Abstract
本发明涉及饲料添加剂技术领域,尤其涉及一种南美白对虾养殖用饲料添加剂及其制备方法和应用,包括以下质量份原料:第一弧菌抑制剂3‑7份、第二弧菌抑制剂1‑3份、全氨基β‑环糊精5‑7份、微胶囊助剂1‑2份、聚氧乙烯40氢化蓖麻油0.3‑0.5份、混合发酵制剂10‑15份、去离子水50‑70份;所述第一弧菌抑制剂由辣木籽素、香兰素按照1:(0.2‑0.5)的质量比混合而成;所述第二弧菌抑制剂为莳萝精油;所述混合发酵制剂由丁酸梭菌发酵制剂和桑黄发酵制剂按照1:(0.15‑0.3)的质量比混合而成。本发明制出的饲料添加剂能有效抑制水体中的致病弧菌,提高南美白对虾的成活率,并具有诱食增香的效果。

技术领域
本发明涉及饲料添加剂技术领域,还涉及微生物发酵技术领域,尤其涉及一种南美白对虾养殖用饲料添加剂及其制备方法和应用。
背景技术
南美白对虾为十足目、对虾科、对虾属水生甲壳动物,学名凡纳滨对虾,又称白皮虾、白对虾、基围虾、明虾,体色为浅青灰色,体表带有细小斑点,壳薄体肥,营养丰富,味道鲜美,成为世界三大主要对虾养殖品种之一。在南美白对虾的养殖过程中,易受弧菌属病原菌感染,诱发南美白对虾患弧菌病,且对虾弧菌病能在短时间内迅速蔓延,导致南美白对虾的大规模死亡。弧菌致病菌包括霍乱弧菌、副溶血性弧菌、创伤弧菌、溶藻弧菌、哈维氏弧菌、拟态弧菌、鳗弧菌等,会导致对虾早期死亡综合征(EMS)、对虾肝胰腺急性坏死(AHPND)等弧菌病。例如,副溶血弧菌是诱发南美白对虾早期死亡综合征的主要病原菌,患病南美白对虾体色呈白浊微红,且伴有肝胰脏肿大、质地松软,部分患病对虾的肝胰脏明显萎缩。副溶血弧菌的致病机理为:1)副溶血弧菌产生溶血毒素:溶血毒素能造成南美白对虾中的细胞发生溶血,导致对虾死亡;2)副溶血弧菌有肠毒素作用:造成南美白对虾消化道组织受损,导致回肠糜烂、胃粘膜炎、消化功能紊乱;3)副溶血弧菌产生脲酶:脲酶使南美白对虾肠液积聚,造成肠道积水,严重时,可能导致拉白便问题;4)副溶血弧菌的胞外分泌物使南美白对虾的免疫力下降,导致生长发育迟缓,并易引发其他疾病。
因此,关于防治致病弧菌的新型饲料添加剂一直是南美白对虾养殖领域的研究热点。如公告号为CN103283990B的发明专利,公开桦褐孔菌多糖作为凡纳滨对虾饲料添加剂的应用,桦褐孔菌多糖在凡纳滨对虾基础饲料中的添加量为500-1500mg/kg凡纳滨对虾基础饲料,能有效提高对虾的生长性能、免疫力和抗氧化能力,为凡纳滨对虾饲料的开发提供了新途径,促进了对虾养殖业的可持续发展。如公告号为CN107494998B的发明专利,公开一种抑制南美白对虾肠道弧菌的复合饲料添加剂,所述添加剂以重量百分比计,包含以下成分:柠檬酸8-12%、丁酸梭菌1012cfu/kg~5×1012cfu/kg、柴胡5-8%、白术3-6%、白芍2-5%、防风2-5%、陈皮1-3%、甘草1-3%,能有效地选择性降低对虾肠道中弧菌的数量,同时又能有效避免丁酸梭菌的应用对肝脏的损伤作用,提高对虾防御外源性弧菌入侵肠道的能力,从而有效预防对虾弧菌病的发生及其引发的死亡。