The invention relates to a medicinal and edible composition for reducing uric acid and treating gout, which comprises the following preparation method:
1. The intelligent screening and fine pretreatment of the raw materials, namely screening out the raw materials with high quality and high activity by means of advanced spectrum analysis technology and biological activity detection means. 2. The multistage targeted extraction technology is a multistage targeted extraction technology of firstly fusing dynamic countercurrent extraction, subcritical fluid extraction and ultrasonic auxiliary extraction. (1) Weighing 15 parts by weight of celery seeds, 10 parts by weight of pagodatree flower bud, 12 parts by weight of chicory, 10 parts by weight of dandelion, 12 parts by weight of mulberry twig, 12 parts by weight of papaya, and 15 parts by weight of fresh cogongrass rhizome
(2) The active ingredients can be extracted efficiently and prepared into oral preparations by intelligent screening and fine pretreatment, multistage targeted extraction process, intelligent concentration and accurate molding steps of the traditional Chinese medicine raw materials.
Experimental design and data
Animal experiment:
The aim of the experiment is to evaluate the uric acid reducing and anti-inflammatory effects of the composition on animal models of hyperuricemia.
The experimental animals are selected from 60 male SD rats, the weight of the experimental animals is 200-220g, and the experimental animals are randomly divided into 6 groups of 10 animals each.
The modeling method is to construct a hyperuricemia model by injecting potassium oxazinate (300 mg/kg) into the abdominal cavity once a day for 7 days.
After successful molding, the control group is given with equal amount of physiological saline, the low dose group is given with 100mg/kg of the composition, the medium dose group is given with 200mg/kg of the composition, the high dose group is given with 400mg/kg of the composition, and the positive control group is given with allopurinol (10 mg/kg), and the stomach is irrigated once a day for 14 days.
The detection index comprises collecting rat blood after administration, detecting serum uric acid, creatinine and urea nitrogen level, collecting joint synovial tissue, and detecting expression level of inflammatory factors TNF-alpha and IL-6.
As a result of the experiment, compared with a control group, serum uric acid levels of the composition groups are obviously reduced (16.3% in a low-dose group, 33.6% in a medium-dose group, 45.7% in a high-dose group and 39.3% in an IL-6-dose group), uric acid reducing effects of the composition groups are equivalent to those of a positive control group, and TNF-alpha and IL-6 expression levels in joint synovial tissues are obviously reduced (15.4% in the low-dose group, 19.3% in the IL-6 group, 34.9% in the medium-dose group, 26.7% in the IL-6 group, 46.8% in the TNF-alpha in the high-dose group and 39.3% in the IL-6 group, and P < 0.05) in each dose group of the composition, so that the composition has good anti-inflammatory effect. The specific data are shown in Table 1.

Human clinical trial:
The experimental purpose is to verify the clinical curative effect and safety of the composition in gout patients.
Subjects were enrolled in 100 patients with confirmed gout, aged 35-65 years, randomized into experimental and control groups of 50 persons each.
The treatment scheme is that the traditional Chinese medicine preparation capsules prepared by the composition are taken by patients in an experimental group every day, 3 capsules are taken every time, and the placebo capsules are taken by patients in a control group, wherein the dosage of the placebo capsules is the same. The treatment course is 3 months.
The detection index comprises detecting serum uric acid, blood sedimentation and C-reactive protein level of the patient each month, recording the times of joint pain and pain degree of the patient (adopting visual simulation scoring method, VAS), and observing the change of liver and kidney function index of the patient.
Experimental results after treatment, serum uric acid levels of patients in the experimental group were reduced by 120. Mu. Mol/L on average compared with those before treatment, and the difference was significant compared with the control group (P <
0.05 Blood sedimentation, C reactive protein level is also obviously reduced (blood sedimentation is reduced by 15mm/h, C reactive protein is reduced by 8mg/L, P is less than 0.05), the number of times of arthralgia attack is reduced by 3 times/month, VAS score is averagely reduced by 2.5 minutes, and liver and kidney function indexes have no obvious abnormal change, so that the composition has good safety when effectively treating gout. The specific data are shown in Table 2.

Safety acute toxicity test:
The experimental animals are 20 healthy Kunming mice, and the weight is 18-22g, and the male and female animals are half.
Administration method the composition extract was administered at the maximum tolerated dose (20 g/kg) and the mice were observed for general status, diet, drinking water, excretion and death after a single administration for 14 consecutive days.
The experimental result shows that the mice have no obvious toxic reaction after being dosed, normal diet and drinking water, no death phenomenon, smooth fur, free movement, bright eyes, no secretion and normal fecal form and color. The composition extract has extremely low acute toxicity, basically has no damage to organisms at extremely large dosage, and preliminarily verifies the edible safety. Specific observations are detailed in table 3.

