。它们对各类细菌及少数芽孢菌有灭活作用,还能使真菌及病毒灭活,且无腐蚀性。苯扎溴铵对病毒具有杀伤作用,但消毒所需时间较长,否则消毒作用较差,故目前已不提倡使用;含氯消毒剂长期使用可使内镜软管受损,临床也较少用。戊二醛是国内目前较普遍使用的消毒剂,但是其气味大,对人体和环境均有危害,而且浸泡时间长,在临床应用中存在其对医患双方健康损害、出现耐药菌等多种问题。过氧乙酸对消化内镜进行消毒明显提高了消毒速率和效果,但是其仍有刺激性气味。而邻苯二甲醛作为一种高效消毒灭菌剂,具有高效、快速、毒性低、刺激性小等优点,日益受到关注和广泛使用。但是,目前,现有技术中可供选择的邻苯二甲醛产品较少。
发明内容
针对现有技术中存在的不足,本发明提供了一种消化内镜消毒液,所述消毒液具有高水平消毒、、浸泡时间短、刺激性小等特点,对真菌、细菌和芽孢等具有显著的杀灭效果。
为实现本发明,采取的技术方案是一种消化内镜消毒液,按质量百分比计,包括邻苯二甲醛0.5~5%、异丙醇5~10%、三氯羟基二苯醚0.5~2%、厚朴酚1.5~5%、表面活性剂1~10%、助溶剂0.5~10%、缓蚀剂0.2~5%、稳定剂0.2~6%、pH调节剂适量、余量为水。
在一个优选的实施方式中,所述表面活性剂选自脂肪醇聚氧乙烯醚、烷基酚聚氧乙烯、脂肪酸烷醇酰胺、聚氧乙烯聚氧丙烯丙二醇醚、十六烷基三甲基溴化铵、双十八烷基二甲基溴化铵、月桂醇聚醚硫酸钠、肉豆蔻醇聚醚硫酸钠或椰油酰氨基丙基甜菜碱的一种或多种。
在一个优选的实施方式中,所述助溶剂选自乙醇、甘油、丙二醇和聚乙二醇中的一种或多种。
在一个优选的实施方式中,所述缓蚀剂选自硅酸盐、钼酸盐、植酸、季戊四醇、六偏磷酸钠和多聚磷酸钠中的一种或多种。
在一个优选的实施方式中,所述稳定剂可以包括螯合剂和/或抗氧化剂;所述螯合剂选自乙二胺四乙酸及其钠盐、三聚磷酸钠、多聚磷酸钠、六偏磷酸钠、焦磷酸钠、乙二胺四乙酸及其钠中的一种或者几种;所述抗氧化剂选自抗坏血酸、抗坏血酸棕榈酸酯、亚硫酸钠、硫代硫酸钠、焦亚硫酸氨、半胱氨酸中的一种或多种。
在一个优选的实施方式中,所述pH值调节剂为无机强碱、碳酸盐、碳酸氢盐、硼酸、硼砂、磷酸二氢钾和磷酸氢二钠中的一种或多种。
在一个优选的实施方式中,所述消化内镜消毒液的pH值为7.5~8.5。
本发明的另一个目的是提供一种消化内镜消毒液的制备方法,包括以下步骤:
将邻苯二甲醛、异丙醇、缓蚀剂和稳定剂与部分水混合,边加热边搅拌,得到混合液1;
将三氯羟基二苯醚、厚朴酚、表面活性剂、助溶剂和剩余水混合,边加热边搅拌,得到混合液2;
将混合液1和混合液2混合,搅拌均与,加入pH调节剂至pH值为7.5-8.5,得到所述消化内镜消毒液。
有益效果
本发明首次将厚朴酚作为有效成分应用于消化内镜的消毒领域。本发明将邻苯二甲醛、异丙醇、三氯羟基二苯醚与厚朴酚复配,采用不同机制的杀菌消毒成分直接或间接作用于致病微生物的生物蛋白质,增强对致病微生物如真菌、细菌、芽孢等的穿透作用,进而增强所述消毒液对上述微生物的杀灭作用,实现了消化内镜等精密医疗器械的高水平消毒。同时,本发明的消毒液还具有稳定性好,明显缩短所的浸泡时间,对消化内镜等医疗器械的腐蚀性小等优点。
本发明的消毒液中还添加了表面活性剂、助溶剂、缓蚀剂、稳定剂等成分。利用表面活性剂的润湿、促渗透等作用,增加有效成分对致病微生物表面的接触面积,促进有效成分对致病微生物的杀灭作用。助溶剂加快邻苯二甲醛、三氯羟基二苯醚、厚朴酚等成分的溶解和相互融合速度,避免有效成分混合不均匀导致的效果不稳定以及长时间搅拌和加热导致的有效成分损失。加入缓蚀剂则降低了邻苯二甲醛等成分消化内镜的腐蚀性。稳定剂则增强了所述消化液的抗氧化性,保证产品品质稳定。