如公告号为CN112369534B的发明专利,公开一种预防对虾早期死亡的饲料添加剂及其制备方法及应用,首先通过对破壁雨生红球藻一次包被,使雨生红球藻与空气隔绝避免氧化,再将一次包被的破壁雨生红球藻藻液与吸附有有机复合酸、三丁酸甘油酯的二氧化硅混合得到混合颗粒,将混合颗粒二次包被,包裹住有机酸有效成分;通过双层包被工艺提高了该添加剂的相对独立性,有效规避了应用过程中有效成分与饲料成分反应,避免对虾饲料在水体中长时间浸泡导致有机酸的损耗,并一定程度解决了虾青素加工工艺中高温耐受性、易氧化的缺点,保证了虾青素的性质稳定及有效吸收,与有机酸成分发挥协同作用,抑制致病原弧菌增殖、改善了对虾肠道菌群平衡,增强了对虾的机体免疫力和抗病性能。如公告号为CN114672446B的发明专利,公开一种丁酸梭菌制剂的制备方法及应用,包括以下步骤:将菌种活化,得到丁酸梭菌发酵液;将丁酸梭菌发酵液离心,收集菌泥,然后加入保护剂溶液,混匀,再加入壁材溶液,混匀,最后冷冻干燥,得到丁酸梭菌制剂;其中,保护剂为脱脂乳、β-环状糊精、葡萄糖和磷酸氢二钾的混合物;壁材为明胶、海藻酸钠或羧甲基纤维素钠;通过保护剂和壁材的双重作用,能够有效的保证丁酸梭菌在凡纳滨对虾肠道中定植,进而能够对凡纳滨对虾肠道健康的修复提供保障。
但是,现有饲料添加剂一般都是利用包衣材料或壁材对活性内容物进行物理包合,如CN112369534B、CN114672446B,由于包衣材料分子间不发生化学交联作用,分子间隙较大,形成的微胶囊饲料添加剂的外壁较为疏松,不够致密,其包合稳定性有待进一步提高,对于挥发性活性成分仍存在溢出散失的可能性,进而影响饲料添加剂抑制弧菌病原菌及诱食的效果,尤其是,当饲料添加剂遇水后或进入养殖水体后,这类饲料添加剂的溶解释放速度仍较高,难以达到长效缓释控释的效果。
发明内容
有鉴于此,本发明的目的在于提出一种南美白对虾养殖用饲料添加剂及其制备方法和应用,一方面,研制一种新型饲料添加剂以进一步提高防治南美白对虾治病弧菌的效果,另一方面,如何提高微胶囊饲料添加剂的包合稳定性,以防止挥发性活性成分的散失,延长缓释时间,保持良好的抗菌诱食效果。
基于上述目的,本发明提供了一种南美白对虾养殖用饲料添加剂,包括以下质量份原料:第一弧菌抑制剂3-7份、第二弧菌抑制剂1-3份、全氨基β-环糊精5-7份、微胶囊助剂1-2份、聚氧乙烯40氢化蓖麻油0.3-0.5份、混合发酵制剂10-15份、去离子水50-70份;
所述第一弧菌抑制剂由辣木籽素、香兰素按照1:(0.2-0.5)的质量比混合而成;

经辣木籽素处理后,对霍乱弧菌、副溶血性弧菌、创伤弧菌、溶藻弧菌、哈维氏弧菌等南美白对虾致病弧菌具有明显的抑制作用。具体地,辣木籽素明显抑制了南美白对虾致病弧菌的生长,使细胞膜通透性和损伤率显著增加,使胞内大分子核酸、蛋白质和糖类物质泄漏到胞外;辣木籽素处理南美白对虾致病弧菌后,抑制弧菌生物膜的形成,降低生物被膜粘附性,干扰弧菌疏水作用和弧菌菌体泳动性;辣木籽素能显著增加弧菌胞内Ca2+含量,导致Ca2+失衡,损伤弧菌的代谢功能,进而导致弧菌凋亡;且辣木籽素还能诱导和促进细胞凋亡进程,且随着辣木籽素用量的提高,细胞凋亡率也逐渐增加;

