Abstract
本发明提供了一种具有防晒、美白及抗衰效果的组合物及其应用,属于化妆品技术领域。本发明组合物包含黄芩根提取物、芦丁、姜黄素和阿魏酸,这四种组分的质量比为黄芩根提取物:芦丁:姜黄素:阿魏酸=2‑18:1‑14:0.1‑8:0.1‑5,在这四种组分的协同作用下,本发明组合物具有优异的防晒、美白、抗衰的效果,而且本发明组合物天然安全、易于工业化生产和推广应用。

技术领域
本发明属于化妆品领域,涉及一种具有防晒、美白及抗衰效果的组合物及其应用。
背景技术
近年来,随着大气臭氧层部分空洞的出现,使其对紫外线的过滤效果减弱,日光的辐射强度增加。虽然太阳紫外线是万物生长的必备条件,但过量的紫外线会损害人体免疫系统,损伤真皮内胶原和弹性硬蛋白,使皮肤弹性下降,出现皱纹,造成皮肤老化。同时,长时间、高强度的紫外线照射会让皮肤产生大量自由基,诱发细胞膜的过氧化反应,致使黑色素细胞产生大量黑色素,然后经由细胞代谢的层层移动,到了肌肤表皮层,形成了我们所看到的色斑和肤色不匀等皮肤问题。因此,注重防晒是保持肌肤健康状态的重要途径。
纵观现在化妆品市场,大多数产品都含有防晒剂,把防晒保护概念结合到日常护肤生活中。但大多数防晒产品为了追求高SPF防晒效果,添加了大量有机紫外吸收剂,这无疑将大大提高皮肤过敏和刺激的可能性。在美白、抗衰已成行业热点的背景下,开发研究兼具防晒、美白、抗衰多种功效为一体的天然植物提取物具有很好的应用前景。
发明内容
为了克服现有技术的上述缺点与不足,本发明的目的在于提供一种具有防晒、美白及抗衰效果的组合物及其应用。
为实现上述目的,本发明采用的技术方案为:
第一方面,本发明提供了一种具有防晒、美白及抗衰效果的组合物,其包含黄芩根提取物、芦丁、姜黄素和阿魏酸,所述黄芩根提取物、芦丁、姜黄素和阿魏酸的质量比为黄芩根提取物:芦丁:姜黄素:阿魏酸=2-18:1-14:0.1-8:0.1-5。
作为本发明组合物进一步优选的实施方式,所述黄芩根提取物、芦丁、姜黄素和阿魏酸的质量比为黄芩根提取物:芦丁:姜黄素:阿魏酸=15-18:10-14:5-8:5。
黄芩根提取物具有抗菌抗炎、抗氧化、免疫调节等作用;芦丁具有抗炎、抗氧化、抗自由基、抗辐射等作用;姜黄素是一种从姜科植物姜黄等的根茎中提取得到的黄色色素,具有抗炎、抗氧化、美白、抗衰等作用;阿魏酸具有美白、抗氧化、防晒等作用。发明人将黄芩根提取物、芦丁、姜黄素以及阿魏酸组合使用,发现这四种组分之间能协同防晒、美白、抗衰,当所述黄芩根提取物、芦丁、姜黄素和阿魏酸的质量比为黄芩根提取物:芦丁:姜黄素:阿魏酸=2-18:1-14:0.1-8:0.1-5时,所述组合物的防晒、美白、抗衰效果都很好,尤其当所述黄芩根提取物、芦丁、姜黄素和阿魏酸的质量比为黄芩根提取物:芦丁:姜黄素:阿魏酸=15-18:10-14:5-8:5时,所述组合物的防晒、美白、抗衰效果都更好。
作为本发明组合物的优选实施方式,所述黄芩根提取物、芦丁、姜黄素和阿魏酸的质量比为黄芩根提取物:芦丁:姜黄素:阿魏酸=18:14:8:5。
作为本发明组合物的优选实施方式,所述黄芩根提取物、芦丁、姜黄素和阿魏酸的质量比为黄芩根提取物:芦丁:姜黄素:阿魏酸=15:10:5:5。
作为本发明组合物的优选实施方式,所述黄芩根提取物包含黄芩苷、汉黄芩素、汉黄芩苷、黄芩素中的至少一种。
第二方面,本发明提供了一种化妆品,所述化妆品包含上述组合物。
作为本发明化妆品的优选实施方式,所述化妆品包含以下质量百分含量的组分:黄芩根提取物0.01-18%,芦丁0.01-14%,姜黄素0.01-8%,阿魏酸0.01-8%。
作为本发明化妆品进一步优选的实施方式,所述化妆品包含以下质量百分含量的组分:黄芩根提取物0.1-15%,芦丁1-12%,姜黄素0.1-7%,阿魏酸0.05-8%。
作为本发明化妆品进一步优选的实施方式,所述组合物质量占所述化妆品质量的11-55%。这样,所述化妆品的SPF值大于15。
作为本发明化妆品的优选实施方式,所述化妆品还包含以下质量百分比的组分:化妆品基质10-40%,余量去离子水
作为本发明化妆品的优选实施方式,所述化妆品基质包含润肤剂、乳化剂、增稠剂、保湿剂、中和剂、防腐剂、物理防晒剂、螯合剂和香料中的至少一种。
作为本发明化妆品的优选实施方式,所述化妆品为液体状、乳液状、膏霜状或固体状。
作为本发明化妆品进一步优选的实施方式,所述化妆品为乳液状或膏霜状。
第三方面,本发明还提供了上述化妆品的制备方法,其包括以下步骤:
(1)将甘油、黄原胶、EDTA二钠、丙二醇和水混合,升温至75-85℃,得到A相;将鲸蜡硬脂基葡糖苷、C14-22醇、C12-20烷基葡糖苷、聚二甲基硅氧烷、单硬脂酸甘油酯、钛白粉浆、异壬酸异壬酯、白油、姜黄素、羟苯甲酯和羟苯丙酯混合,升温至75-85℃,得到B相;将B相加入A中混合并均质乳化5-10min,得到AB乳化相;
(2)将所述AB乳化相降温至60-70℃,再加入黄芩苷、芦丁、阿魏酸、聚丙烯酰胺、C13-14异链烷烃以及月桂醇聚醚-7,均质乳化3-5min,冷却至40℃以下,过滤,即得所述化妆品。
与现有技术相比,本发明具有以下优点和有益效果:
(1)本发明组合物通过黄芩根提取物、芦丁、姜黄素、阿魏酸之间的协同增效作用而具有优异的防晒、美白、抗衰的效果。
(2)本发明组合物中的功效组分来源于植物,天然又安全,可应用于化妆品中,所得化妆品不仅具有优异的防晒、美白、抗衰的功效,而且易于推广应用。
(3)本发明化妆品制备方法简单,易于工业化生产。
具体实施方式
为更好的说明本发明的目的、技术方案和优点,下面将结合具体实施例对本发明作进一步说明。
实施例1-4以及对比例5-8均为涉及一种组合物,这些组合物的组成如表1所示;对比例1-4均涉及一种天然物质,这些天然物质的种类如表1所示。