实施例1
一种消化内镜消毒液,按质量百分比计,包括邻苯二甲醛2.5%、异丙醇6%、三氯羟基二苯醚1%、厚朴酚2.5%、聚氧乙烯聚氧丙烯丙二醇醚5%、丙二醇3%、硅酸钠1%、六偏磷酸钠1.5%,抗坏血酸棕榈酸酯1.5%、pH调节剂适量、余量为水。
所述消化液的制备方法包括以下步骤:
将邻苯二甲醛、异丙醇、硅酸钠、六偏磷酸钠和抗坏血酸棕榈酸酯与部分水混合,边加热边搅拌,得到混合液1;
将三氯羟基二苯醚、厚朴酚、聚氧乙烯聚氧丙烯丙二醇醚、丙二醇和剩余水混合,边加热边搅拌,得到混合液2;

实施例2

所述消化液的制备方法包括以下步骤:
将邻苯二甲醛、异丙醇、钼酸钠、乙二胺四乙酸钠和半胱氨酸与部分水混合,边加热边搅拌,得到混合液1;
将三氯羟基二苯醚、厚朴酚、月桂醇聚醚硫酸钠、聚乙二醇和剩余水混合,边加热边搅拌,得到混合液2;
将混合液1和混合液2混合,搅拌均与,加入pH调节剂至pH值为8.5,得到所述消化内镜消毒液。
实施例3
一种消化内镜消毒液,按质量百分比计,包括邻苯二甲醛1.5%、异丙醇7%、三氯羟基二苯醚1%、厚朴酚2%、椰油酰氨基丙基甜菜碱8%、甘油5%、多聚磷酸钠2.5%、抗坏血酸棕榈酸酯3%、pH调节剂适量、余量为水。
所述消化液的制备方法包括以下步骤:
将邻苯二甲醛、异丙醇、多聚磷酸钠、抗坏血酸棕榈酸酯与部分水混合,边加热边搅拌,得到混合液1;
将三氯羟基二苯醚、厚朴酚、椰油酰氨基丙基甜菜碱、甘油和剩余水混合,边加热边搅拌,得到混合液2;
将混合液1和混合液2混合,搅拌均与,加入pH调节剂至pH值为8.5,得到所述消化内镜消毒液。
对比例1
一种消化内镜消毒液,按质量百分比计,包括邻苯二甲醛1.5%、异丙醇6%、三氯羟基二苯醚1%、苯扎溴铵2.5%、聚氧乙烯聚氧丙烯丙二醇醚5%、丙二醇3%、硅酸钠1%、六偏磷酸钠1.5%,抗坏血酸棕榈酸酯1.5%、pH调节剂适量、余量为水。