Long-term toxicity test:
the experimental animals are selected from 80 SD rats, each half of which has a weight of 150-180g, and are randomly divided into 4 groups of 20 animals each.
Dosing regimen control group was given an equivalent amount of physiological saline, low dose group was given 2g/kg of composition extract, medium dose group was given 5g/kg, high dose group was given 10g/kg, and the administration was performed once daily by gavage for 90 days.
The detection index comprises recording rat weight, food intake and water intake every week, collecting blood for detecting blood convention every half month, and collecting liver and kidney function index (glutamic pyruvic transaminase, glutamic oxaloacetic transaminase, creatinine, urea nitrogen, etc.), and collecting main viscera (heart, liver, spleen, lung, kidney, stomach, intestine, etc.) at the end of test
A pathological histological examination was performed.
The experimental results show that the weight growth trend of rats in each dosage group is similar to that of rats in a control group in the whole experimental period, the weight growth trend of rats in each dosage group has no obvious difference (P is more than 0.05), the food intake and water intake of rats in each dosage group have no obvious fluctuation, the blood routine indexes are all in a normal range, the difference of the liver and kidney function indexes in each dosage group has no statistical significance (P is more than 0.05) compared with the control group, and the pathological tissue examination shows that the tissue structure of each viscera is clear, and the abnormal conditions such as obvious lesions, inflammations, cell necrosis and the like are avoided. The combination shows that the composition has no obvious toxic and side effect on organisms and has good long-term edible safety under long-term and large-dose administration. The detailed data are shown in tables 4-7.

S2, purifying and preparing the traditional Chinese medicine preparation, wherein the preparation method adopts the steps of intelligent screening and fine pretreatment of raw materials, multistage targeted extraction technology, intelligent concentration and precise forming, and can efficiently extract active ingredients and prepare various dosage forms.
S3, the traditional Chinese medicine preparation is applied, in the aspect of accurate medical assistance, personalized medicine application schemes can be customized according to different stages of gout, whole-course digital management is achieved by means of intelligent equipment and a cloud platform, and in daily intelligent health care of families, gout prevention is facilitated through intelligent household kitchen equipment and uric acid monitoring packages, and the traditional Chinese medicine preparation can be used for innovation and health food development, such as uric acid regulation energy bars, gout prevention meal replacement powder and the like.

The oral preparation and the preparation method thereof according to claim 1 are characterized in that S2, raw materials are intelligently screened and finely preprocessed, wherein the raw materials are screened by means of a spectrum analysis technology and a biological activity detection means, batches which are planted organically, have no pesticide residue and have active ingredient content above a specific threshold value are selected, the impurities are removed by a supercritical fluid cleaning technology after harvesting, the materials are dried to the water content of less than 99% under the condition of-40 ℃ to-50 ℃ by a vacuum freeze drying technology, and after flaws and worm-eaten parts of pagodatree flower buds, mulberry twigs and papaya are manually selected and removed, the pagodatree flower buds and the mulberry twigs are cut into small sections of 2 cm to 3 cm and papaya is cut into slices of about 0.5 cm by a numerical control cutting device.

The oral preparation and the preparation method thereof according to claim 1 are characterized in that in S2, the multistage targeted extraction process comprises the steps of firstly mixing pretreated celery seeds, chicory, dandelion and fresh cogongrass rhizome according to a specific proportion, putting into dynamic countercurrent extraction equipment, taking deionized water as a solvent, controlling the feed liquid ratio to be 1:7, the extraction temperature to be 40-50 ℃, shortening the extraction time to 0.5 hour, ultra-filtering and purifying the extract by a ceramic membrane, then accurately extracting fat-soluble active ingredients for pagodatree flower buds, mulberry branches and papaya under the conditions of critical temperature and pressure of 0.6-0.9MPa by adopting subcritical fluid extraction technology, removing solvent residues by adopting supercritical carbon dioxide extraction technology after extraction, and finally merging the two extracts, and putting into ultrasonic extraction equipment for 30-45 minutes at the ultrasonic frequency of 40-60 kHz.
5. The oral preparation and the preparation method thereof according to claim 1 are characterized in that the intelligent concentration and the accurate molding in S2 are carried out, the mixed extract after ultrasonic treatment is connected into an intelligent concentration control system, the concentrated extract is accurately concentrated to thick extract with the relative density of 1.2-1.3 under the conditions of the vacuum degree of 0.08-0.1MPa and the temperature of 45-55 ℃, if the oral preparation is prepared, proper pharmaceutical excipients are added for different dosage forms according to big data analysis and clinical feedback, the 3D printing technology is adopted when capsules are prepared, each capsule contains 0.4-0.6g of composition dry powder, a high-speed tablet press is used for preparing tablets, each tablet contains 0.5-0.7g of composition, a proper amount of flavoring agent and preservative are added into the prepared oral liquid, the oral liquid is diluted to a specified concentration tank, and if the oral liquid is used for dietary addition, the extract is converted into superfine micropowder with the granularity distribution of 1-10 mu m by adopting a low-temperature spray drying technology, and the sealing packaging is carried out.