效果例1——酪氨酸酶活性抑制率测试
酪氨酸酶是一种多酚氧化酶,是皮肤黑色素形成过程中的关键酶、限速酶,酪氨酸酶的活性与黑色素合成量呈正相关,其活性改变可引起色素障碍性皮肤病。酪氨酸酶抑制剂通过抑制酪氨酸酶活性,阻止黑色素形成,可用来预防和治疗色素沉着等疾病。
测试样品:以0.2mol/L pH=6.8的磷酸缓冲液(PBS)为溶剂,分别将实施例1-4以及对比例1-8中的组合物或天然物质配制成浓度为0.1mg/mL的溶液,即得测试样品。
测试方法:分别上述0.1mg/mL的测试样品,加入1mL L-酪氨酸(0.3mol)和1mL pH=6.8的磷酸缓冲液(PBS),在37℃恒温水槽中预热10min,再加入0.1mL酪氨酸酶水溶液(1.0mg/mL,mcllvains缓冲液),充分搅拌,在37℃反应15分钟后,测定475nm吸光度(D1)。另外,用上述磷酸缓冲液代替上述实施例1-4和对比组1-5所制得的组合物配制的浓度为0.1mg/mL的溶液进行同样的反应,测得吸光度(D2)。为了对照,在不加试样溶液和酪氨酸酶的条件下,测得吸光度(D3)。用下式计算出酪氨酸酶活性抑制率:
酪氨酸酶活性抑制率(%)=(D2-D1)/(D2-D3)×100%;
测试结果如表2所示。