所述消化液的制备方法包括以下步骤:
将戊二醛、异丙醇、硅酸钠、六偏磷酸钠和抗坏血酸棕榈酸酯与部分水混合,边加热边搅拌,得到混合液1;
将三氯羟基二苯醚、厚朴酚、聚氧乙烯聚氧丙烯丙二醇醚、丙二醇和剩余水混合,边加热边搅拌,得到混合液2;
将混合液1和混合液2混合,搅拌均与,加入pH调节剂至pH值为8.5,得到所述消化内镜消毒液。
试验例1:考察本发明的消毒液的杀菌效果和内镜消毒模拟现场效果。
1.材料
试验指标菌为金黄色葡萄球菌(ATCC 6538)、枯草杆菌黑色变种芽孢(ATCC9372)、大肠杆菌(8099)和白色念珠菌(ATCC 10231),由中国普通微生物菌种保藏中心提供。
制备菌悬液和染菌载体:取各试验菌种,经增菌培养,取单个典型菌落接种营养琼脂培养基斜面,培养后洗下斜面上菌苔,用稀释液配制成所需浓度菌悬液,备用;依据2002年版《消毒技术规范》培养出枯草杆菌黑色变种芽孢悬,在灭菌的聚四氟乙烯管腔载体,在管腔内污染细菌芽孢,要求回收菌数达到相关规定要求,晾干备用。
2.方法
2.1悬液定量杀菌试验:将本发明的消毒液(实施例1-3)、对比例1-3的消毒液与菌悬液共同放置水浴(20℃)中恒温保持5min。用无菌吸管分别吸取4ml本发明实施例1-3的消毒液和对比例1-3的消毒液注入六个相同的无菌试管中,分别吸取0.5ml菌悬液和0.5ml3%牛血清白蛋白加于上述两个无菌试管中,混合形成均匀的菌药混合液。将菌药混合液相互作用固定时间,吸取0.5ml菌药混合液加于4.5ml中和剂中,混匀。中和作用10min,进行活菌计数,计算杀灭对数值。试验进行3次。
2.2内镜消毒模拟现场试验:按《内镜清洗消毒机消毒效果检验技术规范(试行)》的要求,将实施例1-3和对比例1-3的消毒液装于内镜清洗消毒机的消毒槽内,将模拟内镜体管道在50nm、500nm和950nm处剪开,取染菌载体分别连接在50nm、500nm和950nm处,将模拟内镜体装放于内镜清洗消毒机。按消毒程序(初洗2min、酶洗2min、清洗1min、浸泡消毒5min和末洗2min)进行处理,程序结束后取下载体进行活菌培养计数。试验进行3次。
3.试验结果
3.1悬液定量杀菌效果
试验结果表明,本发明的消毒液对悬液内大肠杆菌、金黄色葡萄球菌和白色念珠菌,分别作用1min和3min,杀灭对数值如下表1所示。对枯草芽胞杆菌黑色变种芽孢作用30min和60min,平均杀灭对数值如下表2所示。本发明实施例1的消毒液对大肠杆菌、金黄色葡萄球菌和白色念珠菌明显优于对比例1。对于枯草芽胞杆菌黑色变种芽孢的作用,本发明实施例1的消毒液仅作用30min即可达到平均杀灭对数值>5的效果,其作用时间相较于对比例1明显缩短。

3.2内镜消毒模拟现场效果
试验结果表明,本发明的消毒液装于内镜清洗消毒机进行模拟现场消毒,按消毒程序(初洗2min、酶洗2min、清洗1min、浸泡消毒5min、末洗2min)处理后,对载体上人工污染的枯草杆菌黑色变种芽孢的消除效果明显优于对比例1。