香兰素又名香草醛,化学名称为3-甲氧基-4-羟基苯甲醛,具有浓郁的香荚兰豆香气及奶香,具有增香、诱食、调味的作用,提高南美白对虾对饲料的摄食率和利用率,与此同时,南美白对虾致病弧菌作为革兰氏阴性菌,细胞壁结构疏松,易与香兰素结合,通过破坏细胞膜完整性和通透性,使胞内环境发生改变,进而抑制酶的活性(例如,香兰素能抑制DNA聚合酶的活性),影响细胞体内的新陈代谢,并阻碍南美白对虾致病弧菌延滞期中遗传物质的合成、表达;
所述第二弧菌抑制剂为莳萝精油,莳萝精油包括香芹酮、α-水芹烯、β-水芹烯、柠檬烯等活性成分,能有效杀灭南美白对虾致病弧菌,如霍乱弧菌、副溶血性弧菌、创伤弧菌、溶藻弧菌、哈维氏弧菌等,莳萝精油还通过导致细胞膜发生畸变和损伤破裂,改变通透性,使细胞内容物泄漏,进而达到抑菌效果,同时具有清除饲料中自由基性能和抗脂质过氧化能力,并有助于提高南美白对虾的免疫力,进而提高其成活率;此外,莳萝精油还能促进南美白对虾伤口的快速愈合,降低感染风险;
最后,辣木籽素、香兰素、莳萝精油具有协同抑制南美白对虾致病弧菌的作用,三者在弧菌抑制机制上具有互补增强的效果,通过不同途径、不同靶点达到多重抗弧菌效果,通过与弧菌细胞膜的脂质相互作用,破坏细胞膜的完整性和通透性,干扰弧菌正常生物代谢,抑制其生长繁殖,并干扰生物膜的形成和维持,破坏弧菌的保护层,从而达到协同增效的效果;