试验结论:对比例1~4均单独使用黄芩根提取物、芦丁、姜黄素、阿魏酸这四种天然物质中的一种,与实施例1对比可看出,单独使用这四种天然物质中的任意一种,酪氨酸酶活性抑制率均显著低于本发明组合物;对比例5~8为使用这四种天然物质中的任意三种所获得的组合物,与实施例1其他条件相同,可看出,相对于单独使用其中一种天然物质,其清除率有提升,但其效果与本发明组合物还存在差异,说明本发明组合物需在这四种提取物的共同协同作用下,才会达到显著的酪氨酸酶活性抑制效果。
效果例2——黑色素生成抑制率测试
测试方法:选择对数生长期的同一代细胞,常规胰酶消化处理后吹打成单细胞悬液细胞计数定量接种于96孔板。24小时后,弃去上清液,分别加入含有实施例1-4和阳性对照药-熊果苷50μg/ml的培养液(各组合物的体积含量相同)。在37℃,体积分数5%的CO2饱和湿性条件下培养,对照孔加RPMI1640培养液,培育三天,中间更换一次培育液,之后弃去上清液,常规胰酶消化处理后,计数细胞密度,离心得到细胞沉淀,用PBS冲洗两次,离心弃去上清液,定量加含10vol%DMSO的1N的NaOH液,80℃水浴50分钟,然后在酶标仪450nm处测定吸光值。用下式计算出黑色素生成的抑制率:
黑色素生成的抑制率(%)=(试验组吸光度/对照组的吸光度)×100%;
测试结果如表3所示。

试验结论:实施例1-3中的组合物都具有抑制黑色素生成的作用,且实施例4的组合物与熊果苷抑制黑色素生成的作用相当,可应用到美白类化妆品中。
效果例3——抗皱测试
自由基可以从多方面对皮肤造成损伤,从而加速皮肤的衰老,因此,抗衰老产品是否具有抗衰老作用可由其对自由基的清除、抑制效果来判定。
(1)DPPH自由基清除能力测试
测试方法:将实施例1-4中的组合物、对比例1-4中的天然物质以及对比例5-8中的组合物配制成浓度为0.1mg/mL的溶液,分别用无水乙醇将其稀释到40、80、120、160、200μM;取DPPH 1.4mg加17.5mL无水乙醇即得0.2mmol/L的DPPH溶液;取100μL上述不同浓度的样品溶液加入96孔酶标板中,然后加入现用现配的DPPH溶液100μL,使样品作用最终浓度为20、40、60、80、100μM。酶标板在30℃下保温30min后测定样品在517nm波长下的吸光度值(A1),同时测定不加DPPH自由基试剂的样品空白(100μL无水乙醇代替DPPH)吸光值(A2)和加DPPH自由基试剂但不加样品(100μL无水乙醇代替样品)的吸光值(A3)。用下式计算出DPPH自由基清除率:

测试结果如表4所示。

(2)超氧阴离子自由基清除能力测试
测试方法:配制pH值为8.20的缓冲体系(Tris-HCl-EDTA),在10mL离心管中加入待测样品0.6mL,加入5.2mL缓冲液,用移液管加入0.045mol/L邻苯三酚溶液0.2mL,速摇匀,等待反应5分钟(时间必须精确)后迅速于干燥比色皿中,在325nm处测吸光度值,记为A0,10min后再测记为A4。以蒸馏水代替样品测定的相应的吸光值分别记为B0和B4。平行操作3次取平均值,用下式计算出超氧阴离子的抑制率:
超氧阴离子的抑制率(%)=[(B4-B0)-(A4-A0)]/(B4-B0)×100%;
测试结果如表5所示。

(3)羟自由基清除能力测试
测试方法:分别取1mL、0.4mmol/L结晶紫,3mL 0.2mol/L pH 7.4磷酸缓冲液,1mL、0.001mol/L FeSO4溶液,1mL待测样品,加入10mL离心管中,最后加1mL30%H2O2反应,于室温下反应1h后12000r/min离心5min,取上清液在510nm下测定吸光值记为C,用蒸馏水代替样品时的吸光值记为C1,用蒸馏水代替样品和H2O2时的吸光值记为C2。平行操作3次取平均值,用下式计算出样品对羟自由基的清除率:

由表4、表5、表6可知,本发明实施例1~4所述组合物均具有较强的清除自由基和抗氧化能力,其中以实施例1~2的效果最佳。
上述效果测试时,对比例1-8与实施例1-4相同条件,可看出,单独使用4种天然物质中的一种或由其中任意3种组成的组合物,自由基清除率均显著低于本发明组合物,说明本发明组合物在这需四者共同协同作用下,才会达到显著的美白、抗氧化效果。
设置实施例5-9,均为一种具有防晒、美白及抗衰效果的乳霜,它们的组成如表7所示,同时设置对照组乳霜,其组成也如表7所示。