试验例2:考察本发明的消毒液的稳定性和金属腐蚀性
1.稳定性考察
将密封包装的本发明实施例1的消毒液置37℃恒温恒湿箱中存放90天。于放置前、后分别取样,测定邻苯二甲醛含量,计算其下降率。经计算,邻苯二甲醛含量下降了约1%。
2.金属腐蚀性试验
将不锈钢、铝和碳钢制成直径(20±1)mm、厚1.0mm、中间设置直径15mm小孔的圆片,用砂纸磨去表面氧化层,经洗涤剂浸泡10min,洗净;用无水乙醇再次脱脂。置50℃干燥1h称重后浸泡于本发明实施例1的消毒液中(100ml/片),浸泡时间为72h,并定期更换消毒液,每日1次。对照组为蒸馏水连续浸泡。浸泡结束后取出金属片,用蒸馏水和软毛刷反复洗净,滤纸吸干,置50℃干燥1h后称重,计算腐蚀速率。经检测,本发明实施例1的消毒液对不锈钢的腐蚀速率为0.0065mm/a,对铝的腐蚀速率是0.0077mm/a,对碳钢的腐蚀速率是0.0051mm/a,均属于基本无腐蚀级别。
基于以上试验结果可以看出,本发明的消毒液配制厚朴酚后对细菌等的杀灭效果明显增强,对芽孢的杀灭时间明显缩短,同时保持了稳定性以及对金属无腐蚀,产生了明显优于现有技术的良好消毒效果。

1.一种消化内镜消毒液,其特征在于,按质量百分比计,所述消毒液包括邻苯二甲醛0.5~5%、异丙醇5~10%、三氯羟基二苯醚0.5~2%、厚朴酚1.5~5%、表面活性剂1~10%、助溶剂0.5~10%、缓蚀剂0.2~5%、稳定剂0.2~6%、pH调节剂适量、余量为水。
2.根据权利要求1所述的消毒液,其特征在于,所述表面活性剂选自脂肪醇聚氧乙烯醚、烷基酚聚氧乙烯、脂肪酸烷醇酰胺、聚氧乙烯聚氧丙烯丙二醇醚、十六烷基三甲基溴化铵、双十八烷基二甲基溴化铵、月桂醇聚醚硫酸钠、肉豆蔻醇聚醚硫酸钠或椰油酰氨基丙基甜菜碱的一种或多种。
3.根据权利要求1所述的消毒液,其特征在于,所述助溶剂选自乙醇、甘油、丙二醇和聚乙二醇中的一种或多种。
4.根据权利要求1所述的消毒液,其特征在于,所述缓蚀剂选自硅酸盐、钼酸盐、植酸、季戊四醇、六偏磷酸钠和多聚磷酸钠中的一种或多种。
5.根据权利要求1所述的消毒液,其特征在于,所述稳定剂可以包括螯合剂和/或抗氧化剂;所述螯合剂选自乙二胺四乙酸及其钠盐、三聚磷酸钠、多聚磷酸钠、六偏磷酸钠、焦磷酸钠、乙二胺四乙酸及其钠中的一种或者几种;所述抗氧化剂选自抗坏血酸、抗坏血酸棕榈酸酯、亚硫酸钠、硫代硫酸钠、焦亚硫酸氨、半胱氨酸中的一种或多种。
6.根据权利要求1所述的消毒液,其特征在于,所述pH值调节剂为无机强碱、碳酸盐、碳酸氢盐、硼酸、硼砂、磷酸二氢钾和磷酸氢二钠中的一种或多种。
7.根据权利要求1所述的消毒液,其特征在于,所述消化内镜消毒液的pH值为7.5~8.5。
8.根据权利要求1所述的消毒液的制备方法,其特征在于,包括以下步骤:
将邻苯二甲醛、异丙醇、缓蚀剂和稳定剂与部分水混合,边加热边搅拌,得到混合液1;
将三氯羟基二苯醚、厚朴酚、表面活性剂、助溶剂和剩余水混合,边加热边搅拌,得到混合液2;
将混合液1和混合液2混合,搅拌均与,加入pH调节剂至pH值为7.5-8.5,得到所述消化内镜消毒液。