A1.制备丁酸梭菌发酵液:将丁酸梭菌接种至RCM液体培养基中,控制丁酸梭菌与RCM液体培养基的质量比为(4-5):100,32-37℃厌氧发酵1-2d,得丁酸梭菌发酵液;该丁酸梭菌发酵液中含有包括丁酸、丙酮酸、乳酸、乙酸以及少量的丙酸、甲酸等有机酸在内的代谢产物,能够穿透革兰氏阴性菌弧菌的细胞壁,在细胞质中解离并干扰弧菌细胞的功能,进而具有一定的广谱抗弧菌性能;同时,代谢产物还具有一定的挥发性香气,进而提高饲料的适口性和诱食效果;此外,丁酸梭菌与寄生在南美白对虾肠道中的致病弧菌存在竞争关系,进而限制弧菌的生长繁殖,并促进南美白对虾肠道有益菌群增殖,增强免疫功能,降低南美白对虾肠炎的发病概率;
A2.制备丁酸梭菌发酵制剂:将丁酸梭菌发酵液进行冷冻干燥,得丁酸梭菌发酵制剂。
进一步地,所述桑黄发酵制剂的制备方法,包括以下步骤:
B1.将桑黄菌丝接种至桑黄液体培养基中,控制桑黄菌丝与桑黄液体培养基的质量比为(0.2-0.4):100,在28-32℃通风条件下,有氧发酵7-10d,压滤后,分离出桑黄菌体和发酵液体;
B2.将桑黄菌体烘干后,超微粉碎至粒径为5-10μm,得桑黄菌体超微粉;
B3.再将所得桑黄菌体超微粉加入所分离出的发酵液体中,60-70℃超声处理1-2h后,得到桑黄发酵液;将桑黄菌体进行超微粉碎、超声处理,有助于提高菌丝体胞内多糖、子实体多糖及黄酮等活性成分的溶出率和提取率;
B4.将桑黄发酵液进行减压浓缩、喷雾干燥后,得桑黄发酵制剂;该桑黄发酵制剂富含桑黄多糖,以及黄酮类化合物(主要为二氢黄酮和黄烷-3-醇黄酮)、萜类化合物(主要为倍半萜、二萜和三萜)、吡喃酮类化合物(具有苯乙烯基吡喃酮骨架和苯并吡喃酮骨架结构)、甾体化合物(主要为麦角甾烷类甾体)等活性成分,具有抗菌、免疫调节、抗氧化、清除自由基、修复南美白对虾受损细胞及组织的作用。
进一步地,所述桑黄液体培养基的配方为:葡萄糖20-25g/L、木质纤维素25-30g/L、土豆粉2-3g/L、蛋白胨3-5g/L、KH2PO40.5-1g/L、K2HPO40.1-0.5g/L,pH=6.7-6.9。
本发明进一步提供南美白对虾养殖用饲料添加剂的制备方法,包括以下步骤:
S1:将全氨基β-环糊精、微胶囊助剂加入去离子水中,超声处理0.5-1h后,再加入聚氧乙烯40氢化蓖麻油,搅拌均匀后,得壁材溶液;
S2:将第一弧菌抑制剂、第二弧菌抑制剂、混合发酵制剂混合均匀后,得芯材混合物;
S3:将芯材混合物加入壁材溶液中,32-37℃搅拌处理0.5-1h,再冷冻干燥后,即得南美白对虾养殖用饲料添加剂;在该过程中,全氨基β-环糊精还能与第一弧菌抑制剂中的辣木籽素、香兰素发生交联作用,进一步加强包埋稳定性以及所制成的微胶囊饲料添加剂的结构稳定性;与此同时,全氨基β-环糊精中的氨基与香兰素中的醛羰基反应,形成具有席夫碱结构的衍生物,进一步强化抗南美白对虾致病弧菌的效果。
本发明还进一步上述制备方法制出的南美白对虾养殖用饲料添加剂在南美白对虾饲料中的应用,该饲料添加剂的用量为南美白对虾饲料质量的0.1-0.15%。
本发明的有益效果:
本发明制出的饲料添加剂能有效抑制水体中的致病弧菌,提高南美白对虾的成活率,不会产生抗药性,避免抗生素对养殖水体造成污染,且有助于提高南美白对虾对饲料的采食率,进而提高饲料有效利用率。具体地:
本发明首次采用辣木籽素、香兰素组配第一弧菌抑制剂,并以莳萝精油作为第二弧菌抑制剂,三者协同增效,明显提高抗致病弧菌的性能和南美白对虾的成活率,提香增香,达到诱食效果。
本发明通过桑黄发酵制剂和丁酸梭菌发酵制剂的科学合理组配,改善南美白对虾肠道环境,进一步提高对致病弧菌的抑制效果,并提高免疫功能,进一步提高南美白对虾的成活率。
最后,利用全氨基β-环糊精、双醛淀粉进行包埋处理,形成致密稳定的微胶囊结构,防止外界杂菌侵入,并避免饲料添加剂中挥发性香气成分的散失,同时,进入养殖水体后,缓慢释放有效成分,延长饲料添加剂对弧菌的抑制时间和诱食时间。

试验一:应用在南美白对虾饲料中的养殖效果测试:
将实施例1-3及对比例1-7制出的饲料添加剂各添加至相同的南美白对虾饲料,饲料添加剂的用量为南美白对虾饲料质量的0.1%,南美白对虾饲料购于无棣瑞兴生物科技有限公司,选取PL10生长期的南美白对虾,每组南美白对虾的养殖数量为200尾,每组设置3个平行实验,每天投喂3次,投喂时间分别为6:00、13:00、21:00,连续养殖28d,并计算成活率、饲料系数,计算公式如下:
成活率=100%×(28d后的活虾尾数-初始虾尾数)/初始虾尾数,初始虾尾数=200;
饲料系数=总投饲量/(28d后的终体重-初始体重);
养殖试验结果如表1所示:

由表1可知,实施例1-3制出的饲料添加剂明显提高了南美白对虾的成活率以及提高配合饲料的有效利用率,说明了本发明制出的饲料添加剂具有优良的抗病原菌和诱食的效果。
试验二:抗弧菌性能测定:
以霍乱弧菌、副溶血弧菌为试验弧菌菌种,分别用无菌水将霍乱弧菌、副溶血弧菌稀释至106CFU/mL,以100mL LB液体培养基为空白对照组,并以添加实施例1-3及对比例1-7制出的饲料添加剂的100mL LB液体培养基作为10个试验组,且饲料添加剂的用量为LB液体培养基质量的0.1%,再向空白对照组和试验组中接种200μL试验弧菌稀释液,并于37℃、200r/min下摇床培养72h,再根据以下公式进行计算:弧菌抑制率=100%×(空白对照组培养后弧菌菌量-试验组培养后弧菌菌量)/空白对照组培养后弧菌菌量。抗弧菌性能测试试验结果如表2所示:

由表2可知,实施例1-3制出的饲料添加剂具有优越的抗致病弧菌的性能,对霍乱弧菌、副溶血弧菌的抑制性能超过97%。
试验三:稳定性测试:
将实施例1-3及对比例5-6制出的饲料添加剂置于90RH%、25℃的环境中暴露7d后,对比实施例1-3及对比例5-6制出的饲料添加剂的香气差异、潮解情况,试验结果如表3所示:

由表3可知,实施例1-3制备的饲料添加剂在90RH%、25℃的环境中能保留饲料添加剂的香气成分,而对比例5-6制备的饲料添加剂更易潮解,香气成分也相对更容易散失,说明了实施例1-3通过全氨基β-环糊精、微胶囊助剂的交联作用提高了外层壁材的致密性和稳定性。

A2.制备丁酸梭菌发酵制剂:将丁酸梭菌发酵液进行冷冻干燥,得丁酸梭菌发酵制剂。
7.根据权利要求1所述的一种南美白对虾养殖用饲料添加剂,其特征在于,所述桑黄发酵制剂的制备方法,包括以下步骤:
B1.将桑黄菌丝接种至桑黄液体培养基中,控制桑黄菌丝与桑黄液体培养基的质量比为(0.2-0.4):100,在28-32℃通风条件下,有氧发酵7-10d,压滤后,分离出桑黄菌体和发酵液体;
B2.将桑黄菌体烘干后,超微粉碎至粒径为5-10μm,得桑黄菌体超微粉;
B3.再将所得桑黄菌体超微粉加入所分离出的发酵液体中,60-70℃超声处理1-2h后,得到桑黄发酵液;
B4.将桑黄发酵液进行减压浓缩、喷雾干燥后,得桑黄发酵制剂。
8.根据权利要求7所述的一种南美白对虾养殖用饲料添加剂,其特征在于,所述桑黄液体培养基的配方为:葡萄糖20-25g/L、木质纤维素25-30g/L、土豆粉2-3g/L、蛋白胨3-5g/L、KH2PO40.5-1g/L、K2HPO40.1-0.5g/L,pH=6.7-6.9。
9.一种根据权利要求1-8任一项所述的南美白对虾养殖用饲料添加剂的制备方法,包括以下步骤:
S1:将全氨基β-环糊精、微胶囊助剂加入去离子水中,超声处理0.5-1h后,再加入聚氧乙烯40氢化蓖麻油,搅拌均匀后,得壁材溶液;
S2:将第一弧菌抑制剂、第二弧菌抑制剂、混合发酵制剂混合均匀后,得芯材混合物;
S3:将芯材混合物加入壁材溶液中,32-37℃搅拌处理0.5-1h,再冷冻干燥后,即得南美白对虾养殖用饲料添加剂。
10.一种根据权利要求9所述的制备方法制出的南美白对虾养殖用饲料添加剂在南美白对虾饲料中的应用,其特征在于:该饲料添加剂的用量为南美白对虾饲料质量的0.1-0.15%。