实施例5-9中的乳霜的制备方法包括以下步骤:
(1)将甘油、黄原胶、EDTA二钠和水混合,升温至75-85℃,得到A相;将鲸蜡硬脂基葡糖苷、C14-22醇、C12-20烷基葡糖苷、聚二甲基硅氧烷、单硬脂酸甘油酯、异壬酸异壬酯、白油、钛白粉浆、姜黄素、羟苯甲酯和羟苯丙酯混合,升温至75-85℃,得到B相;将B相加入A中混合并均质乳化5-10min,得到AB乳化相;
(2)将所述AB乳化相降温至65℃,再加入黄芩苷、芦丁、阿魏酸、聚丙烯酰胺、C13-14异链烷烃以及月桂醇聚醚-7,均质乳化3-5min,冷却至40℃以下,过滤,即得所述乳霜。
采用SPF 290S(Optometrics)对对照组以及实施例5-9中的乳霜进行防晒指数测试,测试结果如表8所示。

与对照组相比,实施例5-9的防晒指数得到了显著提升,SPF值达到15~25,可作为具有防晒作用的日常护肤品使用,同时相比使用化学防晒剂达到该指数的产品对皮肤更安全。
本发明最后所应当说明的是,以上实施例仅用以说明本发明的技术方案而非对本发明保护范围的限制,尽管参照较佳实施例对本发明作了详细说明,本领域的普通技术人员应当理解,可以对本发明的技术方案进行修改或者等同替换,而不脱离本发明技术方案的实质和范围。

1.一种具有防晒、美白及抗衰效果的组合物,其特征在于,所述组合物包含黄芩根提取物、芦丁、姜黄素和阿魏酸,所述黄芩根提取物、芦丁、姜黄素和阿魏酸的质量比为黄芩根提取物:芦丁:姜黄素:阿魏酸=2-18:1-14:0.1-8:0.1-5。
2.根据权利要求1所述的组合物,其特征在于,所述黄芩根提取物、芦丁、姜黄素和阿魏酸的质量比为黄芩根提取物:芦丁:姜黄素:阿魏酸=15-18:10-14:5-8:5。
3.根据权利要求1或2所述的组合物,其特征在于,所述黄芩根提取物包含黄芩苷、汉黄芩素、汉黄芩苷、黄芩素中的至少一种。
4.一种化妆品,其特征在于,所述化妆品包含如权利要求1-3任一项所述的组合物。
5.根据权利要求4所述的化妆品,其特征在于,所述化妆品包含以下质量百分含量的组分:黄芩根提取物0.01-18%,芦丁0.01-14%,姜黄素0.01-8%,阿魏酸0.01-8%。
6.根据权利要求5所述的化妆品,其特征在于,所述化妆品包含以下质量百分含量的组分:黄芩根提取物0.1-15%,芦丁1-12%,姜黄素0.1-7%,阿魏酸0.05-8%。
7.根据权利要求5所述的化妆品,其特征在于,所述组合物质量占所述化妆品质量的11-55%。
8.根据权利要求5所述的化妆品,其特征在于,所述化妆品还包含以下质量百分比的组分:化妆品基质10-40%,余量去离子水;所述化妆品基质包含润肤剂、乳化剂、增稠剂、保湿剂、中和剂、防腐剂、物理防晒剂、螯合剂和香料中的至少一种。
9.根据权利要求4所述的化妆品,其特征在于,所述化妆品为液体状、乳液状、膏霜状或固体状。
10.一种如权利要求4-9任一项所述的化妆品的制备方法,其特征在于,所述制备方法包括以下步骤:
(1)将甘油、黄原胶、EDTA二钠、丙二醇和水混合,升温至75-85℃,得到A相;将鲸蜡硬脂基葡糖苷、C14-22醇、C12-20烷基葡糖苷、聚二甲基硅氧烷、单硬脂酸甘油酯、异壬酸异壬酯、钛白粉浆、白油、姜黄素、羟苯甲酯和羟苯丙酯混合,升温至75-85℃,得到B相;将B相加入A中混合并均质乳化5-10min,得到AB乳化相;
(2)将所述AB乳化相降温至60-70℃,再加入黄芩苷、芦丁、阿魏酸、聚丙烯酰胺、C13-14异链烷烃以及月桂醇聚醚-7,均质乳化3-5min,冷却至45℃以下,过滤,即得所述化妆品。
